    Description / Table of Contents: Investigation of Chromatographic Resolutions of Racemates, VI. Polymeric Amino Acid Derivatives as Optically Active AdsorbensStarting with optically active amino acids, the polyacrylamides 3a-h, 5a-d and 6a, b the polymethacrylamides 8a-c and the polyacrylic ester 6f were synthesized. The separation efficiencies of these polymers were determined by chromatography of racemic mandelic acid and mandelamide, respectively. The best separations of the enantiomers of both racemates were observed on the phenylalanine derivatives 3e and 5a.
    Notes: Die Polyacrylamide 3a-h, 5a-d und 6a, b die Polymethacrylamide 8a-c und der Polyacrylsäureester 6f wurden aus optisch aktiven Aminosäurederivaten hergestellt und auf ihre Trennwirkung durch Chromatographie racem. Mandelsäure sowie racem. Mandelamids geprüft. Die beste Trennung der Enantiomeren beider Racemate wurde an den Phenylalaninderivaten 3e und 5a beobachtet.

    Notes: Chloroquine Enantiomers by Chromatographic Resolution and SynthesisThe chromatographic resolution of the antimalarial drug chloroquine (1) on an optically active polyamide is described. The results reveal that the optical purity of chloroquine formerly resolved with d-bromocamphorsulfonic acid was 12% only. A facile synthesis of the enantiomers via the optically active diamine 4 is given.

The amide 5c was resolved on a preparative scale yielding both optically pure enantiomers.
    Notes: Die optisch aktiven Polyamide 1, 2a und b trennen Racemate von α-substituierten 2-Phenylacetamiden (3a-g), Mandelsäurederivaten (4a-c), N-substituierten Acetamiden (5a-d, 6, 7), N-Acylaminosäureestern (8a-h) und Benzoinderivaten (9a-c). Das Amid 5c wurde im präparativen Maßstab in beide optisch reinen Enantiomeren aufgetrennt.

    Notes: Information on spatial correlation in the tangent direction along electron microscope images of filamentous molecule is shown to be obtainable by the analysis of statistical fluctuations in curvature, yielding an absolute measure of the persistence parameter amicro. The relationship of amicro, a local, microscopic parameter, to the persistence length introduced by Kratky and Porod is discussed. The hypotheses underlying the assumed theoretical model concern (1) the shape of the angle distribution, assumed to be Gaussian; (2) the passage from a three- to a two-dimensional situation, which is supposed to occur by deformation of the flexible chain in a manner that preserves the memory of the spatial correlation in orientation (except for the blocking of one degree of freedom); and (3) the adsorption conditions, which should meet the equilibrium requirement as closely as possible. The analytical method has been checked on computer simulated “Gaussian” molecules: the study of the simulated sample was essential in solving the problems connected with minimum statistics requirements and the effect of the reading error. Experimental images obtained for T2 DNA fragments at different ionic strengths by Kleinschmidt's adsorption technique have been analyzed by means of an automatic flying spot digitizer, the “Precision Encoder and Pattern Recognition.” The results show that adsorbed molecules do in fact “remember” the rigidity they possessed in solution and that the Gaussian hypothesis is well verified. Consequently, the slopes of log cosθ(l) or θ2(l) may be used indifferently in the estimate of amicro. The dependence of this parameter on ionic strength in the range explored shows the expected behavior.

    Notes: When exposed to a low pH, in various ionic strength conditions and in sufficiently dilute solutions, DNA undergoes a transition, revealed by an increase in optical density. A careful analysis shows that, associated with this transition, there is an effective decrease in absorbance, overcompensated by an increase in scattering. The conditions for the new transition can be summarized conveniently by a graph in a pH-Na+ molarity diagram. If the pH of a DNA solution is progressively lowered at constant Na+ concentration, one finds first the melting transition (I), and at lower pH values, the new transition (II). If the same experiment is performed on pre-denatured DNA, only transition II will be found. If native DNA is brought directly to the low pH conditions, without allowing it to denature irreversibly at intermediate pH values, transition II is reversible (with a small hysteresis effect). DNA, initially native, neutralized after prolonged exposures to the low pH, recovers the buoyant density value of native DNA, along with the absorption and scattering properties of the native state. The experiments are consistent with the interpretation that a new state exists in which DNA, still double stranded, assumes a very compact shape (of the order of 1500 Å in diameter for T2 DNA), with a hyperchromicity value of 10-14% above the native value. Nearly monodisperse suspensions of DNA molecule in this apparent state may be obtained only at very low concentrations (∼0.25 μg/ml). At 1 μg DNA/ml aggregation is noticeable. The possible connection with the condition of intraphage DNA is discussed.

    Notes: The use of a simple chemical equilibrium and of the law of mass action leads to correct prediction when applied to the study of the role of the counterions in the process of acid melting of the DNA molecule. This approach allows an estimate to be given of the enthalpy variation per base pair associated with this process. Experiments have been carried out to test (a) the linearity of the dependence of pHm vs. 1/T; (b) the stabilizing effect of Na+ concentration. The enthalphy variation per base pair, deduced from the slope of the pHm vs. 1/T plot, and from the number of H+ bound per base pair, is in agreement with direct measurement.6,8
