    Notes: Abstract 14-3-3 Proteins are found to bind to a growing number of eukaryotic proteins and evidence is accumulating that 14-3-3 proteins serve as modulators of enzyme activity. Several 14-3-3 protein recognition motifs have been identified and an increasing number of target proteins have been found to contain more than one binding site for a 14-3-3 protein. It is thus possible that 14-3-3 dimers function as clamps that simultaneously bind to two motifs within a single binding partner. Phosphorylation of a number of binding motifs has been shown to increase the affinity for 14-3-3 proteins but other mechanisms also regulate the association. It has recently been demonstrated that fusicoccin induces a tight association between 14-3-3 proteins and the plant plasma membrane H+-ATPase. Phorbol esters and other hydrophobic molecules may have a similar effect on the association between 14-3-3 proteins and specific binding partners.

    Description / Table of Contents: Contents The solution of network problems in power engineering is often simplified by the use of coordinate transformations. These transformations take advantage of the symmetrical properties of electrical equipment. All possible transformations are deduced in this paper. It is found that the well-known oαβ-(Clarke)-, odq-(Park)- and o12-(symmetrical)-coordinates have to be supplemented by certain oGH-coordinates to form a complete set of transformations. These oGH-coordinates generalize the concept of the complex vector as known in the literature. The proper choice of the coordinate system is essential for the efficient use of these transformations. Criteria for the proper choice are given.
    Notes: Übersicht In der elektrischen Energietechnik wird die Lösung von Netzproblemen durch Anwendung von Koordinatentransformationen erleichtert. Dabei werden Symmetrieeigenschaften elektrischer Betriebsmittel ausgenutzt. Die möglichen Transformationen werden hergeleitet. Man stellt fest, daß die bekannten oαβ (Clarke)-, odq (Park)- und o12 (symmetrische)-Komponenten noch um die neu vorgestellten oGH-Komponenten zu ergänzen sind, um ein vollständiges System von Transformationen zu erhalten. Die oGH-Komponenten präzisieren den in der Literatur eingeführten Begriff “Raumzeiger”. Die richtige Wahl des Komponentensystems ist entscheidend für die Nutzung der möglichen Rechenvorteile. Kriterien für die geeignete Auswahl werden angegeben.

    Notes: Summary Cu/Zn Superoxide dismutase (SOD) was quantified by enzyme immunoassay for prenatal diagnosis of Down's syndrome. Overall, 154 samples of amniotic fluid, 72 samples of amniotic cells and 31 samples of chorionic tissue were investigated. Due to the large biological variance of the SOD concentrations in normal pregnancies (range for amniotic fluid 10.5–154.9, for amniotic cells 40.0–338.8, and for chorionic tissue 132.2–649.5 g SOD/g protein) the cases of Down's syndrome detected by karyotype analysis were not reliably identified by Cu/Zn SOD quantification. As in erythrocytes obtained from patients with Down's syndrome, a trisomy 21 was easily and accurately detected in the erythrocytes from very small quantities (about 50 μl) of umbilical blood. The SOD concentrations in normal cases (n = 40) varied between 11.4 and 17.3 and in the cases of trisomy 21, as confirmed by karyotyping (n = 4), between 22.5 and 23.2ng/one million cells. SOD quantification in fetal erythrocyte is a helpful additional method in prenatal Down syndrome diagnosis under certain conditions, which are discussed.

    Notes: Summary Postmortem scanning electron microscopy of human phalanges in a chronic uremic hemodialysis patient with hyperparathyroidism showed the presence of confluent abnormal rounded formations with a radial rosette-like crystalline pattern in the diaphysis as well as in the epiphyseal part of the bones. These fan-shaped configurations were found either as individual formations within bone trabeculae or as numerous aggregated crystalline deposits replacing large parts of the bone structure. The microdissected content of such large areas submitted to X-ray diffraction analysis revealed the predominant presence of calcium oxalate monohydrate or whewellite with some traces of hydroxyapatite. Oxalate titration analysis indicated the presence of 25% of oxalate, corresponding to 45% in weight of whewellite.

    Notes: Abstract Two ornithine carbamoyltransferases (OCT 1 and OCT 2) were isolated from Pseudomonas syringae pv. phaseolicola and purified by precipitation with ammonium sulfate, heat denaturation, chromatography on DEAE-Sephadex A-50 and Sephadex G-200. Molecular weights of both enzymes: 110,000; optimal activity: pH 8.5 to 9.5 (OCT 1), pH 8.4 (OCT 2); apparent K m for ornithine: 7·10-4 (both enzymes); apparent K m for carbamoylphosphate: 7·10-4 (OCT 1), 2.8·10-3 (OCT 2). Both enzymes possess only an anabolic function. OCT 1 is highly inhibited by low concentrations of phaseolotoxin and Orn-P(O)(NH2)-NH-SO3H, OCT 2 is insensitive to both compounds. The inhibition of OCT 1 is reversible.

    Notes: Abstract The production of biomass, polysaccharide storage material and H2 from malate was studied in the wild-type and mutants RdcI, RdcII and RdcI/cII of Rhodobacter capsulatus. The mutants are defective in either copy I, copy II or both copies of the nitrogenase genes nifA and nifB. Stationary phase levels of biomass, polysaccharide and H2 were determined in phototrophic batch cultures grown with 30 mM of d,l-malate and either 2, 5, or 8 mM of ammonium or 7 mM of glutamate. Calculation of the amounts of malate converted into the three products revealed that, at 8 mM of ammonium and 7 mM of glutamate, malate consumption and product formation were balanced. But with decreasing ammonium concentrations malate not converted into biomass was utilized with decreasing efficiency in polysaccharide and H2 formation. This suggests formation of unknown products at the lower ammonium concentrations. Under conditions of optimal N supply, 80% of the malate not used for biomass production was converted by the wild-type and strain RdcII to H2 and CO2. Mutant RdcI exhibited slightly decreased H2 production. The double mutant did not evolve H2 but accumulated increased amounts of polysaccharide. However, the amounts of polysaccharide were lower than should be expected if all of the spare malate, not utilized by the double mutant for H2 production, was converted into storage material. This and incomplete conversion of malate into known products at low ammonium supplies suggests that polysaccharide accumulation does not compete with the process of H2 formation for malate.

    Notes: Abstract The insertion mutant HF231 of Alcaligenes eutrophus H16 failed to grow anaerobically on nitrate and nitrite. When grown under oxygen limitation, mutant HF231 specifically excreted coproporphyrin III, an intermediate of heme biosynthesis. With the help of a Tn5-labeled fragment, we identified and cloned the corresponding wild-type fragment. Sequence analysis of the mutant locus revealed an open reading frame consisting of 1,473 bp, predicting a protein of 491 amino acids that corresponds to a size of 54.2 kDa. In the non-coding upstream region, consensus elements that are indicative for binding sites of the anaerobic transcriptional regulator Fnr were identified. The deduced polypeptide showed extensive sequence similarity with various bacterial oxygen-independent coproporphyrinogen III oxidases designated HemN. HemN catalyzes the oxidative decarboxylation of coproporphyrinogen III to yield protoporphyrinogen IX. Anaerobic growth on nitrate and nitrite of mutant HF231 was restored by introducing the hemN gene of A. eutrophus or of Pseudomonas aeruginosa on a broad-host-range vector. Likewise, the A. eutrophus hemN complemented heme biosynthesis of a Salmonella typhimurium hemF/hemN double mutant during anaerobic and aerobic growth. Analysis of a transcriptional lacZ gene fusion showed that expression of hemN in A. eutrophus is nitrate-independent and repressed by oxygen.
