    Notes: Summary Determination of Magnesium by Spectrofluorimetry and Synchronous Scanning First and Second Derivative Spectrofluorimetry with 2-Quinizarinsulpbonate An analytical method has been developed for the fluorimetric determination of microgram amounts of magnesium in solution. The method is based on the reaction of magnesium with 2-quinizarinsulphonate. Synchronous scanned first and second derivative fluorimetry has been employed to increase the sensitivity of the method. The influence of reaction variables as well as instrumental parameters and the composition of the complex is discussed. The interference of various foreign cations and anions has also been examined and in some cases eliminated or reduced by addition of cyanide.

    Notes: Summary The establishment of suspension cell cultures from trimmed cotyledons of pepper (Capsicum annuum L.) provides a new experimental system for studying the relationship between release of peroxidase (EC 1.11.1.7) into the free intercellular spaces and plant cell growth. In contrast with several other species, the total peroxidase activity in the medium increased continuously during the post-exponential growth phase of the pepper cell culture, and this was correlated with the growth inhibition of pepper cells cultivated in suspension. The increase in the peroxidase activity in the culture medium was the consequence of a differential release of isoperoxidases, prominently marked by a primary release of basic isoperoxidases, followed by a strong increase in the level of acidic isoperoxidases. Thus, pepper cells cultures constitute a new experimental system for studying the regulation of the sequential release of basic and acidic isoperoxidases, which occurs during the growth cessation of plant cells.

    Notes: Abstract Inorganic-carbon transport was investigated in the eukaryotic marine microalgaeStichococcus minor, Nannochloropsis oculata and aMonallantus sp. Photosynthetic O2 evolution at constant inorganic-carbon concentration but varying pH showed thatS. minor had a greater capacity for CO2 rather than HCO 3 − utilization but forN. oculata andMonallantus HCO 3 − was the preferred source of inorganic carbon. All three microalgae had a low affinity for CO2 as shown by the measurement of inorganic-carbon-dependent photosynthetic O2 evolution at pH 5.0. At pH 8.3, where HCO 3 − is the predominant form of inorganic carbon, the concentration of inorganic carbon required for half-maximal rate of photosynthetic O2 evolution [K 0.5 (CO2)] was 53 μM forMonallantus sp. and 125 μM forN. oculata, values compatible with HCO 3 − transport. Neither extra- nor intracellular carbonic anhydrase was detected in these three microalgal species. It is concluded that these microalgae lack a specific transport system for CO2 but that HCO 3 − transport occurs inN. oculata andMonallantus, and in the absence of intracellular carbonic anhydrase the conversion of HCO 3 − to CO2 may be facilitated by the internal pH of the cell.

    Notes: Abstract Air-grown cells of a marine, small-celled (2 μm diameter) strain of Stichococcus bacillaris contained appreciable carbonic-anhydrase activity but this was repressed when cells were grown on air enriched with 5% (v/v) CO2. Assay of carbonic-anhydrase activity using intact cells and cell extracts showed all activity was intracellular in this Stichococcus strain. Measurement of inorganic-carbon-dependent photosynthetic O2 evolution at pH 5.0, where CO2 is the predominant form of inorganic carbon, showed that the concentration of inorganic carbon required for half-maximal rate of photosynthetic O2 evolution [K0.5(CO2)] was 4.0 μM for both air- and CO2-grown cells. At pH 8.3 the K0.5(CO2) was 0.3 mM for air-grown and 0.6 mM for CO2-grown cells. Sodium ions did not enhance bicarbonate utilization. Measurement of the internal inorganic-carbon pool (HCO 3 − +CO2) by the silicone-oil-layer centrifugal filtering technique showed that air- and CO2-grown cells were able to concentrate inorganic carbon up to 20-fold in relation to the external medium at pH 5.0 but not at pH 8.3. In this alga the high affinity for CO2 and inorganic-carbon accumulation in CO2- and air-grown cells results from active CO2 transport that is not dependent on carbonic-anhydrase activity.

    Notes: Summary Over three years, theAdh and α-Gpdh loci have been studied in two cellar populations ofDrosophila melanogaster and in two field populations which were each near to one of the cellars. Analyses of gene frequencies indicate that the divergence among subpopulations is greater in theAdh locus than in the α-Gpdh locus. Selection for or againstAdh S allele acting on theIn(2L)t inversion influences of the α-Gpdh alleles. This phenomenon may contribute to explain the maintenance of theAdh and α-Gpdh polymorphism and of theIn(2L)t inversion.

    Notes: Summary The effect of i.v. infusion of glucose on the hepatic handling of bilirubin was examined in rabbits. A significant increase in the excretion of conjugated bilirubin into the bile was observed, accompanied by a decrease in bilirubinemia. Hepatic bilirubin concentrations were lowered and the UDP-glucose concentrations and liver UDP-glucuronosyl and UDP-glucosyl transferase activities increased.

    Notes: Summary Fasting (48 h) results in dopamine-beta-hydroxylase (DBH) release both in adrenal gland and spleen, suggestive of an increase in the activity of these organs. Cold exposure (48 h) produces a dissociation of the, sympathoadrenal response. When both stimuli are simultaneously employed, the DBH response suggests the preponderance of the response to fasting. Plasma DBH is decreased in all groups studied, this could be due to its half-life and the splenic DBH depletion.

    Notes: Abstract Ge diffusion into GaAs from thin evaporated layers as sources is reported. Irradiation with aQ-switched ruby laser gives rise ton-type diffused layers of a thickness from 240 to 710 Å. A strong compensation of the diffused layers, that cannot be removed by thermal annealing, was observed. From the present experimental results it can be inferred that the diffusion coefficient increases at the melting point by 5 to 6 orders of magnitude.
