    Notes: Abstract A system is described for the automatic sorting of zooplankton by isopycnic sedimentation at one gravity in density gradients of the silica sol Ludox AM containing 1% w/v sodium trimetaphosphate or 1% w/v dextran sulfate. The apparatus, called a “Rho Spectrometer”, consists of a 15.6–1 cylinder, a moveable piston for displacing the gradient from the top, and an ejector tube for collecting successive layers of the gradient. A gradient generator suitable for up to 30 Rho Spectrometers is also described. In a typical separation, 100 ml or so of preserved zooplankton are layered over a 13–1 gradient, which is 0 to 45% v/v in Ludox AM. Following an overnight period of sedimentation and equilibration, fish eggs are recovered from the top of the gradient, invertebrates from the central part of the gradient, and fish larvae from the bottom of the gradient. In many cases 90% of the fish larvae are separated from 95% of the invertebrates. Rho spectrometry should facilitate the analysis of large numbers of plankton samples through a substantial decrease in the amount of manual sorting required. However, in its present state of development, it is not suitable for all plankton samples; the zone of herring larvae, for example, is strongly overlapped by a number of invertebrates. In addition, samples heavily contaminated with Phaeocystis spp. or hydroids fail to sediment freely.

    Notes: Abstract  The ability to introduce and express foreign genes can greatly facilitate in vitro studies of the molecular biology of an organism. We report here the first use of modified retroviral vectors to achieve foreign gene expression in primary cultured cells from the Oka organ and ovary of the blue shrimp Penaeus stylirostris (Litopenaeus) stylirostris (Holthuis 1980). Of the heterologous promoters tested, the Moloney murine leukemia virus and Rous sarcoma virus promoters mediated the highest levels of reporter gene expression. Firefly luciferase and Escherichia coliβ-galactosidase were successfully used as reporter genes in cultured cells from both Oka organ and ovary. These in vitro systems can be exploited for the study of shrimp cellular biology and regulation of gene expression.

    Notes: Abstract A model is presented for the operation of the striatum. The model posits that the basal ganglia are responsible for driving smooth transitions of state for an organism. We propose that this is accomplished through the computation of a potential function within the striatum on which a gradient descent is performed toward the goal state. The model suggests that various somatotopic regions of the striatum correspond to state spaces, each of which pertains to a different aspect of the organism. This paper discusses this model only in the context of motor control, i.e., egomotion and limb movement. The model appears to account for a variety of experimental results, and for some unusual properties of the striatum.

    Notes: Abstract. The juxtaposition of hypokinetic and hyperkinetic symptoms in Parkinson's disease (PD) presents a challenge in modeling the basal ganglia. We propose a model of the striatum that can account for the mixture of symptoms seen in PD. In the model, the problem of motor planning is cast in terms of a particle in a potential, where potentials are generated internally in striatal modules, subject to afferent control. Planned movement is governed by Hamilton's equations, where potential energy is supplied by potentials expressed in the striatum. To test the model in realistic situations, a dynamic simulation of a two-link robot arm was used. Normal movement is modeled and shown to exhibit observed experimental properties. Symptoms of PD are reproduced by modeling hypothetical consequences of PD pathology.

    Notes: Summary Tissue proteins believed to contain a relatively high concentration of C-terminal carboxyl groups emit an intense blue fluorescence after being treated with first a hot mixture of acetic anhydride and pyridine, second salicylhydrazide and last zinc acetate. Characteristically they do not fluoresce when the zinc treatment is omitted. Muscular tissues emit the strongest fluorescence, but normally neither mucosubstances nor loose connective tissues fluoresce at all. These and other results are consistent with Barrnett and Seligman's view that acetic anhydride in the presence of hot pyridine transforms the C-terminal carboxyl groups of proteins into methyl ketones. They do not support Karnovsky's more recent theory that hot acetic anhydride more or less exclusively converts side-chain carboxyl groups of proteins into mixed acid anhydrides instead.

    Notes: Abstract Hypocotylary explants obtained from 30- to 40-day-old slash pine (Pinus elliottii, Engelm.) seedlings treated with 6-benzylaminopurine produced multiple buds that eventually elongated into axillary shoots. The explants were pulse treated (45-s dip) with 6-benzylaminopurine (22.2, 111, 222 μM) plus a control and cultured on three different basal media containing activated charcoal (0.5% w/v). Hormonal concentration and basal medium were compared for the number and size of axillary shoots induced after 12 and 29 days. The greatest number of axillary shoots was produced by explants that were pulse treated with 111 μM 6-benzylaminopurine and cultured on Gresshoff and Doy medium. The axillary shoots were fewer in number per explant than shoots previously reported resulting from hormonally induced advantitious buds of slash pine, but the axillary shoots developed more rapidly.

    Notes: Abstract Peanut (Arachis hypogaea L.) somatic embryos were produced from the embryo axes of mature, dry seeds of cultivar GK-7. Percent embryogenic explants ranged from 88–100% using 10–40 mg/1 of 2,4-D in the induction medium. Neither 2,4-D concentration nor photoperiod during the induction period had a large effect on percent embryogenesis, mean number of embryos per explant, or embryo morphology. However, embryos obtained from cultures grown in the dark were easier to remove from the explant than those under a 16-h photoperiod. Somatic embryos developed on the epicotyl portion of the embryo axis, primarily on the young, expanding leaves. A survey of 14 genotypes indicated that genotype had a large influence on embryogenic capacity, with all genotypes being embryogenic to some extent. The ability to recover somatic embryos from axes of harvested, stored seeds represents significant advantages for the establishment of peanut embryogenic cultures, including the use of simple sterilization procedures and a constant source of explant tissue.
