    Notes: Abstract The structural gene encoding the Rieske iron-sulfur protein from Thermus thermophilus HB8 has been cloned and sequenced. The gene encodes a protein of 209 amino acids that begins with a hydrophilic N-terminus followed by a stretch of 21 hydrophobic amino acids that could serve as a transmembrane helix. The remainder of the protein has a hydrophobicity pattern typical of a water-soluble protein. A phylogenetic analysis of 26 Rieske proteins that are part of bc 1 or b 6 f complexes shows that they fall into three major groups: eubacterial and mitochondrial, cyanobacterial and plastid, and five highly divergent outliers, including that of Thermus. Although the overall homology with other Rieske proteins is very low, the C-terminal half of the Thermus protein contains the signature sequence CTHLGC-(13X)-CPCH that most likely provides the ligands of the [2Fe-2S] cluster. It is proposed that this region of the protein represents a small domain that folds independently and that the encoding DNA sequence may have been transferred during evolution to several unrelated genes to provide the cluster attachment site to proteins of different origin. The role of individual residues in this domain of the Thermus protein is discussed vis-a-vis the three-dimensional structure of the bovine protein (Iwata et al., 1996 Structure 4, 567–579).

    Notes: Abstract An MIP-AES element specific detector has been directly coupled to an HPLC separation system by means of hydraulic high-pressure nebulization (HHPN) with an appropriate desolvation unit. This system was applied to Cr(III)/Cr(VI) speciation analysis. The linear dynamic range of Cr(III) and Cr(VI) determination was two orders of magnitude, absolute detection limits were 13 ng for Cr(III) and 18 ng for Cr (VI), respectively. The matrix effect caused by the easily ionizable elements (Na, Ca, K) was significant in real samples.

    Notes: Abstract Taxol (NSC-125973) is a poorly soluble plant product that exhibits excellent antimitotic properties. This study involves the development of a new formulation for taxol. The stability of taxol in a 50% triacetin emulsion as well as possible methods of intravenous administration of this dosage form was examined. A stable emulsion was found at taxol concentrations of 10 and 15 mg/ml of emulsion. The 50% triacetin emulsion showed an intravenous LD50 of 1.2 ml/kg in Swiss-Webster mice. The 10 mg/ml taxol formulation was demonstrated to be stable upon addition to 5% dextrose iv fluids provided that small packing systems were used. The taxol–triacetin emulsion can also be intravenously injected at various rates, and it may prove to be a useful formulation for taxol.

    Notes: Summary Isolated cardiac cells from bullforg atrial tissue can be readily prepared by digestion of intact fragments of atrial tissue with trypsin and collagenase. These isolated cells have dimensions of about 5 μm in width and range in length from 300 μm to over 500 μm. Such isolated cells may prove useful for the investigation of contractile activity of cardiac muscle at the single cell level and at the sarcomere level within the single cell.

    Notes: Abstract The studies reported here suggest that the immunomodulatory effects of 1,1-dimethylhydrazine (UDMH) are associated, in part, with interference with interleukin 2 (IL-2) regulatory action. Concanavalin A (Con A)-stimulated DNA synthesis in murine splenocytes was inhibited from 18.6 to 44.1% at sub-toxic concentrations of UDMH (10 to 50 μg/ml) and IL-2-dependent DNA synthesis in CTLL-20 cells was inhibited from 11.3 to 41.58% at subtoxic concentrations of UDMH (10 to 50 μg/ml). In addition, UDMH suppressed phorbol myristic acetate (PMA)-stimulated IL-2 production in EL-4 cells by up to 30% and slightly suppressed IL-2 production by Con A-stimulated murine splenocytes. In all cases, inhibition was evident at sub-toxic UDMH concentrations and was demonstrated to be independent of inactivation of IL-2 or interference with IL-2 absorption. It is suggested that UDMH has the potential to modify immune function through interference with IL-2 production and especially the lymphoproliferative response to IL-2.

    Notes: Summary A series of double mutant homozygotes have been produced from three wilty tomato mutants; flacca, sitiens and notabilis. The phenotypic interaction between the mutant genes has been studied. The severity of phenotype in the double mutants does not correspond to that predicted from the single mutant homozygotes. The results are discussed in relation to the probable involvement of the mutants in abscisic acid metabolism.

    Notes: Abstract Laboratory tests were developed to allow identification of the effects of toxicants on the predator-prey interaction between Hydra oligactis Pallas and Daphnia magna Strauss. Three test methods, which differed with respect to initial Daphnia population, test period, and management of predation level, were used to determine the influence of the organochlorine insecticide lindane on the interaction. The criteria examined were the population structures of the two species. The lindane treatments did not alter the production of Hydra. However, the final population structure of Daphnia was variously affected by the toxicant in the different test systems. The most sensitive endpoint, with a lowest observed effect concentration of 4 μg/L lindane determined after a 14 day exposure period, was increased recruitment of juvenile Daphnia in a test system which incorporated management of the Hydra. The results are compared to previously reported data derived from single species chronic tests, and the suitability of the test methods for hazard evaluation is discussed.

    Notes: Abstract It has recently been shown thatnitrogen contained in humic substances can be releasedas low molecular weight species through photochemicalprocesses or reactions. Nitrogen released in thismanner may then become available for biological useand chemical reactivity. This report providesadditional kinetic data for nitrogen photochemistry oforganic matter in natural waters from Louisiana andfor Suwannee River humic and fulvic acids. Naturalwater samples were fractionated into colloidal(〉0.2 μm, 〈1000 Dalton) and dissolved (〉1000Dalton) fractions and the kinetics of these fractionsas well as 0.2 μm filtered water samples weremeasured. For irradiations of less than 10 hours ofsimulated noon-time sun, samples showed linearproduction of ammonium and first-order fading at350 nm. Ammonium photoproduction was enhanced asmuch as three-fold by lowering pH, but linear kineticswere maintained. Light-dark cycling showed continuedrelease of ammonium immediately after irradiation,followed by uptake at longer post irradiation times.A two step mechanism was proposed to explain theobserved kinetics. Calculated net daytime release ofammonium from natural surface waters ranged from 0.33to 1.9 μM h-1 in the samples studied,corresponding to a photic zone conversion rate of9–20% of the total organic nitrogen per day intoammonium.
