    Notes: . To assess the potential role of G-proteins in chemokinesis, Paramecium tetraurelia was pre-incubated with the G-protein modulator pertussis toxin. Pertussis toxin pretreatment significantly reduced Paramecium chemoattraction to sodium acetate and ammonium chloride in T-maze behavioral assays and depressed the frequency of avoidance reactions, indicating that heterotrimeric G-proteins may be involved with the motility response. To determine whether G-proteins exert their effect via the ciliary voltage-sensitive calcium channel, we examined responses of P. tetraurelia to the potent voltage-sensitive calcium channel agonist, deltamethrin. Pertussis toxin preincubation significantly reduced the toxic effects of deltamethrin exposure as determined by survival under depolarizing conditions and reduced the duration of backward swimming episodes in behavioral bioassays. Furthermore, non-hydrolyzable analogs of guanine nucleotides altered deltamethrin-stimulated calcium influx via calcium channels in isolated ciliary vesicles. Heterotrimeric G-protein subunits were subsequently detected in ciliary vesicles of P. tetraurelia by antibodies produced against Ga and Gβ subunits, and by 32P-ADP-ribosylation, indicating that proteins of the appropriate molecular weight are the target of pertussis toxin in these vesicles. These findings provide additional evidence that heterotrimeric G-proteins are associated with ciliary vesicles and that they play a role in the modulation of swimming behavior and the toxic action of deltamethrin in Paramecium.

    Notes: Peronospora viciae sporulated frequently following localized inoculation of leaflets of Pisum sativum cv. Krupp Pelushka but rarely on cv. Early Onward (60 and 〈 1% of plants respectively). Similar results were obtained following inoculation of seedling shoot apices. The proportions of sporangia that germinated, formed appressoria, invaded via stomata or cuticle, and established colonies, were similar on leaflets of both cultivars. After 2 days, total hyphal lengths of most colonies were greater than 100 μm (94% in Krupp Pelushka, 64% in Early Onward), but in Krupp Pelushka there were five times as many haustoria per unit length of hypha. Subsequently, only occasional colonies in Early Onward grew to the same extent as in Krupp Pelushka, with 8% and 88% respectively attaining lengths 〉 800 μm after 4 days. Reduced growth of hyphae in Early Onward was associated with increased frequency of host cells stained by trypan blue.

    Notes: The extent of lack of flatness of the film in air survey cameras was examined by the use of interferometry. Following an assessment of the merits of vacuum back and register glass flattening devices, a roller flattener was developed for use in conjunction with a register glass and with discrete vacuum application. Encouraging results have been obtained in the early stages of its use.

    Notes: : Artificial streams can assist in assessing the potential impact of pollutants on the structure and function of aquatic communities. While most artificial streams are constructed within a controlled environment or are maintained by recirculating water, we constructed a series of artificial streams along a river bank using river water on a once through basis. This system was used to develop and test methods for structural and functional analyses of Aufwuchs communities. Only the flow rate and stream depth were controlled while individual streams were perturbed to obtain communities of altered ecological condition. This experimental system provided a relatively inexpensive series of treated and untreated streams which allowed an evaluation and comparison of methods using communities under various stressed and nonstressed conditions. Without the stabilizing effects of a controlled atmosphere or recirculating water, our approach demonstrated a degree of variability which approached that of the river. We inferred that the Aufwuchs community present in the artificial streams maintained a high degree of structural complexity and functional dynamism, providing a strong test of our methods and an opportunity to examine current ecological theory.

    Notes: The sweetness of lactulose over the range of concentration 5–35% (W/V), measured by a trained panel using paired comparison with standard reference solutions of sucrose of varying concentrations, is 48% to 62% of that of sucrose. In addition, sensitivity thresholds and recognition thresholds for sweetness of lactulose and sucrose were determined by a rating-scale method. The sweetness of a mix ture containing 10% (W/V) lactulose and 5% (W/V) sucrose showed a synergistic effect of 22%, and a mixture of 5% (W/V) lactulose and 2.5% (W/V) sucrose showed 12% synergism. Partial hydrolysis of lactulose to give a mixture containing 5% (W/V) lactulose, 2.5% (W/V) galactose, and 2.5% (W/V) fructose caused a 6% synergistic effect on sweetness.
