Notes: A practical approach towards evaluating form deviations of 2D complex contour profiles from the coordinate measurement data of discrete points is proposed. In this approach, a 2D complex contour is represented approximately by the combination of straight line and circular arc segments under the condition of satisfying the prespecified interpolation accuracy. The form deviation of a measured point is calculated relative to the approximate contour and the form deviation of the whole contour is evaluated using either the minimum zone criterion (MZC) or the least squares criterion (LSC). It has been demonstrated that the difference between the deviation definitions, one relative to the original contour and the other to the approximate contour, is not larger than the prespecified accuracy value, i.e. the evaluation precision of the form deviation can be sufficiently guaranteed. The effectiveness and the efficiency of the approach are verified with a practical example of a planar cam contour. Moreover, a weighted LSC technique is proposed in order to obtain a rational evaluation result for the measured points with non-uniform length interval.

Notes: Abstract Three hydrographic surveys were carried out in Deep Bay, which is located in the eastern part of Pearl estuary between Shenzhen, the most successful special economic zone of PRC, and Hong Kong. Data on current, size distributions of bottom sediment, suspended solids, and some water quality parameters were obtained. This information is of value for mathematical modeling of tidal circulation and sediment transport in the bay, and also useful in the planning of further development in this area. The sedimentation rate in Deep Bay was estimated by two different approaches, viz. comparison of historical navigation maps and210Pb dating. Information obtained from the maps indicated that average sedimentation rate between 1898 and 1949 was about 8 mm/yr, while a 15 mm/yr figure was obtained from the210Pb dating for recent years.

Notes: Summary A new enzyme-linked immunosorbent assay (ELISA) was used to study the kinetics of transformation of aflatoxin B1 into aflatoxin M1 in lactating mice. Aflatoxin M1 concentration in the milk samples reached a maximum 30 min after injection of aflatoxin B1 and decreased thereafter. At the maximum time, the levels of aflatoxin M1 in the samples were proportional to the dosages administered. Aflatoxin B1 was also detected in the milk samples but at a lower concentration.

Notes: Abstract  The proteasome regulator PA28, which can be upregulated by IFN, is important in the modulation of proteasome activity. Since the proteasome has been implicated in the processing of the major histocompatibility complex (MHC) class I antigens, it was of interest to determine the regulatory elements of PA28 at the genomic level. Although PA28 has been found in different species, the gene layout on the chromosome was not determined. In this study, the genetic organization of mouse PA28b was characterized. Two copies of the PA28b gene, namely b1 and b2, were found by restriction fragment mapping and Southern hybridization. By fluorescence in situ hybridization, the location of the two PA28b genes was determined on chromosomes 11 and 14. PA28b1 has 11 exons, whereas PA28b2 has no introns and appears to be a nonfunctional pseudogene. The 5' promoter region of PA28b1 contains several transcriptional factor binding sites including two IFN responsive elements. The expression levels of PA28 and other gene products involved in MHC class I antigen presentation appear to be correlated in various tissues. Notably, PA28 is expressed at high levels in immunological tissues such as spleen and peripheral blood leukocytes. Taken together, PA28 seems to be co-regulated with other molecules involved in MHC class I antigen presentation.

Notes: Methylaspartate ammonia lyase (MAL) catalyses the reversible α,β-elimination of ammonia from L-threo-(2S,3S)-3-methylaspartic acid to give mesaconic acid. Crystals of Citrobacter amalonaticus MAL have been obtained by the hanging-drop method of vapour diffusion using ammonium sulfate as the precipitant. Three crystal forms were obtained from identical crystallization conditions, two of which (forms A and B) diffract to high resolution, whilst the third form diffracted poorly. Crystals of form A diffract to beyond 2.1 Å and have been characterized as belonging to one of the enantiomorphic space groups P4122 or P4322, with unit-cell parameters a = b = 66.0, c = 233.1 Å, α = β = γ = 90° and a monomer in the asymmetric unit. Crystals of form B diffract to beyond 1.5 Å and belong to space group C222, with unit-cell parameters a = 128.3, b = 237.4, c = 65.8 Å, α = β = γ = 90° and a dimer in the asymmetric unit. Determination of the structure of MAL will be an important step in resolving current conflicts concerning the enzyme mechanism which differ between one which places MAL as a member of the superfamily of ammonia lyases whose catalytic activity requires a cofactor formed by post-translational modification of the enzyme and another which links MAL to the enolase superfamily.

Notes: Crystal structures of the active-site mutants D99A and H48Q and the calcium-loop mutant D49E of bovine phospholipase A2 have been determined at around 1.9 Å resolution. The D99A mutant is isomorphous to the orthorhombic recombinant enzyme, space group P212121. The H48Q and the calcium-loop mutant D49E are isomorphous to the trigonal recombinant enzyme, space group P3121. The two active-site mutants show no major structural perturbations. The structural water is absent in D99A and, therefore, the hydrogen-bonding scheme is changed. In H48Q, the catalytic water is present and hydrogen bonded to Gln48 N, but the second water found in native His48 is absent. In the calcium-loop mutant D49E, the two water molecules forming the pentagonal bipyramid around calcium are absent and only one O atom of the Glu49 carboxylate group is coordinated to calcium, resulting in only four ligands.

Notes: The fourth-order elastic constants (FOEC) of Laue groups RI, RII, HI, CII, and I have been calculated with the method of Hearmon. By use also of the results of Ghate for other Laue groups, the FOEC schemes for all crystal classes have been worked out. By the method of direct inspection the fifth- and sixth-order elastic constants have also been calculated for Laue groups N, M, O, TI, TII, CI, and CII. The number of independent constants for each Laue group agrees with the group-theoretical predictions.

Notes: The detective quantum efficiency (DQE) characterizes the measurement uncertainty introduced by a detector. A formulation of the DQE for CCD and vidicon-based X-ray integrating detectors is developed and illustrated with five model detectors. A comparison of the calculated and measured DQEs for two of these detectors is presented. From an examination of the DQEs for the model detectors, generalizations are made regarding detector designs.
