    Notes: Heats of formation, protonation energies, charge distributions, dipole moments, and geometrical structures of thirty-two 1-, 2-, 1,5-, and 2,5-substituted tetrazoles have been investigated by different semiempirical SCF MO calculations. MNDO and, in some cases, AM1 methods are the most acceptable for tetrazole derivatives. A number of linear correlations between available experimental data and the calculated energies and charge characteristics have been established. © 1992 John Wiley & Sons, Inc.

    Notes: Rat brain microvessel endothelial cells were immortalized by transfection with a plasmid containing the E1A adenovirus gene. One clone, called RBE4, was further characterized. These cells display a nontransformed phenotype and express typical endothelial markers, Factor VIII-related antigen and Bandeiraea simplicifolia binding sites. When RBE4 cells were grown in the presence of bFGF and on collagen-coated dishes, confluent cultures developed sprouts that extend above the monolayer and organized into three-dimensional structures. The activity of the blood-brain barrier-associated enzyme, gamma-glutamyl transpeptidase (γGTP), was expressed in these structures, not in the surrounding monolayer. Similar results were obtained with the microvessel-related enzyme alkaline phosphatase (ALP). Addition of agents that elevate intracellular cAMP reduced the formation of three-dimensional structures, but every cell inside the aggregates still expressed γCTP and ALP activities. Such structures, associated with high levels of γCTP and ALP activities, were also induced by astroglial factors, including (1) plasma membranes from newborn rat primary astrocytes or rat glioma C6 cells, (2) C6 conditioned media, or (3) diffusible factors produced by primary astrocytes grown in the presence of, but not in contact with RBE4 cells. RBE4 cells thus remain sensitive to angiogenic and astroglial factors for the expression of the blood-brain barrier-related γCTP activity, as well as for ALP activity, and could constitute the basis of a valuable in vitro model of the blood-brain barrier. © 1994 wiley-Liss, Inc.

    Notes: Programmed death (apoptosis) of the rat myelocytic leukemic cell line IPC-81 was triggered by cyclic adenosine monophosphate (cAMP) analogs or by agents (cholera toxin, prostaglandins) increasing the endogenous cAMP level. The induction of cell death by cholera toxin was preceded by increased activation of cAMP-kinase. Cell lysis started already 5 hr after cAMP challenge and was preceded by internucleosomal DNA fragmentation and morphological changes characteristic of apoptosis. The cell suicide could be prevented by inhibitors of macromolecular synthesis. cAMP analogs induced cell death in a positively cooperative manner (apparent Hill coefficient of 2.9), indicating that triggering of the apoptotic process was under stringent control. There was a strong synergism between cAMP analogs complementing each other in the activation of cAMP-dependent protein kinase I (cAKI). No such synergism was noted for analogs complementing each other in the activation of cAKII. It is concluded that apoptosis can be induced solely by activation of cAKI. The IPC-81 cells expressed about four times more cAKI than cAKII. The expression of cAK subunits, on the protein and mRNA levels, was only minimally affected by cholera toxin treatment.

    Notes: A mutually consistent method to calculate excitation energies and corresponding transition densities is proposed. The method is based on the extended Brillouin's theorem that is derived from the nonstationary variation principle. Within the proposed procedure, the Brillouin's conditions, which appear in this extension, are used as a set of nonlinear equations for molecular orbitals and configuration interaction coefficients of the trial ground- and excited-state functions. The excitation energy is an eigenvalue of the set. To some extent, this procedure is related to the variational treatment of the conventional random-phase approximation within the equation-of-motion method. The basic features of the proposed procedure are discussed and it is illustrated by numerical examples. © 1993 John Wiley & Sons, Inc.

    Notes: In the present paper, preconditioning of iterative equation solvers for the Navier-Stokes equations is investigated. The Navier-Stokes equations are solved for the mixed finite element formulation. The linear equation solvers used are the orthomin and the Bi-CGSTAB algorithms. The storage structure of the equation matrix is given special attention in order to avoid swapping and thereby increase the speed of the preconditioner. The preconditioners considered are Jacobian, SSOR and incomplete LU preconditioning of the matrix associated with the velocities. A new incomplete LU preconditioning with fill-in for the pressure matrix at locations in the matrix where the corner nodes are coupled is designed. For all preconditioners, inner iterations are investigated for possible improvement of the preconditioning. Numerical experiments are executed both in two and three dimensions.

    Notes: Brain Na+ K+ ATPase activity has been found to decrease in experimental trypanosomiasis in rats infected with Trypanosoma congolense. Some physical features that affect membrane fluidity were also observed to be altered. The levels of cholesterol in the brain and free fatty acids in the serum were found to increase in the infected animals. These findings might be relevant to the development of brain lesions.
