    Effect of apholate and hempa on nucleic acid and protein syntheses in the yellow-fever mosquito (1969)

    Description / Table of Contents: Zusammenfassung Wachstumskurve, Nukleinsäure- und Eiweißgehalt verschiedener Entwicklungsstadien der Gelbfieber-Mücke, Aedes aegypti, wurden untersucht. Die Larven dieser Mücke wurden mit sterilisierenden Dosen der Chemosterilantia Apholate und Hempa behandelt und ihre Wirkungen auf die genannten Parameter geprüft. Das Körpergewicht nahm in den frühen Stadien allmählich zu und zeigte vom Ende des 4. Stadiums bis zur Puppe einen steilen Anstieg. Adulte wogen weniger als die Puppen, Weibchen mehr als Männchen. In den Kontrollen folgte der DNS- und RNS-Gehalt im allgemeinen der Wachstumskurve. Der RNS-Gehalt war bis zum späten 4. Stadium größer als der DNS-Gehalt, in den späteren Stadien kehrte sich das Verhältnis um. Der Eiweißgehalt folgte ebenfalls der Wachstumskurve mit Ausnahme bei den Weibchen, wo er höher war als in der Puppe. Im allgemeinen schien die Behandlung mit den Chemosterilantien Apholate und Hempa den RNS-, DNS- und Eiweißgehalt im ganzen Insekt nicht zu verändern.
    Notes: Abstract The growth curve, nucleic acid and protein content of the various life stages of Aedes aegypti were studied. The larvae of this mosquito were treated with sterilizing doses of the chemosterilants apholate and hempa, and their effects on the above parameters were also investigated. The body-weight increased gradually in the earlier instars and showed a sharp rise from late-fourth instar to the pupa. Adults weighed less than the pupae. Females weighed more than the males. In the controls, DNA and RNA content generally followed the growth curve. RNA content was more than DNA up to the late-fourth instar and the ratio reversed in the later stages. Protein content also followed the growth curve except in adult female, where it was more than in the pupa. In general the treatment with the chemosterilants, apholate and hempa did not seem to alter RNA, DNA and protein content in the whole insect.

    Notes: Abstract Palladium(II) complexes of an imine-thiophene ligand N,N′ bis(1-methylenethiophene)-1,3-diaminopropane and an amine-phenol ligand N,N′-bis(2-hydroxybenzyl)1,3-diamino-2,2-dimethylpropane were synthesized by reacting K2PdCl4 with alcoholic solutions of the ligands. X-ray structures of the complexes were determined using data from a CAD-4 Nonius diffractometer (MoKα radiation). The structures were solved by conventional methods resulting inR factors of 0.044 and 0.061 using 1841 and 2241 independent reflections, respectively. Crystal data: Complex I, orthorhombic, space groupPbca,a=11.323(2),b=14.819(2),c=23.873(3)Å,V=4006(1)Å3,Z=8. Complex II: Space groupP21/c,a=9.169(2),b=31.920(4),c=8.902(6),β=90.38(2)°,V=2605(1)Å3.Z=4. Both complexes showed coordination of the ligands through the nitrogens only while the sulphur and the oxygen atoms were uncoordinated. Square planar coordination is completed bycis chloro ligands.

    Notes: Abstract A palladium(II) complex of an amine-phenol ligand N,N′-bis(2-hydroxybenzyl)1,4-diaminobutane has been synthesized and characterized by NMR, IR, and X-ray diffraction studies. Diffraction data were collected with a Nonius CAD-4 diffractometer with θ-2θ scan and MoKα radiation. The structure was solved by conventional methods resulting in anR factor of 0.022 for 1058 independent reflections. The X-ray structure shows a discrete mononuclear square planar complex in which the Pd is coordinated to the two nitrogens and two oxygens of the ligand. The complex is neutral with the Pd(II) charge balanced by deprotonation of both phenolic oxygens. Crystal data: Space group Pna21,a+8.223 Å,b+22.477 Å,c+9.190 Å;V+1698.8 Å3;Z+4.

Die normalen Männchen haben weniger Hämolymphe (13–16 μl) als Weibchen (18–37 μl) in den ersten 120 h des adulten Lebens. Apholatbehandlung reduziert signifikant das Hämolymphvolumen beider Geschlechter. Es sind signifikant mehr Hämocyten im Umlauf bei adulten Weibchen als bei Männchen. Zählung der Gesamthämocyten zeigt bei behandelten Männchen und Weibchen ein anfängliches Anwachsen bis zu 48 h, später aber eine drastische Reduktion. Die Zahl der Plasmatocyten der behandelten Insekten erwies sich als abnehmend und die Prohämocyten werden der überwiegende Typ, während in der Kontrolle die Plasmatocyten der Haupttyp der Hämocyten sind.
    Notes: Abstract Topical application of sterilizing doses of apholate causes haematological changes in the adults ofDysdercus koenigii. The normal males have less haemolymph (13–16 μl) than females (18–37 μl) for the first 120 hr of adult life. Apholate treatment significantly reduces the haemolymph volume of both the sexes. There are significantly more haemocytes in circulation in adult females than in males. The total haemocyte count shows an initial increase up to 48 hr, but later indicates a drastic reduction in the treated males and females. The plasmatocytes of the treated insects are found to reduce in numbers and the prohaemocytes become the predominant type, while in controls the plasmatocytes are the major type of haemocytes.

Behandlung der Larven mit sterilisierenden Dosen der Chemosterilantien verursachte Hemmung der DNS-Synthese in den Gonaden. Die Hemmung erwies sich in den Hoden als stärker als in den Ovarien. Die Bedeutung dieser Befunde in Beziehung zur Wirkungsweise dieser Chemosterilantien wird diskutiert.
    Notes: Abstract Effect of the chemosterilants, apholate and hempa, on the incorporation of tritiated thymidine in DNA of the gonads of Aedes aegypti was studied. Larval treatment with sterilizing doses of the chemosterilants caused inhibition of DNA synthesis in the gonads. The inhibition was found to be more in testes than in ovaries. The significance of these findings in relation to the mode of action of these chemosterilants is discussed.
