    Notes: Abstract Adaptation of solitary corals, Fungia repanda and F. echinata, and their zooxanthellae to low light and ultraviolet light B (UV-B) was studied with respect to changes in their protein contents, photosynthetic pigment contents and the photosynthesis-irradiance (P-I) curves. The corals were collected from 1 to 50 m depths in the Republic of Belau (Paulau) in 1990 and 1991. The chlorophyll a content in a unit surface area of the coral did not change significantly with the depth of the habitat, whereas cellular chlorophyll a in the algae increased with the depth. Zooxanthellae density and protein content in a unit surface area of Fungia spp. decreased with the depth. Photosynthetic parameters normalized by a unit surface area of the Fungia spp., maximum gross photosynthetic rate (P gmax area-1) and dark respiration rate (R area-1), were negatively correlated with the depth, while initial slope of the P-I curve (α) did not show significant correlation with the depth. Compensation light intensity (Ic) decreased with the depth. In isolated zooxanthellae, P max chl a -1, and R chl a -1 decreased with the depth, while αchl a was constant. P gmax cell-1 and R cell-1 did not change significantly but αcell increased with the depth. Ic decreased with the depth as in the intact corals. Reduction of protein content in a unit area of the coral from deeper habitat implies decrease of host animal tissues. Reduction of Ic can be explained by decrease of R area-1, which may be due to the diminution of animal tissues. The photoadaptational response to low light intensity of intact Fungia spp. was found to be a combination of the photoadaptation of symbiotic algae and the decrease of host animal tissue. In order to study their adaptation to ultraviolet (UV) radiation, P-I curves of Fungia spp. and isolated zooxanthellae were analyzed before and after UV-B irradiation. 1 h UV-B irradiation showed no effect on the photosynthetic rate of the shallow water (1 m) corals, while it inhibited the photosynthesis of the deep water (30 m) corals and zooxanthellae isolated from both shallow and deep water corals. These results indicate that the host, Fungia spp., in shallow water have protective mechanism for intense UV-B in their habitat. These photoadaptational mechanisms seem to allow the Fungia spp. to have wide vertical distribution where light intensity spans more than two orders of magnitude.

    Notes: Abstract. Organic molecules adhered to the point of a micropipette were transferred onto the surface of a polymer film by pulsed laser light guided through a quartz optical fiber which was inserted into the micropipette. Controlled by a high-resolution piezoelectric driver designed for scanning probe microscopy, the distance between the micropipette point and the polymer surface was kept to a few tens of nm. After laser irradiation, the deposited molecules on the polymer surface were examined with optical transmission, fluorescence, atomic force, and scanning near-field optical microscopy. It was demonstrated that various kinds of organic molecules can be implanted into sub-micrometer regions at the surfaces of polymer films without their decomposition. The hypothetical mechanism of the molecular transfer is discussed in relation to the morphology of crystalline molecules adhered at the micropipette apertures.

    Notes: Summary The plasma (or serum) protein binding of disopyramide (DSP) in five haemodialysis patients was studied using an ultrafiltration technique. There was an increase in the free fraction of DSP in the plasma on dialysis days in comparison to the levels on interdialysis days, which was associated with an elevation of the free fatty acid levels in the plasma together with the increase of the free fraction of DSP. The inhibitory effect of free fatty acids on DSP binding in an in vitro study was enhanced in proportion to their concentrations, and was shown to be due to competition at one binding site by experiments with oleic acid as a representative displacer. Certain endogenous organic acids, such as indoxyl sulphate, 2-hydroxyhippuric acid and hippuric acid, which are characteristically elevated in chronic renal failure, scarcely affected the protein binding of DSP. The findings indicate that free DSP should be monitored in patients with elevated plasma free fatty acid levels, such as those on haemodialysis therapy.

    Notes: Summary The binding of disopyramide (DSP) to plasma (or serum) proteins was determined using an ultrafiltration technique in three patients undergoing haemodialysis. An increase in the free fraction (FF) of DSP during dialysis occurred together with elevation of the free fatty acid (FFA) level in plasma. The effect of FFA on protein binding in vitro was examined using DSP- and FFA-spiked solutions containing human α1-acid glycoprotein and serum albumin. The FF of DSP rose in proportion to increasing FFA levels, supporting the in vivo observations. The findings suggest that the free concentration of DSP should be routinely monitored, especially in haemodialysis patients.

