    Notizen: The complete intracellular cycle of five cloned stocks of Trypanosoma cruzi was quantified. Marked but stable interclonal differences were found in the length of the pre-replicative lag period (18.2-34.2 h), amastigote doubling lime (8.6-21.5 h), and duration of the complete intracellular cycle (96-215 h). Strong correlations were demonstrated between these characteristics as well as to the growth rate of the epimastigote stage of the same clones grown in liquid medium. These data demonstrate that the marked heterogeneity of the natural population of T. cruzi extends to the intracellular cycle of the parasite and has important implications for our understanding of Chagas’ disease.

    Notizen: . Restriction endonuclease mapping of the Trypanosoma cruzi kinetoplast DNA maxicircle was performed in nine cloned stocks using maxicircle probes from T. brucei. Analysis of the maxicircle 13–15-kbp encoding region allowed cloned stocks to be divided into three groups: A, B, and C. Parasites from groups A and B had 3% sequence divergence while parasites from group C showed 16–17% sequence divergence with regard to parasites from groups A and B. Cross-hybridization experiments demonstrated that the 23–25-kbp maxicircle divergent region was similar in sequence in group A and B, but different in group C parasites. The high homology of the T. cruzi and T. brucei encoding regions allowed the use of T. brucei probes to detect T. cruzi transcripts, whose overall picture did not differ for parasites from any of the nine assayed clones.

    Notizen: The infection of bovine embryo skin and muscle cells by trypomastigotes of four Trypanosoma cruzi clones (CA-I/71, /72, Miranda/76, /80) was quantified. Stable and reproducible intra-isolate differences were observed; an almost 70-fold difference in infectivity occurred between clones. The CA-I/71 clone was not susceptible to N-acetyl-D-glucosamine at a concentration that inhibits the infection of vertebrate cells by Ernestina and Y-strain parasites. Eight other monosaccharides that are common constitutents of vertebrate cell surface glycoproteins also failed to inhibit the infection of vertebrate cells by the CA-I/71 clone.

    Notizen: Nineteen clones of Trypanosoma cruzi were obtained as single-cell isolates from Triatoma infestans. Ten of the clones were isolates from a patient with chronic Chagas' disease; nine clones were isolates from a dog infected with T. cruzi strain CA-I isolated originally from a chronic chagasic patient. The growth kinetics and peak modal Coulter volume of these clones were characterized. Significant inter- and intra-group differences between growth rates and peak modal volumes were found. These data indicate that subpopulations and, consequently, genetic heterogeneity of T. cruzi exist in chronic chagasic patients. All of the clones infected vertebrate cells in vitro.

    Notizen: . Total DNA/organism was determined by flow cytometry on stocks of 33 single-cell-isolate clones and one strain of mithramycin-stained Trypanosoma cruzi. Interstrain differences in mean total DNA/group of 34% and interclone differences in total DNA/organism of 41% were found. Microspectrofluorometric analyses of the trypomastigote stage of selected clones confirmed the flow cytometry data and indicated that the total DNA/organism differences were due to differences in DNA of both the nucleus and kinetoplast with the nucleus being the major contributing factor. These data imply that the potential for genetic diversity in T. cruzi may be very large.

    Notizen: An extracellular serine peptidase, purified from the culture supernatant of the sub-Arctic psychrophilic bacterium strain PA-43, is monomeric, with a relative molecular mass of 76 000, and an unusually low pI of 3.8. The peptidase is active towards N-succinyl AAPF p-nitroanilide and N-succinyl AAPL p-nitroanilide, indicating a chymotrypsin-like substrate specificity. It is inhibited by the serine peptidase inactivator phenylmethylsulfonyl fluoride, but not by EDTA or EGTA, suggesting that added metal ions are not necessary for activity. The enzyme is most active at pH 8.3 and at 55–60°C, although it is unstable at 60°C. It is nevertheless remarkably stable for an enzyme from a psychrophilic microorganism, remaining active after 1 week at 20°C and after five freeze–thaw cycles. Comparison of the N-terminal 40 amino acid residues with other archived sequences revealed highest similarity to the alkaline serine protease (aprx) from Bacillus subtilis.

    Notizen: : To adequately manage impacts of ongoing or future land use changes in a watershed, the magnitude of their hydrologic impacts needs to be assessed. A grid based daily streamflow model was calibrated with two years of observed streamflow data, using time periods when land use data are available and verified by comparison of model predictions with observed streamflow data. Streamflow data were separated into direct runoff and baseflow to estimate the impacts of urbanization on each hydrologic component. Analysis of the ratio between direct runoff and total runoff from 30 years of simulation results and the change in these ratios with urbanization shows that estimated annual direct runoff increased from 49.2 percent (1973) to 63.1 percent (1984) and 65.0 percent (1991), indicating the effects of urbanization are greater on direct runoff than on total runoff. The direct runoff ratio also varies with annual rainfall, with dry year ratios larger than those for wet years. This suggests that the impact of urbanization on areas that are sensitive to runoff ratios, such as stream ecosystems, might be more serious during drier years than in wetter years in terms of water quality and water yield. This indicates that sustainable base-flow is important to maintaining sound stream ecosystems.

    Notizen: : An index of watershed susceptibility to surface water contamination by herbicides could be used to improve source water assessments for public drinking water supplies, prioritize watershed restoration projects, and direct funding and educational efforts to areas where the greatest environmental benefit can be realized. The goal of this study is to use streamflow and herbicide concentration data to develop and evaluate a method for estimating comparative watershed susceptibility to herbicide loss. United States Geological Survey (USGS) concentration data for five relatively water soluble herbicides (alachlor, atrazine, cyanazine, metolachior, and simazine) were analyzed for 16 Indiana watersheds. Correlation was assessed between observed herbicide losses and: (1) a herbicide runoff index using GIS-based land use, soil type, SCS runoff curve number, tillage practice, herbicide use estimates, and combinations of these factors; and (2) predicted herbicide losses from a non-point source pollution model (NAPRA-Web, an Internet-based interface for GLEAMS). The highest adjusted R2 value was found between herbicide concentration and the runoff curve number alone, ranging from 0.25 to 0.56. Predictions from the simulation model showed a poorer correlation with observed herbicide loss. This indicates potential for using the runoff curve number as a simple herbicide contamination susceptibility index.

    Notizen: : The U.S. Department of Agriculture Curve Number (CN) method is one of the most common and widely used techniques for estimating surface runoff and has been incorporated into a number of popular hydrologic models. The CN method has traditionally been applied using compositing techniques in which the area weighted average of all curve numbers is calculated for a watershed or a small number of sub-watersheds. CN compositing was originally developed as a time saving procedure, reducing the number of runoff calculations required. However, with the proliferation of high speed computers and geographic information systems, it is now feasible to use distributed CNs when applying the CN method. To determine the effect of using composited versus distributed CNs on runoff estimates, two simulations of idealized watersheds were developed to compare runoff depths using composite and distributed CNs. The results of these simulations were compared to the results of similar analyses performed on an urbanizing watershed located in central Indiana and show that runoff depth estimates using distributed CNs are as much as 100 percent higher than when composited CNs are used. Underestimation of runoff due to CN compositing is a result of the curvilinear relationship between CN and runoff depth and is most severe for wide CN ranges, low CN values, and low precipitation depths. For larger design storms, however, the difference in runoff computed using composite and distributed CNs is minimal.
