    Description: Experimentalists, data analysts, and theoreticians were brought together to study and interpret the results of four solstice campaigns conducted prior to the symposium. Emphasis was placed on campaigns III and V. Topics discussed included: review of the data and evaluation of the consistency with available theoretical models; interpretation of the data; recommendations on how the Atmospheric Tides Middle Atmosphere Program (ATMAP) results should reach the open literature such that the advancement of middle atmosphere science and the proprietary rights of the individual experimenters are best served; and recommendations concerning future campaigns and the future emphasis of ATMAP.

    Description: A binational, multidisciplinary research effort in Iceland is directed at an analysis of MSS imagery from ERTS-1 to study a variety of geologic, hydrologic, oceanographic, and agricultural phenomena. A preliminary evaluation of available MSS imagery of Iceland has yielded several significant results - some of which may have direct importance to the Icelandic economy. Initial findings can be summarized as follows: (1) recent lava flows can be delineated from older flows at Askja and Hekla; (2) MSS imagery from ERTS-1 and VHRR visible and infrared imagery from NOAA-2 recorded the vocanic eruption on Heimaey, Vestmann Islands; (3) coastline changes, particularly changes in the position of bars and beaches along the south coast are mappable; and (4) areas covered with new and residual snow can be mapped, and the appearance of newly fallen snow on ERTS-1, MSS band 7 appears dark where it is melting. ERTS-1 imagery provides a means of updating various types of maps of Iceland and will permit the compilation of special maps specifically aimed at those dynamic environmental phenomena which impact on the Icelandic economy.

    Description: The author has identified the following significant results. The wide variety of geological and geophysical phenomena which can be observed in Iceland, and particularly their very direct relation to the management of the country's natural resources, has provided great impetus to the use of ERTS-1 imagery to measure and map the dynamic natural phenomena in Iceland. MSS imagery is being used to study a large variety of geological and geophysical eruptive products, geologic structure, volcanic geomorphology, hydrologic, oceanographic, and agricultural phenomena of Iceland. Some of the preliminary results from this research projects are: (1) a large number of geological and volcanic features can be studied from ERTS-1 imagery, particularly imagery acquired at low sun angle, which had not previously been recognized; (2) under optimum conditions the ERTS-1 satellite can discern geothermal areas by their snow melt pattern or warm spring discharge into frozen lakes; (3) various maps at scales of 1:1 million and 1:500,000 can be updated and made more accurate with ERTS-1 imagery; (4) the correlation of water reserves with snowcover can improve the basis for planning electrical production in the management of water resources; (5) false-color composites (MSS) permitted the mapping of four types of vegetation: forested; grasslands, reclaimed, and cultivated areas, and the seasonal change of the vegetation, all of high value to rangeland management.

    Description: Budgets of O3, NO(x), and NO(y), and acetic acid in the Arctic Boundary Layer Expedition (ABLE 3A) flight region are constructed using photochemical model statistics based on aircraft observations. A Lagrangian model is used to reconstruct the photochemical history of two aged biomass fire plumes sampled by the ABLE 3A aircraft. It is shown that anthropogenic influence on O3 levels in the Arctic may manifest itself not by long-range transport of pollution-derived O3, but rather by a decrease of the regional photochemical sink due to the presence of small amounts of NO(x). The low concentrations of NO(x) measured in ABLE 3A were sufficient to reduce the rate of photochemical loss appreciably relative to a NO(x)-free atmosphere, thus increasing the O3 lifetime. It is shown that decomposition of PAN can account for most of the NO(x) measured below 4-km altitude, but for only 20 percent at 6-km altitude. A lifetime of 29 days is estimated for NO(y) in the ABLE 23A flight region.

    Description: The eddy correlation method was used to measure the net ecosystem exchange of carbon dioxide continuously from April 1990 to December 1991 in a deciduous forest in central Massachusetts. The annual net uptake was 3.7 +/- 0.7 metric tons of carbon per hectare per year. Ecosystem respiration calculated from the relation between nighttime exchange and soil temperature, was 7.4 metric tons of carbon per hectare per year, implying gross ecosystem production of 11.1 metric tons of carbon per hectare per year. The observed rate of accumulation of carbon reflects recovery from agricultural development in the 1800s. Carbon uptake rates were notably larger than those assumed for temperate forests in global carbon studies. Carbon storage in temperate forests can play an important role in determining future concentrations of atmospheric carbon dioxide.

