    Notes: Resonant power absorption is an important phenomenon during microwave heating. The resonances that occur when plane electromagnetic waves are incident on infinitely long cylinders and slabs are investigated as a function of sample dimensions. For cylinders two kinds of incident waves are studied: TMz when the electric field is oriented along the axis of the cylinder and TEz when the magnetic field is oriented along the axis. At a resonant condition the overage power absorbed by the sample is a local maximum. Due to attenuation within the sample the resonances decrease in intensity as the sample size increases. Using the dielectric properties of water, resonances are found to be a function of the ratio of the sample dimension to the wavelength of radiation, λs, in the sample. For slabs of thickness L and integer values of n, resonances occurred at L/λs = 0.5n; for cylinders of diameter D, resonances occurred at D/λs = 0.5n - 0.257. The generality of these relations to predict resonances in other food samples are shown using existing dielectric data. Resonances in cylinders for both polarizations occurred at similar radii. However, the radius at which the first resonance occurred for the TMz case was absent in the TEz case.

    Notes: A primary method of reducing membrane fouling during cross-flow microfiltration is periodic reverse filtration. This in situ method of cleaning the membrane forces clear fluid in the reverse direction through the membrane and readjusts the particle or solute accumulation on the retentate side of the membrane. This work focuses on the design of a high-frequency, reverse-filtration strategy to maximize the flux for washed yeast suspensions through 0.2- μm cellulose acetate flat sheet membranes. Several experiments were conducted with reverse-filtration times ranging from 0.5-4 s and forward-filtration times ranging from 1-40 s. For every back-filtration time, there exists an optimum forward-filtration time that gives the maximum global average flux. The optimum average flux increases with decreasing back filtration times and feed concentrations, but shows little dependence on cross-flow velocity and reverse filtration transmembrane pressure. The optimum flux with rapid backflushing is 20 to 30 times higher than the long-term flux in the absence of backflushing. A theory presented assumes that cake formation during forward filtration follows dead-end filtration theory and the cake is instantly removed during reverse filtration. The measured average flux per cycle follows the trends predicted by the theory, but the measured values exceed the predictions, presumably due to brief delays in cake removal and cake formation at the start of reverse and forward filtration, respectively, during each cycle.

    Notes: In this work the experimental observations are explained with the aid of a simple air-entrapment model based on the concept of two levels of porosity of fiber mats. A simple model that includes liquid bypassing with initial air trapping, subsequent capillary invasion of regular fiber bundles with air compression, and finally mobilization is proposed to explain air-entrapment phenomena. The simple model successfully rationalizes the observed air trapping and compression during initial liquid-fiber contact. An empirical equation for the mobilization efficiency is adapted to the model to estimate void content. The velocity dependence of the trapped void content predicted by the model is in good agreement with observations.

    Notes: Wetting of glass fibers was visualized using an oil with the same refractive index. With both a video-enhanced microscope and a high-magnification video camera we followed the flow front and qualitatively measured the entrapped air bubbles by image analysis. Due to different permeabilities between the fiber mats and bundles, air bubbles are entrapped. Two major kinds of air bubbles are observed: small cylindrical micro voids between fiber filaments (inside fiber bundles) and spherical macro voids outside fiber bundles. These air bubbles can be described by three major mechanisms initial liquid bypassing/air trapping, later capillary invasion of disordered fiber bundles, and air bubble mobilization. Both random and unidirectional fiber mats were used in this study. Vacuum as well as different fluid viscosities and surface tensions were also investigated, which led us to guidelines of optimum processing parameters of fiber wetting for resin transfer molding and structural reaction injection molding. They are low viscosity, vacuum, high mold temperature, and high pressure.

    Notes: Protein recovery from a bacterial lysate was accomplished using microfiltration membranes in a flat crossflow filter and in a cylindrical rotary filter. Severe membrane fouling yielded relatively low long-term permeate flux values of 10-4-10-3 cm/s (where I cm/s = 3.6 × 104 L/m2 - h). The permeate flux was found to be nearly independent of transmembrane pressure and to increase with increasing shear rate and decreasing solids concentration. The flux increased with shear to approximately the one-third power or greater for the flat filter and the one-half power or greater for the rotary filter; the stronger dependence for the rotary filter is thought to result from Taylor vortices enhancing the back transport of debris carried to the membrane surface by the permeate flow. The average protein transmission or sieving coefficient was measured at approximately 0.6, but considerable scatter in the transmission data was observed. The largest sieving coefficients were obtained for dilute suspensions at high shear rate. The rotary filter provided higher fluxes than did the flat filter for dilute suspensions, but not for concentrated suspensions. © 1995 John Wiley & Sons, Inc.

