    Notes: Abstract Optical characterization is presented for a silicon oxide amorphous matrix (prepared by the sol–gel method) doped with chlorophyll. The gel fabrication procedure and the doping process are first described. Then the optical behaviour from ultraviolet to far infrared is analysed. Gel samples with and without chlorophyll are studied morphologically by SEM in order to determine the spatial homogeneity of the gel matrix and the chlorophyll distribution. Finally, a mathematical model is proposed for behaviour of the gel-doped absorption coefficient as a function of chlorophyll concentration.

    Notes: Abstract The paper deals with the problem of building an inventory of information systems for the public administration, with reference to an ongoing project in Italy. We describe the investigation techniques defined for collecting information and the techniques developed for a systematic analysis of the large set of conceptual schemas resulting from the investigation. These schemas describe the data used by the public administration work processes. In particular, we describe the conceptual schema of the inventory, which is the basis for discussing the methodology of investigation, the choice of units of investigation, the data collection and merging, and the access to information. Then, we present the schema analysis techniques developed to analyse semi-automatically the large set of conceptual schemas resulting from the investigation. In particular, we illustrate indexing techniques for identifying representative descriptors of schemas and similarity techniques to compare schemas for their classification into families. Finally, the tool developed to support the storage, analysis and classification of schemas is described and experimentation results are discussed.

    Notes: Summary The most relevant properties of hypercycles were previously studied mainly from a theoretical point of view. We have developed a Monte Carlo method simulating hypercyclic organization to obtain information about the dynamics of this prebiotic organization. Nucleation, growth, and selective properties have been tested and the results obtained are in good agreement with those of the theoretical predictions. The influence of hypercyclic organization of the “error threshold” has also been studied. As a consequence of the emergence of a hypercycle, the value of this threshold decreases. The amount of this decrease depends on the population size. Moreover, for some interval of quality factor values, either the hypercycle organization or an error catastrophe can be produced, depending on the initial conditions. The influence of these phenomena on both the dynamic behavior and evolutionary advantages of the hypercycle, as well as their decisive roles on genome size, are discussed.

    Notes: Abstract HLA-A2 antigens are divided into four subtypes, designated A2.1 to A2.4, by the use of cytolytic T lymphocytes (CTL). The A2.4 subtype consists of a functionally heterogeneous group of variants that are not recognized by A2.1-, A2.2-, or A2.3-specific CTL lines while it is indistinguishable from A2.1 by isoelectric focusing. The structure of an A2.4 variant expressed on donor KNE has been established by comparative peptide mapping with A2.1 and radiochemical sequencing. It was found to differ from A2.1 by a single amino acid change of Cys to Tyr at position 99. This position is only moderately polymorphic and has not previously been found to vary in any other HLA or H-2 variants. The nature of the change is compatible with its generation by one-point mutation from A2.1. The only other previously characterized A2.4 variant, CLA, differs from A2.1 by a single amino acid replacement at position 9. Both residues 9 and 99 are located in homologous positions within the α1 and α2 domains, respectively. The results shown contribute to the molecular interpretation of the heterogeneity of CTL recognition within the HLA-A2.4 group of antigens.

    Notes: Abstract The Y chromosome gene SRY plays an important role in normal male sexual development and is thought to be the testis-determining factor. We describe a familial nonsense mutation in SRY, shared by two XY sisters with complete gonadal dysgenesis and, in a mosaic manner, by their father. This mutation, consisting of a C to T transition in position 1 of codon 97 of SRY, results in a truncated peptide with an incomplete DNA-binding domain. The mutation is also present in the father of the two cases, but a portion of wild-type SRY also remains. Our data suggest that the father suffered a postzygotic mutation early in development, but that he retained a remnant of functional SRY protein that accounts for his normal development.

    Notes: Abstract The structure of an HLA-A2.4 functional variant (A2.4c) expressed on donor KLO has been examined by comparative peptide mapping with other HLA-A2 antigens of known structure and radiochemical sequencing. All the peptide differences between A2.4c and A2.1 could be accounted for by five amino acid changes at positions 9, 43, 66, 95, and 156. The nature of residues 9, 43, and 95 in A2.4c was determined by sequencing to be identical to those in A2.2Y. The nature of residue 156 in A2.4c was also assigned as identical to that in A2.2Y on the basis of the identity of the corresponding peptide in its chromatographic comparison with A2.2Y. Position 66 was unique to A2.4c. It was determined to be an Asn residue instead of the Lys present in all other HLA-A2 antigens of known structure. This was the only detected amino acid difference between A2.4c and A2.2Y. The results indicate that, from a structural point of view, A2.4c is most closely related to the A2.2 subtype antigens and not to other A2.4 antigens. The data are compatible with the assumption that A2.4c was derived from A2.2Y by a single point mutation event.

    Notes: Abstract The primary structure of the HLA-A2 subtype A*0204 (isoelectric focusing variant A2.A) has been determined. cDNA encoding this subtype was amplified by the polymerase chain reaction. Four independent full-lenght cDNA clones encoding A*0204 were analyzed to obtain a consensus sequence for this subtype. A*0204 differs from A*0201 by a single nucleotide change of G to T through the coding regions, resulting in an Arg to Met change at position 97. This substitution accounts for the isoelectric focusing pattern of the subtype. The same change occurs in other HLA-A specificities in association with other changes in its vicinity. The absence of additional substitutions in A*0204 suggests that it could have arisen from A*0201 by point mutation, and that recurrent mutations may take place during HLA diversification. The spatial location of this change implies that A*0204 must be a functional variant. Comparison of its sequence with other HLA-A2 subtypes reveals that much of the HLA-A2 subtype polymorphism is generated by variations in four neighboring positions, including position 97, which are located in two adjacent β-strands on the floor of the peptide binding site of the molecule.

    Notes: Abstract The HLA-A2 antigen expressed by donor OZB can be distinguished from the main HLA-A2.1 subtype by isoelectric focusing - it is one charge unit more acidic — and by some alloreactive T-cell clones but not by cytolytic T lymphocyte lines. The structure of variant OZB has been examined by comparative peptide mapping with A2.1 and radiochemical sequence analysis. The two molecules were found to differ in a single tryptic peptide from the 0 region, spanning residues 220–243. The amino acid sequence of this peptide from variant OZB revealed that there was only one amino acid change of Glu instead of Ala at position 236, a hitherto invariant residue in class I HLA antigens. All previously characterized HLA or H-2 natural variants have structural changes restricted to the α 1 and/or α2 domains. Thus, variant OZB is unique in that (1) it has one amino acid change in α3 and (2) it has no changes in αl and α2. The only detected substitution of this variant may be accounted for by a single base change at the DNA level, suggesting that it might have resulted from a point mutation in the A2.1 gene. The structural features of variant OZB open a novel way to examine the influence of polymorphism in α3 on cytolytic T-cell recognition of naturally occurring class I antigens.
