    Description: Primary triple-negative breast cancers (TNBCs), a tumour type defined by lack of oestrogen receptor, progesterone receptor and ERBB2 gene amplification, represent approximately 16% of all breast cancers. Here we show in 104 TNBC cases that at the time of diagnosis these cancers exhibit a wide and continuous spectrum of genomic evolution, with some having only a handful of coding somatic aberrations in a few pathways, whereas others contain hundreds of coding somatic mutations. High-throughput RNA sequencing (RNA-seq) revealed that only approximately 36% of mutations are expressed. Using deep re-sequencing measurements of allelic abundance for 2,414 somatic mutations, we determine for the first time-to our knowledge-in an epithelial tumour subtype, the relative abundance of clonal frequencies among cases representative of the population. We show that TNBCs vary widely in their clonal frequencies at the time of diagnosis, with the basal subtype of TNBC showing more variation than non-basal TNBC. Although p53 (also known as TP53), PIK3CA and PTEN somatic mutations seem to be clonally dominant compared to other genes, in some tumours their clonal frequencies are incompatible with founder status. Mutations in cytoskeletal, cell shape and motility proteins occurred at lower clonal frequencies, suggesting that they occurred later during tumour progression.     Description: Many long non-coding RNAs (lncRNAs) affect gene expression, but the mechanisms by which they act are still largely unknown. One of the best-studied lncRNAs is Xist, which is required for transcriptional silencing of one X chromosome during development in female mammals. Despite extensive efforts to define the mechanism of Xist-mediated transcriptional silencing, we still do not know any proteins required for this role. The main challenge is that there are currently no methods to comprehensively define the proteins that directly interact with a lncRNA in the cell. Here we develop a method to purify a lncRNA from cells and identify proteins interacting with it directly using quantitative mass spectrometry. We identify ten proteins that specifically associate with Xist, three of these proteins--SHARP, SAF-A and LBR--are required for Xist-mediated transcriptional silencing. We show that SHARP, which interacts with the SMRT co-repressor that activates HDAC3, is not only essential for silencing, but is also required for the exclusion of RNA polymerase II (Pol II) from the inactive X. Both SMRT and HDAC3 are also required for silencing and Pol II exclusion. In addition to silencing transcription, SHARP and HDAC3 are required for Xist-mediated recruitment of the polycomb repressive complex 2 (PRC2) across the X chromosome.     Description: We review some emerging trends in transduction, connectivity and data analytics for Point-of-Care Testing (POCT) of infectious and non-communicable diseases. The patient need for POCT is described along with developments in portable diagnostics, specifically in respect of Lab-on-chip and microfluidic systems. We describe some novel electrochemical and photonic systems and the use of mobile phones in terms of hardware components and device connectivity for POCT. Developments in data analytics that are applicable for POCT are described with an overview of data structures and recent AI/Machine learning trends. The most important methodologies of machine learning, including deep learning methods, are summarised. The potential value of trends within POCT systems for clinical diagnostics within Lower Middle Income Countries (LMICs) and the Least Developed Countries (LDCs) are highlighted.

    Description: Soil moisture is an integral quantity parameter in hydrology and agriculture practices. Satellite remote sensing has been widely applied to estimate surface soil moisture. However, it is still a challenge to retrieve surface soil moisture content (SMC) data in the heterogeneous catchment at high spatial resolution. Therefore, it is necessary to improve the retrieval of SMC from remote sensing data, which is important in the planning and efficient use of land resources. Many methods based on satellite-derived vegetation indices have already been developed to estimate SMC in various climatic and geographic conditions. Soil moisture retrievals were performed using statistical and machine learning methods as well as physical modeling techniques. In this study, an important experiment of soil moisture retrieval for investigating the capability of the machine learning methods was conducted in the early spring season in a semi-arid region of Iran. We applied random forest (RF), support vector machine (SVM), artificial neural network (ANN), and elastic net regression (EN) algorithms to soil moisture retrieval by optical and thermal sensors of Landsat 8 and knowledge of land-use types on previously untested conditions in a semi-arid region of Iran. The statistical comparisons show that RF method provided the highest Nash–Sutcliffe efficiency value (0.73) for soil moisture retrieval covered by the different land-use types. Combinations of surface reflectance and auxiliary geospatial data can provide more valuable information for SMC estimation, which shows promise for precision agriculture applications.

