    Description: With the advent of molecular phylogenetics, the systematics and taxonomy of Patellogastropoda have been greatly improved. Eight families and 36 genera are currently recognized in the order. We review recent published papers that have used molecular data, discuss the resulting advances in systematics and evolution of Patellogastropoda and suggest directions of future research.

    Description: This study was designed to elucidate details of the structure and formation process of the alternate lamellar pattern known to exist in lamellar bone. For this purpose, we examined basic internal lamellae in femurs of young rats by transmission and scanning electron microscopy, the latter employing two different macerations with NaOH at concentrations of 10 and 24%. Observations after the maceration with 10% NaOH showed that the regular and periodic rotation of collagen fibrils caused an alternation between two types of lamellae: one consisting of transversely and nearly transversely cut fibrils, and the other consisting of longitudinally and nearly longitudinally cut fibrils. This finding confirms the consistency of the twisted plywood model. The maceration method with 24% NaOH removed bone components other than cells, thus allowing for three-dimensional observations of osteoblast morphology. Osteoblasts extended finger-like processes paralleling the inner bone surface, and grouped in such a way that, within a group, the processes arranged in a similar direction. Transmission electron microscopy showed that newly deposited fibrils were arranged alongside these processes. For the formation of the alternating pattern, our findings suggest that: (1) osteoblasts control the collagen fibril arrangement through their finger-like process position; (2) osteoblasts behave similarly within a group; (3) osteoblasts move their processes synchronously and periodically to promote alternating different fibril orientation; and (4) this dynamic sequential deposition of fibrils results in the alternate lamellar (or twisted plywood) pattern.

    Description: p53 is one of the most important known tumor suppressor genes, and it is inactivated in approximately half of human cancers. p53 family members execute various functions, such as apoptosis induction and cell cycle arrest, by modulating transcriptional regulation. Therefore, the direct transcriptional targets of the p53 family must be explored to elucidate the functional mechanisms of family members. To identify the direct transcriptional targets of p53 family members, we performed chromatin immunoprecipitation together with next-generation sequencing (ChIP-seq) and searched for p53-binding motifs across the entire human genome. Among the identified ChIP-seq peaks, approximately half were located in an intergenic region. Therefore, we assumed large intergenic non-coding RNAs (lincRNAs) to be major targets of the p53 family. Recent reports have revealed that lincRNAs play an important role in various biological and pathological processes, such as development, differentiation, stemness and carcinogenesis. Through a combination of ChIP-seq and in silico analyses, we found 23 lincRNAs that are upregulated by the p53 family. Additionally, knockdown of specific lincRNAs modulated p53-induced apoptosis and promoted the transcription of a gene cluster. Our results suggest that p53 family members, and lincRNAs constitute a complex transcriptional network involved in various biological functions and tumor suppression.

    Description: This study aimed at elucidating whether estrogen deficiency would affect the synthesis of an osteocyte-derived factor, sclerostin, in the mesial region of alveolar bone. Eight 9-week-old Wistar female rats were ovariectomized (OVX) and eight other rats were Sham-operated (Sham). After 4 weeks, the interradicular septa of mandibular first molar were embedded in paraffin and then histochemically examined. Sclerostin-positive osteocytes were located in the superficial layer of the mesial region of Sham bones, whereas the OVX mesial region showed less sclerostin-reactive osteocytes. There was no significant difference in the distribution of estrogen receptor α and terminal deoxynucleotidyl transferase-mediated deoxyuridinetriphosphate nick end-labeling -positive cells in the groups studied. The Sham mesial region featured many osteoclasts, and OVX specimens showed numerous osteoclasts in association with intense immunolabeling of the receptor activator of the nuclear factor kB ligand. Contrary to the observations in Sham specimens, a complex meshwork of cement lines was seen in the OVX mesial region, accompanied by an irregularly distributed osteocytic lacunar–canalicular system. In conclusion, estrogen deficiency appears to inhibit osteocyte-derived sclerostin synthesis in the mesial region of the interradicular septum, in a process that seems to be mediated by accelerated bone remodeling rather than by direct effects on osteocytes.

