Notes: The dynamics of a system of two microbial populations having complementary metabolism are investigated by means of simple mathematical models of growth. Complementary metabolism as used here means that each population produces a substance - not present in the initial or feed medium - required by the other for growth. The simple models indicate that (1) something other than lack of the substrate or growth factor produced by its partner must limit the growth of at least one population and (2) the coexistence steady state of such populations in continuous culture is not stable with respect to large perturbations, though it is stable with respect to a wide range of perturbations.

Notes: During the cultivation of E. coli in an airlift tower-loop bioreactor, the following properties were measured: transverse profiles of Sauter bubble diameter, dS; local relative gas holdup, EG; bubble rise velocity, uBS; local mean velocity, ū turbulence intensity, u′; macrotime scale, TEL; dissipation time scale, τE; power spectrum, E(n); and energy dissipation spectrum D(n) at different distances from the aerator. The influence, distance from the aerator, absence and/or presnece of cells, and batch and/or continuous-culture operation on the behavior of the two-phase system are discussed on the basis of these properties.

Notes: Based on the material balance principle applied to microbial reactions in continuous bioprocesses, the concept of reaction rate control has been developed theoretically. This concept provides a more direct way of controlling biological activities than the control of physical or chemical parameters in practice today. From an analysis of dynamic and steady-state experiments, two control systems for carbon dioxide production rate control during the continuous culture of baker's yeast have been designed and evaluated experimentally. In these control methods, intracellular NADH concentration is used as an immediate indication of the onset of glucose repression. A more sophisticated master controller based on the respiratory quotient can be combined with these control methods. The resulting control system provides a means to indirectly optimize biomass production while preventing ethanol formation in the continuous culture of baker's yeast.

Notes: Rheophoresis, i.e. thin-layer electrophoresis in particulate gels with controlled evaporation, combines in one experiment the chromatographic and the electrophoretic characteristics of macromlecules in such a way that they can be determined independently of each other. In this paper theory and experimental corroboration for molecular size determination are presented. We describe the migration velocity of any molecule as the sum of an electrophoretic, an electroosmotic and a rheophoretic component. The latter is the velocity originating from the liquid flow produced by evaporation. It is a function of the position along the gel axis and the cross-section available to the molecule, but totally independent of the molecular charge. As a result of the position dependence, the rheophoretic component can be isolated from the two others and the available cross-section determined by means of a simple plot. Thus, the chromatographic partition coefficient is readily calculated and the effective hydrodynamic radius of the molecule obtained.