    Description: Eddy correlation flux measurements and concentration profiles of total hydrocarbons (THC) and CO2 were combined to provide a comprehensive record of atmosphere-biosphere exchange for these gases over a 30-day period in July-August 1988 in the Yukon-Kuskokwin River Delta of Alaska. Over 90 percent of net ecosystem exchanges of THC were due to methane. Lakes and wet meadow tundra provided the major sources of methane. The average fluxes from lake, dry tundra, and wet tundra were 11 +/- 3, 29 +/- 3, and 57 +/- 6 mg CH4/sq m/d, respectively. The mean remission rate for the site was 25 mg/sq m/d. Maximum uptake of CO2 by the tundra was 1.4 gC/sq m/d between 1000 and 1500 hrs, and nocturnal respiration averaged 0.73 gC/sq m/d. Net uptake of CO2 was 0.30 gC/sq m/d for the 30 days of measurement; methane flux accounted for 6 percent of CO2 net uptake.

    Notes: In this report, we have compared the physical properties and actin-binding characteristics of several bacterially produced nonmuscle and striated muscle tropomyosins, and we have examined the effects of these isoforms on the interactions of actin with two structurally distinct classes of myosin: striated muscle myosin-II and brush border (BB) myosin-I. All of the bacterially produced nonmuscle tropomyosins bind to F-actin with the expected stoichiometry and with affinities comparable to that of a tissue produced α-tropomyosin, although the striated muscle tropomyosin CTm7 has a lower affinity of F-actin than a tissue-purified striated muscle α tropomyosin. The bacterially produced isoforms also protect F-actin from severing by villin as effectively as tissue-purified striated muscle α-tropomyosin. The bacterially produced 284 amino acid striated muscle tropomyosin isoform CTm7, the 284 amino acid nonmuscle tropomyosin isoform CTm4, and two chimeric tropomyosins (CTm47 and CTm74) all inhibit the actin-activated MgATPase activity of muscle myosin S1 by ∼ 70-85%, comparable to the inhibition seen with tissue-purified striated muscle α tropomyosin. The 248 amino acid tropomyosin XTm4 stimulated the actin-activated MgATPase activity of muscle myosin S1 approximately two- to threefold. The in vitro sliding of actin filaments translocated by muscle myosin-II (2.4 μm/sec at 19°C, 5.0 μm/s at 24°C) increased 25-65% in the presence of XTm4. Tropomyosins CTm4, CTm7, CTm47, and CTm74 had no detectable effect on myosin-II motility. The actin-activated MgATPase activity of BB myosin-I was inhibited 75-90% by all of the tropomyosin isoforms tested, including the 248 amino acid tropomyosin XTm4. BB myosin-I motility (50 nm/s) was completely inhibited by both the 248 and 284 amino acid tropomyosins. These results demonstrate that bacterially produced tropomyosins can differentially regulate myosin enzymology and mechanochemistry, and suggest a role for tropomyosin in the coordinated regulation of myosin isoforms in vivo. © 1994 Wiley-Liss, Inc.

    Notes: Earlier studies have demonstrated that a high (mM) extracellular Ca2+ concentration triggers intracellular [Ca2+] signals with a consequent inhibition of bone resorptive activity. We now report that micromolar concentrations of the divalent cation, Ni2+, elicited rapid and concentration-dependent elevations of cytosolic [Ca2+]. The peak change in cytosolic [Ca2+] increased monotonically with the application of [Ni2+] in the 50-5,000 μM range in solutions containing 1.25 mM-[Ca2+] and 0.8 mM-[Mg2+]. The resulting concentration-response function suggested Ni2+-induced activation of a single class of binding site (Hill coefficient = 1). The triggering process also exhibited a concentration-dependent inactivation in which conditioning Ni2+ applications in the range 5-1,500 μM-[Ni2+] inhibited subsequent responses to a maximally effective [Ni2+] of 5,000 μM. Ni2+-induced cytosolic [Ca2+] responses were not dependent on extracellular [Ca2+]. Thus, when 5,000 μM-[Ni2+] was applied to osteoclasts in Ca2+-free, ethylene glycol bis-(aminoethyl ether) tetraacetic acid (EGTA)-containing medium (≤5 nM-[Ca2+] and 0.8 mM-[Mg2+]), cytosolic [Ca2+] responses resembled those obtained in the presence of 1.25 mM-[Ca2+]. Prior depletion of intracellular Ca2+ stores by ionomycin prevented Ni2+-induced cytosolic [Ca2+] responses, suggesting a major role for intracellular Ca2+ redistribution in the response to Ni2+. The effects of Ni2+ were also modulated by the extracellular concentration of the divalent cations, Ca2+ and Mg2+. When these cations were not added to the culture medium (0 μM-[Ca2+] and [Mg2+]), even low [Ni2+] ranging between 5 pM and 50 μM elicited progressively larger cytosolic [Ca2+] transients. However, the response magnitude decreased at higher, 250-5,000 μM-[Ni2+], resulting in a “hooked” concentration-response curve. Furthermore, increasing extracellular [Mg2+] or [Ca2+] (0-1 mM) diminished the response to 50 μM-[Ni2+], a concentration on the rising phase of the “hook.” Similar increases (0-10 mM) in extracellular [Mg2+] or [Ca2+] increased the response to 5,000 μM-[Ni2+], a concentration on the falling phase of the “hook”. These findings are consistent with the existence of a membrane receptor strongly sensitive to Ni2+ as well as the divalent cations, Ca2+ and Mg2+. Receptor occupancy apparently activates intracellular Ca2+ release followed by inactivation. Furthermore, repriming is independent of intracellular Ca2+ stores, suggesting that such inactivation operates at a transduction step between receptor occupancy and intracellular Ca2+ release. © 1993 Wiley-Liss, Inc.

