    Notes: Summary We studied mice from five strains (BALB/c, C3H/HeSnJ, C57BL/6J, Csb and 129/ReJ) at two ages (young, 10±1 weeks; and old, 67±3 weeks) for the induction of sister chromatid exchanges (SCEs) in vivo by methyl nitrosourea (MNU). The SCE frequency is genotype-specific. The F1 phenotype resembles the ‘low’ responding parent. SCE induction is significantly lower in the older animals of each strain than their younger counterparts, and the reduction of SCE/cell with old age is strain-specific. A general explanation for these results must include strain differences in relative mutagenic sensitivity, genotype-specific pattern of reduction in DNA repair and other such factors affecting SCE formation, with old age.

    Notes: Summary Permanent lymphoblastoid cell lines are important in the molecular analysis and characterization of human genetic disorders, when immortalized cells must be banked for future diagnostic or research purposes. However, routine methods for transformation using Epstein-Barr virus (EBV) require blood volumes that may be difficult to collect from clinically compromised neonates and small children. Here we report a modified transformation procedure utilizing blood samples of small volume (less than 1.0ml), which we have found to be particularly useful for the immortalization of lymphocytes destined for future molecular genetic studies.

    Notes: Rainbow trout were fed for 10 weeks with a nutritionally balanced diet containing 30% by weight of activated sewage sludge. The whole body concentrations of nine heavy metals (Cr, Mn, Fe, Co, Ni, Cu, Zn, Cd, Pb), together with four major cations (Ca, Mg, Na, K) were determined at the beginning and end of the experiment and at three intermediate stages. Fish fed on the diet containing sewage sludge had significantly elevated levels of Cr, Fe, Ni, Pb and reduced levels of Na and K compared with controls, though the values obtained for all groups fell within the range reported for uncontaminated fish. The Ni and Zn showed a marked increase towards the end of the experiment, suggesting that they might have continued to rise after 70 days.

    Notes: Summary The genetic basis of seed setting was evaluated in seven clones of alfalfa selected under predominantly self-pollinating conditions. They were hand crossed in all possible combinations. Their compatibility was studied by the percentage of flowers forming pods and number of seeds per pod during crossing. The variances for GCA, SCA and reciprocal effects were significant for percentage of pod set with a narrow sense héritability of 64 %. This suggested maternal influence of clones on percent pod set, controlled primarily by additive genetic components. GCA was the only significant component for number of seeds per pod with a narrow sense heritability of 71%. There were wide differences between the clones in their relative magnitude of GCA, SCA and reciprocal effects for both traits used as compatibility indexes. Performance of the diallel crosses was judged by studying seed yield and its related characters, namely seeds per pod, dry matter per plant, frost resistance, plant vigor and plant height. Although GCA and SCA variances were significant for all characters, reciprocal differences in general were absent. The SCA values were very high as compared to GCA. Narrow sense heritability values were very low while broad sense heritability were much higher. This suggested that almost none of the variation was due to additive genetic components and all the variability is controlled by interactions of a digenic, trigenic and quadrigenic nature and heterzozygosity. Heterosis was evaluated by comparing the seed yield of single crosses with their mid-parent and high-parent, and very high values were observed. Thus selection of better genes may not be feasible and further improvement in selected clones may have to be brought about by utilization of various interactions and heterosis. An attempt was made to find combinations of characters that may be used for the selection of seed yield but none were found to be satisfactory.

    Notes: Summary Two sets of diallel crosses involving seven tetraploid and five hexaploid alfalfa lines, previously selected for seed and forage yield, are studied. On the basis of combining ability and heritability estimates the following conclusion is drawn. The dominant and epistatic interactions (heterozygosity) are the major components in the expression of variability for yield and characters associated with yield in tetraploid populations. In hexaploid populations, however, there is an appreciable amount of additive variance, and the additive effect of genes along with interaction of various forms may account for the expressed variability for all the characters studied. Better genes and desirable interactions (possibly through heterozygosity) are the two essential genetic components of yield. The selection of better genes is feasible, but by itself will not produce the desired results. Future improvement may therefore depend on the utilization of heterosis and other forms of interactions. There is, in general, a very strong genetic correlation between yield and its components, and therefore, when a direct selection for yield is not feasible, the selection program may rely on a number of closely related characters.

    Notes: Abstract A case of cutaneous infection in a 25-year-old male caused byPaecilomyces variotii is described. Animal pathogenicity studies with normal and cortisone-treated mice revealed the predeliction ofP. variotii for skin and liver in both normal and cortisone-treated mice and for lungs and heart only in immunosuppressed mice. 5-fluorocytosine gave the best MIC value forP. variotii in vitro. This report documents for the first time thatP. variotii causes cutaneous infection.

    Notes: Abstract Aspergillus terreus was isolated from a case of Keratomycosis. The patient, a 50 year old, female presented with a large corneal ulcer with hypopyon. The direct microscopic examination of the scrapings revealed hyaline, thin, septate and branched hyphae. In vitro some antimycotics (amphotericin B,5-fluorocytosine, oxiconazole, amorolfine and ketoconazole) were tested against A. terreus by agar dilution method. Ketoconazole with MIC of 3 gmg/ml after 7 days of incubation was most effective followed by oxiconazole (10μg/ml). Experimental corneal ulcer was produced by injecting intralamellary 0.1 ml of the spore suspension containing 10×106 cfu/ml into the eyes of previously immunocompressed albino rabbits. Histopathologic examination showed infiltration and large destruction of the corneal stroma. Subconjunctival oxiconazole therapy exhibited complete cure. Based on our findings, a clinical evaluation of oxiconazole in human keratomycosis seems to be justified.
