    Notizen: Procedures have been developed for the isolation of pharmacologically active indole compounds from biological samples and for the introduction of electron-capturing groups, pentafluorobenzyl and trifluoroacetyl, onto the indole nitrogen atom. The resulting derivatives have good gas chromatographic properties and strong electron affinities which make them highly suitable for detection capture negative chemical ionization mass spectrometry. These procedures were used to identify 1-methyl-1,2,3,4-tetrahydro-β-carboline as a component of rat brain and lung.

    Notizen: Details are presented of an ultra-sensitive gas chromatographic/mass spectrometric assay for phenylacetic acid in plasma and cerebrospinal fluid based on measurements of the relative intensities of the carboxylate anions, derived from the penta- and tetraflourobenzyl esters under electron capture chemical ionization conditions, of unlabeled and a (13C2)-labeled internal standard. The limits of detection for the penta- and tetrafluorobenzyl esters are 0.85 and 4.0 pg respectively, and the assay is capable of measuring phenylacetic acid concentrations in samples as small as 20 μl of CSF and plasma. The penta- and tetrafluorobenzyl esters are chromatographically separated on the gas chromatograph column, which allows for their co-injection and independent measurement from the same chromatogram.

    Notizen: The tricyclic indole derivative 6-hydroxymethtryptoline (7-hydroxy-1-methyltetrahydro-β-carboline) was identified as a normal constituent of human and cat urine. The identification was based on the correct retention time on a chiral capillary gas chromatographic column and on selected ion monitoring of the molecular ion with the mass spectrometer set at high resolving power (RP 10 000). The quantitation utilized a 13C-labeled analog as internal standard and demonstrated levels of 135 ± 35 and 174 ± 42 (pmol ml-1, mean ±SEM) for the S(-) and R(+) enantiomers, respectively, in human urine. In cat urine, the levels were 203 ± 55 and 108 ± 44 (pmol ml-1, mean ± SEM) for the S(-) and R(+) enantiomers, respectively. In human urine, the compound occurred predominantly in a conjugated form while in cat urine the free compound predominated. It was concluded that 6-hydroxymethtryptoline is naturally occurring in mammalian urine and that it most likely constitutes a metabolite of endogenous methtryptoline (1-methyltetrahydro-β-carboline).

    Notizen: Tris(isopropylphenyl/phenyl) phosphates and triethylene glycol dicaprate/caprylate mixtures of typical industrial compositions have been analyzed by fast atom bombardment (FAB) mass spectrometry, FAB collisional activation (CA) mass-analyzed ion kinetic energy spectroscopy and FAB/electron impact (EI) CA B/E linked scans. Data obtained by these state-of-the-art methodologies are compared qualitatively and semi-quantitatively with those provided by classical gas chromatography (GC)/EI MS. To identify the plasticizers in cases of suspected contamination, GC/EI MS still appears to be the method of choice whenever ultimate sensitivity and specificity are needed. In cases where these requirements are less demanding, FAB mass spectrometry and FAB tandem mass spectrometry may be sufficient and more convenient.

    Notizen: Six years after coining the term ‘mass spectrometric (MS) peptide charting’ for the component analysis of peptide mixtures in a whole tissue, body fluid, or an extract thereof, we offer our current perspective of this field. Matrixassisted laser desorption/ionization and electrospray ionization have replaced plasma desorption and fast atom bombardment as ionization methods of choice. At the same time, the upper mass range has been extended to now include most peptides and proteins of interest to research on cell-cell communication. In addition to qualitative aspects, quantitative applications of MS charting have become important. In combination with new database search algorithms, on-line liquid chromatography-tandem mass spectrometry promises greater dividends from MS charting than are achievable with molecular mass matching alone. We discuss what is and is not yet possible, and consider foreseeable means for overcoming current limitations. Our intent is to encourage researchers in the biological and medical sciences to take advantage of this powerful methodology in their various fields of endeavor.

    Notizen: Platinum catalyzed hydrogen-deuterium exchange labeling of phenylalanine and tyrosine containing peptides in deuterium oxide is exemplified by the preparation of deuterium labeled leucine enkephalin and related peptides. NMR and mass spectral evidence indicates that the side chains of the aromatic residues are the sites of label incorporation. Time course studies with N-acetyltyrosine and tyrosyl-glycine suggest that the exchange of the aromatic protons occurs most rapidly followed by exchange of the benzylic protons adjacent to the aromatic ring.

    Beschreibung: Many surface proteins are anchored to the cell wall of Gram-positive bacteria and are involved in the pathogenesis of these organisms. A hybrid molecule was designed that, when expressed in Staphylococcus aureus, was anchored to the cell wall and could be released by controlled enzymatic digestion. By a combination of molecular biology and mass spectrometry techniques, the structure of the cell wall anchor of surface proteins in S. aureus was revealed.     Beschreibung: The rostral migratory stream (RMS) is the main pathway by which newly born subventricular zone cells reach the olfactory bulb (OB) in rodents. However, the RMS in the adult human brain has been elusive. We demonstrate the presence of a human RMS, which is unexpectedly organized around a lateral ventricular extension reaching the OB, and illustrate the neuroblasts in it.