    Notes: Summary The effect of three new fluoroquinolones on theophylline kinetics and the urinary excretion of metabolites was studied in 5 healthy subjects (3 male, 2 female). All subjects received serial, single i.v. infusions of theophylline (aminophylline, 250 mg) over 60 min after 200 mg doses of a quinolone (enoxacin, ofloxacin, norfloxacin) every 8 h for 3 consecutive days, the quinolone being administered up to the day following theophylline administration. Pretreatment with ofloxacin and norfloxacin did not influence theophylline disposition, but theophylline clearance fell from 0.054 to 0.027 l·h−1·kg−1 in the presence of enoxacin, without a change in the apparent volume of distribution. Enoxacin, too, was the sole compound to increase the urinary excretion of theophylline (33.2 vs 43.9 mg, before vs after treatment), and significantly to decrease the excretion of 3-methylxanthine (3-MX), 1-methyluric acid (1-MU) and 1,3-dimethyluric acid (1,3-DMU) in 24-h urine samples (from 19.8 to 7.16 mg, from 28.3 to 10.3 mg and from 68.8 to 49.5 mg, respectively). The effect of the quinolones on hepatic drug metabolizing enzyme activity was investigated in each subject using the ratios of 6-hydroxycortisol to total 17-hydroxycorticosteroids and to free cortisol in 24-h urines as an index of the hepatic P-450-dependent enzyme system. No significant difference in ratio was observed between control and other treatments. It is concluded that the theophylline-enoxacin interaction was largely due to inhibition of a metabolic system other than the common hepatic P-450 system.

    Notes: Abstract Octacalcium phosphate(OCP), Ca8(HPO4)2(PO4)4·5H2O, consists of alternative stackings of layers with an apatitic structure and a brushite-like composition. Here we consider whether or not OCP is able to complex with organic substances. The interplanar spacing (d100) of OCP prepared in the presence of dicarboxylates (RC2O4 2−; R=organic group) expanded from the original value of 18.7 Å to 19.2–26.1 Å depending on the length of R. Examples of R were CnH2n(n=1–6), CH(CH3)CH2, C(CH3)=CH, CH=CH, CH2CH=CHCH2 and C6H4. Structural considerations and experimental data suggested that dicarboxylates were incorporated into the OCP structure through the replacement of HPO4 2− by RC2O4 2−.

The sigmoidal curves plotting serum parathyroid hormone (PTH) against serum Ca in primary hyperparathyroidism and secondary hyperparathyroidism due to renal failure deviate to the right. We previously found the leftward curve shift in PTH-deficient hypoparathyroidism. In the present study, we investigated the curve shift in pseudohypoparathyroidism (PHP) with secondary hyperparathyroidism due to target organ resistance to PTH. In renal failure the sigmoidal curves move to the left after vitamin D3 treatment. We also examined the effect of vitamin D3 on the curve shift in pseudohypoparathyroidism (PHP) and idiopathic hypoparathyroidism (IHP). Before vitamin D3 treatment, the sigmoidal curve deviated to the left in both types of hypoparathyroidism. After vitamin D3 treatment it moved to the right. These results indicate that vitamin D3 and/or extracellular Ca modify the relationship between PTH and Ca dynamics even in hypoparathyroid disorders with decreased or increased maximum serum PTH. Following vitamin D3 treatment, the point plotting baseline serum PTH against baseline serum Ca moved to the right at first in accordance with the rightward shift of the sigmoidal curve and then the point moved downward in PHP or downward in IHP. These changes suggest that vitamin D3 resets PTH secretion at a higher extracellular Ca level at first and then suppresses it in a time-dependent manner. 1,25(OH)2D3 and/or extracellular Ca may be the determinant factors of the sigmoidal curve shift in hypoparathyroid disorders. Mechanisms other than the Ca sensing system error may contribute to the curve shift.