    Notes: Osteoclasts are known to possess a divalent cation-sensitive receptor, the Ca2+ receptor (CaR). The latter monitors changes in the local Ca2+ concentration generated as a result of hydroxyapatite dissolution. CaR activation elevates cytosolic [Ca2+] and thereby inhibits osteoclastic bone resorption. Recent studies have used Ni2+ as a surrogate CaR agonist to elicit changes in cytosolic [Ca2+]. This article examines the effects of membrane potential changes on the kinetics of the cytosolic [Ca2+] signal resulting from such Ni2+-induced CaR activation. Membrane potential was altered through variations in the extracellular [K+] in combination with applications of the K+ ionophore, valinomycin. Membrane potential changes were confirmed by independent electrophysiological patch clamp studies of whole osteoclasts. The application of valinomycin produced a distinct, sustained elevation of cytosolic [Ca2+] in single fura 2-loaded cells, a “primary” response. This response was independent of valinomycin concentration (between 5 nM to 5 μM) and persisted in Ca2+-free, EGTA-containing solutions. It also persisted both in high (105 mM) and low (5 mM) extracellular [K+]. A gradual “secondary” elevation of cytosolic [Ca2+] then followed with the continued application of valinomycin, but this was eliminated by sequestering the extracellular [Ca2+] or by increasing extracellular [K+] from 5 to 105 mM. In a separate set of experiments, the presence of 5 μM [valinomycin]-([K+] = 5 mM) prolonged the cytosolic [Ca2+] signal elicited by 50 μM-[Ni2+] application. These prolonged kinetics persisted in low extracellular [Ca2+] (zero-added Ca2+), but reverted to a rapid time-course in the presence of 105 mM-[K+] or at higher [Ni2+] (500 μM and 5 mM). The experiments thus indicate that membrane voltage modifies the kinetics of CaR activation by Ni2+ and therefore suggests that the CaR is an integral protein in the osteoclast surface membrane. © 1995 Wiley-Liss, Inc.

    Notes: The influence of aging and dietary restriction on increase in intracellular free calcium ([Ca2+]i) of CD4+ lymphocytes from Macaca mulatta was examined after stimulation with anti-CD3 mAb. We used a flow cytometric assay with the dye indo-1 and either direct or reciprocal immunofluorescent staining to dientify CD4+ cells. After stimulation with anti-CD3 mAb, intracellular free calcium responses were reduced in CD4+ lymphocytes from old male and female ad libitum fed monkeys compared to young and adult male or female monkeys. Old female monkeys had significantly lower [Ca2+]i than did old male monkeys. The reduced responses were in part related to a decreased percentage of responding cells. Dietary restrition of males over a four-year period did not alter [Ca2+]i response compared to ad libitum fed male monkeys. Female monkeys of all ages (which were restricted only for four months) also had similar [Ca2+]i responses to ad libitum fed controls. Our data suggest that age-related changes in [Ca2+]i responses are similar between humans and M. mulatta, and that over these intervals, no effects of caloric restrictions can be detected. © 1995 Wiley-Liss, Inc.
