    Notes: Der wesentlichste Anteil des für das Pflanzenwachstum der Erde notwendigen Stickstoffes wird biologisch durch Mikroorganismen gebunden. Nur wenige Prozente gebundenen Stickstoffes werden auf anderem Wege, etwa künstliche Stickstoff-Düngemittel, verfügbar. Die Voraussetzungen und die Primärschritte der biologischen Stickstoff-Bindung werden diskutiert. Versuche und neue Forschungsergebnisse zeigen, daß es möglich ist, die Ausnützung biologischer Stickstoff-Bindung systematisch weiterzuentwickeln und derart die Bodenfruchtbarkeit und landwirtschaftliche Produktion wesentlich zu erhöhen.

    Notes: Der quantitativ wichtigste Syntheseweg der Pflanzen zu Aminosäuren verläuft über das Ammoniak, wenn auch dem Hydroxylamin als Zwischenprodukt der Nitrat-Reduktion eine wesentliche Rolle zufallen mag. Es ist gelungen, die Oxime der im Stoffwechsel zentralen α-Ketosäuren sowohl in Hefe als in grünen Pflanzen zu identifizieren. Eine unerwartete Anzahl neuer Aminosäuren wurde aus verschiedenen Pflanzen isoliert und strukturell aufgeklärt. Die entsprechenden neuen α-Ketosäuren wurden auch in den entsprechenden Pflanzen in den meisten Fällen gefunden, wodurch die experimentelle Begründung der Annahme vorhanden ist, daß diese Aminosäuren durch Transaminierung entstehen können. Die mit Hilfe moderner Untersuchungsmethoden (Austauscher, Papierchromatographie) in den letzten Jahren aus Pflanzen und Mikroorganismen isolierten Keto- und Aminosäuren werden beschrieben.

    Notes: The deposition of the basement membrane glycoproteins, laminin, fibronectin, and type IV procollagen was studied by indirect immunofluorescence microscopy during the attachment and differentiation of murine C-1300 neuroblastoma cells. A typical cytoplasmic perinuclear staining for the basement membrane antigens was seen both in undifferentiated and differentiated cells. Freshly seeded suspended cells lacked surface fluorescence but in two hours after plating, distinct punctate laminin deposits became discernible on the ventral surface of the cells. Notably, in sparsely seeded undifferentiated cultures, the cell-associated extracellular laminin deposits could only be detected under the primary attaching cells, whereas daughter cells in clonal cell colonies lacked such fluorescence. In cultures induced to neurite formation with dibutyryl cyclic AMP, laminin deposition was also detected in association with the growing cytoplasmic extensions. No distinct differences were found between the secreted proteins of cultures of differentiated and nondifferentiated neuroblastoma cells, but the patterns of fucosylation of high-molecular weight proteins in the two cultures were markedly different.We conclude that cultured neuroblastoma cells both synthesize, secrete and deposit laminin. The distribution of laminin during neuroblastoma cell attachment and neurite extension suggests that this glycoprotein may be involved in cell-to-substratum interactions in C-1300 cell cultures.

    Notes: The presence of prosome proteins (p25K and p27K) was shown and their distribution was studied in oogenesis of Xenopus laevis using immunoblotting and immunofluorescence. These proteins form numerous granular clusters of variable size all over the cell. At previteilogenic stages, the prosome antibodies homogeneously stain the oocyte nucleus and the evenly distributed relatively large clusters in the cytoplasm. As the oocyte grows, the pattern of distribution of the prosome proteins undergoes changes: animal-vegetal and cortical gradients appear in the cytoplasm. In the course of oocyte maturation the size of clusters diminishes. Artificial activation of the egg leads to a dorso-ventral gradient in distribution of the prosome proteins. In this way, specific localization of prosome proteins is first visualized during formation of the dorso-ventral polarity. Co-localization of prosome proteins and actin and myosin was found in the oocyte by double staining. Small clusters of prosomes dispersed in the cytoplasm acquire capability of movement (after artificial activation) due, in all likelihood, to persisting connection with the acto-myosin complex of the egg. © 1994 Wiley-Liss, Inc.

    Notes: The distribution of contractile proteins, actin and myosin, and an actin-binding protein, spectrin, was studied in oogenesis of Xenopus laevis. These proteins are present in oocytes already at the previtellogenic stages, which are characterized by their diffuse distribution. The localization of proteins changed with the beginning of vitellogenesis. At all vitellogenic stages, including the fully grown oocyte, animal-vegetal differences were noted in localization of actin and myosin: in the animal hemisphere they appear as fibrillar-like structures, while in the vegetal one they are localized around the yolk platelets. By the end of the oocyte's growth, a cortical gradient appeared: predominant localization of actin and myosin in the cortical area. As the oocyte maturation proceeded, the distribution of actin and myosin again became diffuse and nonuniform, so that a cortical gradient appears. At the beginning of vitellogenesis spectrin is distributed as a network all over the ooplasm, while in the fully grown oocyte it is localized mostly in teh subcortical area of the animal hemisphere and, as individual inclusions, in other regions of the oocyte. No spectrin is found by the end of maturation. Actin, myosin, and spectrin are also present in the oocyte's nuclei. Changes in the distribution of contractile proteins and spectrin during oocyte maturation are discussed with respect to the development of cortical contractility, as well as to the changes in spatial distribution of yolk platelets and regional sensitivity of the maturing oocyte to cytochalasin B. © 1994 Wiley-Liss, Inc.