    Description: We perform a weak-lensing study of the nearby cool-core galaxy clusters, Hydra A ( z  = 0.0538) and A478 ( z  = 0.0881), of which the brightest cluster galaxies (BCGs) host the powerful activities of active galactic nuclei (AGNs). For each cluster, the observed tangential shear profile is described well by either a single Navarro–Frenk–White model or a two-component model including the BCG as an unresolved point mass. For A478, we determine the BCG and its host-halo masses from a joint fit to weak-lensing and stellar photometry measurements. We find that the choice of initial mass functions (IMFs) can introduce a factor of 2 uncertainty in the BCG mass, whereas the BCG host-halo mass is constrained well by data. We perform a joint analysis of the weak-lensing and stellar kinematics data available for the Hydra A cluster, which allows us to constrain the central mass profile without assuming specific IMFs. We find that the central mass profile ( r  〈 300 kpc) determined from the joint analysis is in excellent agreement with those from independent measurements, including dynamical masses estimated from the cold gas disc component, X-ray hydrostatic total mass estimates, and the central stellar mass estimated with the Salpeter IMF. The observed dark matter fraction around the BCG for Hydra A is found to be smaller than those predicted by adiabatic contraction models, suggesting the importance of other physical processes, such as AGN feedback and/or dissipationless mergers.

    Description: RASSF6, a member of RASSF tumour suppressor proteins, binds to mammalian Ste20-like kinases (MST1/2), core kinases of the proapoptotic Hippo pathway and cooperates with the Hippo pathway to induce apoptosis. We originally identified RASSF6 as a putative interactor of membrane-associated guanylate kinase inverted (MAGI)-1 by the yeast two-hybrid screening. We used human kidney cDNA library for the screening. MAGI-1 is abundantly expressed in kidney and is a core component of the slit diaphragm. These findings suggest that RASSF6 is expressed in kidney. However, the function of RASSF6 in kidney is not yet studied. We performed this study to confirm the interaction of RASSF6 with MAGI-1, to analyse the expression of RASSF6 in kidney and to gain insight into the function of RASSF6 in kidney. RASSF6 binds to PDZ domains of MAGI-1 through its C-terminal PDZ-binding motif and is coimmunoprecipitated with MAGI-1 from rat liver. RASSF6 is localized in normal kidney glomerulus but disappears when the slit diaphragm is disrupted in nephrotic kidney. RASSF6 is also localized on apical membranes in renal proximal tubular epithelial cells. We demonstrated that RASSF6 as well as the Hippo pathway are involved in the sorbitol-induced apoptosis in immortalized human proximal renal tubular epithelial HK-2 cells.

    Description: Ti 0.6 Fe 0.4 O 2 and Ti 0.8 Co 0.2 O 2 nanosheets are Fe- and Co-doped titanium oxides, respectively, and they are synthesized by the exfoliation of lepidocrocite-type layered titanates. We have investigated these nanosheets by electron energy-loss spectroscopy (EELS) using a monochromated transmission electron microscope. The energy-loss near-edge structures (ELNESs) of Fe- L and Co- L indicate that Fe 3+ and Co 2+ ions are substituted in the octahedral sites in each nanosheet. The Ti- L edges of Ti 0.6 Fe 0.4 O 2 and Ti 0.8 Co 0.2 O 2 nanosheets correspond to the octahedral coordination of Ti 4+ and oxygen atoms as well as an undoped titania nanosheet (Ti 0.87 O 2 ). On the other hand, the electron transitions from 2p 3/2 to 3d e g in Ti- L 3 regions are different in each nanosheet. We have also investigated the electron-beam-induced damage of Ti 0.6 Fe 0.4 O 2 and Ti 0.8 Co 0.2 O 2 nanosheets. The results indicated that Fe 3+ ions in the Ti 0.6 Fe 0.4 O 2 nanosheets were selectively reduced to Fe 2+ ions in the reduction process by electron irradiation. In contrast, the chemical shift of the Ti- L edge of the Ti 0.8 Co 0.2 O 2 nanosheets indicated that Ti 4+ ions were reduced. These results suggest that the substitution of 3d metals in titania nanosheets affects their crystal and electronic structures and material properties such as their long-range atomic configuration and reduction mechanism.

