    Notizen: M-shell x-ray production/fluorescence (M XRF) cross-sections for five elements with 81 ≤ Z ≤ 92 were measured at 5.96-keV incident photon energy using thin targets. From the measured M XRF cross-sections the average M-shell fluorescence yields were derived. Experimental results for XRF cross-sections and fluorescence yields were compared with theory. The experimental results were found to be higher than the theoretical values by 10-17%. Further experimental investigations on M XRF cross-sections for more elements at different excitation energies are required in order to check the validity of the theoretical parameters used to calculate the M XRF cross-sections.

    Notizen: The individual steps of the consecutive reactions arising from metastable molecular ions, derived from vitamin D3, vitamin D2 and their respective provitamins (7-dehydrocholesterol, ergosterol), were examined in different field-free regions of a triple-sector mass spectrometer of B/E/E geometry. The comparison of the translational energy release (T) and the metastable peak shapes corresponding to these reactions, as well as unimolecular and collision-induced dissociation mass-analysed ion kinetic energy spectra, showed that there are probably two structures of the [M - H2O]+· and [M - CH3·]+ ions depending upon whether the respective ions are formed in the ion source through high-energy reactions, or from the fragmentation of metastable molecular ions through slow, low-energy processes which occur in the first field-free region.

    Notizen: The fragmentation of 1-phenyl-, l-(2′-pyridyl)- and 1-(4′-methyl-2′-quinolyl)-4-acetoacetyI-3-methyIpyrazol-5-ols (compounds 1, 2 and 3, respectively) on electron impact has been studied and the major processes interpreted. The common feature in the mass spectra of these compounds is the loss of ketene, acetonyl radical, acetone and two molecules of ketene from the molecular ion. Whereas the ion generated after the last process, which corresponds to 1-substituted-3-methyIpyrazol-5-ols, loses methyl cyanide in the case of 1, similar ions in the case of 2 and 3 lose Ċ2HO moiety, necessitating an intramolecular hydrogen transfer followed by ring fission and subsequent loss of methyl cyanide. All these and other related processes have been substantiated with the help of accurate mass measurements of the fragment ions and B/E linked-scan spectra.

    Notizen: The 13C NMR spectra of 36 differently substituted 4-methylcoumarins (4-methyl-2H-1-benzopyran-2-ones) have been recorded. The effects of an alkyl side-chain at C-3 on the chemical shift values and that of a methoxy or an acetoxy group in place of a hydroxy group have been observed and rationalized. This study may be helpful in the structure elucidation of new and natural 4-methylcoumarins.

    Notizen: In situ hybridization using nucleic acid probes was used to detect cell- and tissue-specific transcript(s) of embryonic genes during development and differentiation. This highly sensitive technique has the potential to provide valuable information on the regulation of low-abundance housekeeping genes during development. We have determined the experimental conditions required to detect the catalase message in adult mouse liver. Catalase effects the breakdown of H2O2 to O2 and H2O and offers protection against the toxic effects of oxygen radicals. We used a cloned 550 bp BamHI-Pstl fragment from a mouse catalase cDNA (pMCT-1) to generate 35S-labeled sense and antisense riboprobes. The experimental conditions used were sensitive enough to quantitate the abundance of silver grains generated by the antisense riboprobe on the adult liver, a tissue known to be positive for this message. The hybridization protocol was applied to serial sections of 13- and 18-day-old mouse embryos. The results suggest that the catalase expression in the liver and brain begins with somite formation and increases with development and differentiation. On the other hand, this message appears to be absent in mesenchyme, particularly in day 13 embryos. The message in positive tissues appears evenly distributed throughout the cell. The observed expression of the catalase message in the adult liver is approximately six times that in the embryonic liver. It is compatible with the enzyme activity results and emphasizes the sensitivity of the in situ hybridization method (over northern blot, etc.) used in this study.

    Notizen: A simple, convenient and precise technique for deconvolving the Lorentzian (true) Raman linewidth, ΓL, from the observed Raman linewidth, (Δ1/2)R [the FWHM (full width at half-maximum) of the Raman line in question], through polynomial fitting, was developed. The precision of this technique is a consequence of the fact that the values of ΓL/(Δ1/2)R (= Y), obtained by exact numerical evaluation of the Voigt profiles for the Raman bands, were flitted to third- and fourth-degree polynomials in S/(Δ1/2)R (= X), S being the spectral slit width measured as the linewidth (FWHM) of a narrow spectral line recorded at the same slit settings as the Raman line. The procedure of obtaining ΓL, from the knowledge of S,(Δν1/2)R and X, which yields better results, is discussed. Another technique of deconvolution is described and discussed, involving the measurement of Raman linewidths of a particular Raman line for about five or six different slit widths. The measured linewidths (Δ1/2)R, when extrapolated to S → 0, give the value of ΓL directly. A careful comparison with the other deconvolution techniques reveals that the method of polynomial fitting is relatively more precise than other semi-analytical deconvolution techniques, and the extrapolation technique also gives reasonably good results. The applicability of both methods was tested by taking some selected Raman lines.