    Description: In this work, we present a battery-less wireless Micro-Electro-Mechanical (MEMS)-based respiration sensor capable of measuring the respiration profile of a human subject from up to 2 m distance from the transceiver unit for a mean excitation power of 80 µW and a measured SNR of 124.8 dB at 0.5 m measurement distance. The sensor with a footprint of ~10 cm2 is designed to be inexpensive, maximize user mobility, and cater to applications where disposability is desirable to minimize the sanitation burden. The sensing system is composed of a custom UHF RFID antenna, a low-loss piezoelectric MEMS resonator with two modes within the frequency range of interest, and a base transceiver unit. The difference in temperature and moisture content of inhaled and exhaled air modulates the resonance frequency of the MEMS resonator which in turn is used to monitor respiration. To detect changes in the resonance frequency of the MEMS devices, the sensor is excited by a pulsed sinusoidal signal received through an external antenna directly coupled to the device. The signal reflected from the device through the antenna is then analyzed via Fast Fourier Transform (FFT) to extract and monitor the resonance frequency of the resonator. By tracking the resonance frequency over time, the respiration profile of a patient is tracked. A compensation method for the removal of motion-induced artifacts and drift is proposed and implemented using the difference in the resonance frequency of two resonance modes of the same resonator.

    Description: This paper presents a new constant frequency, direct grid-connected wind-based induction generator system (IGS). The proposed system includes a six-phase cage rotor with two separate three-phase balanced stator windings and a three-phase SV-PWM inverter which is used as a STATCOM. The first stator winding is connected to the STATCOM and is used to excite the machine. The main frequency of the STATCOM is considered to be constant and equal to the main grid frequency. In the second stator winding, the frequency of the induced emf is equal to the constant frequency, so the generator output frequency is independent of the load power demand and its prime mover speed. The second stator winding is directly connected to the main grid without an intermediate back-to-back converter. In order to regulate the IGS output active and reactive power components, a sliding mode control (SMC) is designed. Assuming unbalanced three-phase voltages for the main grid, a second SMC is developed to remove the machine output’s negative sequence currents. Moreover, a conventional PI controller is used to force the average exchanging active power between the machine and STATCOM to zero. This PI controller generates the reference value of the rotor angular speed. An adjustable speed pitch angle-controlled wind turbine is used as the IGS’s prime mover. The effectiveness and capability of the proposed control scheme have been supported by the simulation results.

    Description: In this paper, a sliding mode control is presented for direct torque and stator flux control of interior permanent magnet synchronous motor in a rotor speed sensorless drive system. The control scheme is developed in a specific synchronous rotating reference frame (X-Y) in which the stator current space vector coincides with the direct (X) axis. For this control technique no need to have any knowledge of machine parameters such as stator two-axis inductances, rotor permanent magnets flux linkage, and even the rotor initial position. However, the on-line actual stator resistance value is required to estimate the stator flux components in the stator stationary two-axis reference frame. In this control strategy, two simple methods are described for estimating the rotor speed and stator resistance. Some simulation and experimental results are presented to support the validity and effectiveness of the proposed control scheme.

    Description: Unlike men, who are disproportionately affected by severe disease progression and mortality from COVID-19, women may be more affected by the economic, social and psychological consequences of the pandemic. Psychological distress and mental health problems are general risk factors for increases in the use of alcohol and other substances as a dysfunctional coping mechanism. Methods: An analysis was carried out of the female subset (n = 2153) of a population-based, cross-sectional online survey (October–December 2020), covering the “second wave” of the COVID-19 pandemic in Germany. Results: Among women, 23% increased their alcohol use, 28.4% increased their nicotine use and 44% increased their illicit substance use during the COVID-19 pandemic. Twenty percent reported major depressive symptoms and 23.4% symptoms of generalized anxiety. Generalized anxiety proved to be a significant predictor of increases in alcohol and nicotine use in logistic regression. Discussion: The mental health burden remained high during the second wave of COVID-19 and alcohol, nicotine and other substance use increased. However, the association between mental health and substance use was weak. Psychological distress does not seem to be the main motivator of substance use.