    Description: Social network analysis provides a broad and complex perspective on animal sociality that is widely applicable to almost any species. Recent applications demonstrate the utility of network analysis for advancing our understanding of the dynamics, selection pressures, development, and evolution of complex social systems. However, most studies of animal social networks rely primarily on a descriptive approach. To propel the field of animal social networks beyond exploratory analyses and to facilitate the integration of quantitative methods that allow for the testing of ecologically and evolutionarily relevant hypotheses, we review methodological and conceptual advances in network science, which are underutilized in studies of animal sociality. First, we highlight how the use of statistical modeling and triadic motifs analysis can advance our understanding of the processes that structure networks. Second, we discuss how the consideration of temporal changes and spatial constraints can shed light on the dynamics of social networks. Third, we consider how the study of variation at multiple scales can potentially transform our understanding of the structure and function of animal networks. We direct readers to analytical tools that facilitate the adoption of these new concepts and methods. Our goal is to provide behavioral ecologists with a toolbox of current methods that can stimulate novel insights into the ecological influences and evolutionary pressures structuring networks and advance our understanding of the proximate and ultimate processes that drive animal sociality.

    Description: Phosphatidylinositol (PtdIns) is a membrane phospholipid composed of diacylglycerol and a D - myo -inositol head group. In mammals, the hydroxyl groups at the D3, D4 and D5 positions of the inositol ring can be phosphorylated to yield seven phosphoinositide derivatives. PtdIns-3,5-bisphosphate [PtdIns(3,5)P 2 ] is the most recently discovered species of phosphoinositide that is generated by the phosphorylation of PtdIns(3)P at the D5 position by PtdIns phosphate kinase and catabolized through the dephosphorylation by myotubularin family of phosphatases. Genetic and biochemical analyses of the enzymes metabolizing PtdIns(3,5)P 2 have revealed that this phospholipid is involved in the control of endolysosomal systems and plays crucial roles in various mammalian tissues. In this article, we review the current state of knowledge of the metabolic/physiological functions of PtdIns(3,5)P 2 , and describe how disruption of these functions may contribute to human diseases.

    Description: The shells of a broad range of land snails were digitally imaged in the near-ultraviolet (NUV), visible range (VIS) and near infrared (NIR). NIR images were recorded in both incident and transmitted illumination. In most cases, shell and periostracal pigmentation observed in the VIS was completely translucent in the NIR, while its contrast was enhanced in the NUV. Exceptions to the above rule fit into four main categories. (1) Snails with green or tan shells or periostraca, presumably functional as camouflage among vegetation, were often highly absorbing in the NUV, thus matching the optical characteristics of green vegetation in this range. (2) Pigmented spiral stripes in the shells of several Camaenidae and Helicidae were adjacent to nonpigmented areas that display a heightened reflectivity and reduced translucence in the VIS and NIR. This enhanced the contrast of the colour pattern in the VIS, but appeared to lack functions in the NIR. (3) Snails from arid or desert environments exposed to high levels of sunlight often have largely white shells, highly reflective throughout the studied range of wavelengths. This is likely an adaptation to reduce the thermal effects of solar irradiation, and may also be a form of masquerading camouflage. (4) Among numerous Bradybaenidae and a few Camaenidae, Helicidae, Orthalicidae and Enidae, reflective patches or stripes, white in the VIS, were present in the periostracum or outermost shell layer. These structures were highly reflective (and highly opaque to transmitted radiation) across the NUV, VIS and NIR. They may have a dual function as disruptive camouflage in the NUV and VIS, and as reflectors to reduce the thermal effects of solar irradiation in the VIS and NIR.