    Notes: A promising method for reducing membrane fouling during crossflow microfiltration of biological suspensions is backpulsing. Very short backpulses (0.1-1.0 s) have been used to increase the net flux for washed bacterial suspensions and whole bacterial fermentation broths. The net fluxes under optimum backpulsing conditions for the washed bacteria are approximately 10-fold higher than those obtained during normal crossflow microfiltration operation, whereas only a 2-fold improvement in the net flux is achieved for the fermentation broths. A theory is presented that is based on external fouling during forward filtration and nonuniform cleaning of the membrane during reverse filtration. The model contains an adjustable parameter which is a measure of the cleaning efficiency during backpulsing; the cleaning efficiency found by fitting the model to the experiments increases with increasing frequency and duration of the backpulses. The theory predicts an optimum backpulsing frequency, as was observed experimentally. An economic analysis shows that crossflow microfiltration with backpulsing has lower costs than centrifugation, rotary vacuum filtration, and crossflow microfiltration without backpulsing. © 1998 John Wiley & Sons, Inc.     Notes: An automated, 96-well parallel array synthesizer for solid-phase organic synthesis has been designed and constructed. The instrument employs a unique reagent array delivery format, in which each reagent utilized has a dedicated plumbing system. An inert atmosphere is maintained during all phases of a synthesis, and temperature can be controlled via a thermal transfer plate which holds the injection molded reaction block. The reaction plate assembly slides in the X-axis direction, while eight nozzle blocks holding the reagent lines slide in the Y-axis direction, allowing for the extremely rapid delivery of any of 64 reagents to 96 wells. In addition, there are six banks of fixed nozzle blocks, which deliver the same reagent or solvent to eight wells at once, for a total of 72 possible reagents. The instrument is controlled by software which allows the straightforward programming of the synthesis of a larger number of compounds. This is accomplished by supplying a general synthetic procedure in the form of a command file, which calls upon certain reagents to be added to specific wells via lookup in a sequence file. The bottle position, flow rate, and concentration of each reagent is stored in a separate reagent table file. To demonstrate the utility of the parallel array synthesizer, a small combinatorial library of hydroxamic acids was prepared in high throughput mode for biological screening. Approximately 1300 compounds were prepared on a 10 μmole scale (3-5 mg) in a few weeks. The resulting crude compounds were generally 〈80% pure, and were utilized directly for high throughput screening in antibacterial assays. Several active wells were found, and the activity was verified by solution-phase synthesis of analytically pure material, indicating that the system described herein is an efficient means for the parallel synthesis of compounds for lead discovery. © 1998 John Wiley & Sons, Inc.     Notes: A fusion protein was genetically engineered that contains an antimicrobial peptide, designated P2, at its carboxy terminus and bovine prochymosin at its amino terminus. Bovine prochymosin was chosen as the fusion partner because of its complete insolubility in Escherichia coli, a property utilized to protect the cells from the toxic effects of the antimicrobial peptide. This fusion protein was purified by centrifugation as an insoluble inclusion body. A methionine linker between prochymosin and the P2 peptide enabled P2 to be released by digestion with cyanogen bromide. Cation exchange HPLC followed by reversed-phase HPLC were used to purify the P2 peptide. The recombinant P2 peptide's molecular mass was confirmed by mass spectrometry to within 0.1% of the theoretical value (2480.9 Da), and the antimicrobial activity of the purified recombinant P2 against E. coli D31 was determined to be identical to that of the chemically synthesized peptide (minimal inhibitory concentration of 5 mg/mL). Although the yield of the fusion protein after expression by the cells was high (16% of the total cell protein), the percentage recovery of the P2 peptide in the inclusion bodies was relatively low, which appears to be due to losses in the cyanogen bromide digestion step. © 1998 John Wiley & Sons, Inc.
