    Notes: Abstract We have developed a vector strategy that allows transfer of plasmid DNA by conjugation from Escherichia coli to various Gram-positive bacteria in which transformation via natural competence has not been demonstrated. The prototype vector constructed, pAT187, contains the origins of replication of pBR322 and of the broad host range streptococcal plasmid pAMβ1, a kanamycin resistance gene known to be expressed in both Gram-negative and Gram-positive bacteria, and the origin of transfer of the IncP plasmid RK2. This shuttle plasmid can be mobilised efficiently by the self-transferable IncP plasmid pRK212.1 co-resident in the E. coli donors, and was successfully transferred by filter matings at frequencies of 2 × 10−8 to 5 × 10−7 to Enterococcus faecalis, Streptococcus lactis, Streptococcus agalactiae, Bacillus thuringiensis, Listeria monocytogenes and Staphylococcus aureus.

    Notes: The antigenic specificities of H-2-restricted, tumour-specific cytolytic T lymphocytes (CTL) were studied at the molecular level using CTL from BALB.B and C57BL/6 (H-2b) mice sensitized to an H-2b Gross murine leukaemia virus (MuLV)-induced tumour. Target cells were produced by the double trans-fection of mouse L cells (H-29 with the cloned H-2Kb or H-2P gene and retro-viral DNA derived from a molecular clone of Akv MuLV (closely related to Gross MuLV). Doubly transfected L cells which express either H-2Kb or H-2Db antigen and retroviral antigens are lysed in a virus-specific manner by Gross MuLV-immune CTL. The existence of two independent Gross MuLV-immune CTL subpopulations, one restricted by H-2Kb and the other by H-2Db, is thus confirmed. Gross MuLV-immune CTL from both BALB.B and C57BL/6 mice killed L cells that express Akv MuLV gag gene products and H-2Kb or H-2Db antigen. In contrast, only CTL from C57BL/6 mice killed L cells that express Akv MuLV env gene products and H-2Kb or H-2Db. This indicates that specific recognition of MuLV-induced antigens by CTL can be selective and varies according to the origin of the CTL.

    Notes: Faceted garnets from a wide range of geological ages, environments and locations have been studied in polished grain mounts by a combination of backscattered electron microscopy and elemental mapping using energy-dispersive X-ray analysis. In all cases, the areas apparently showing positive relief on the faceted garnet surfaces are compositionally identical to the adjacent grain cores despite a wide variation in detrital garnet compositions. In one case, zoning within the grain core can be traced into the faceted areas on the grain surface. Thus, faceted areas must be considered to form part of the original detrital grains. Together with previously published studies on experimental garnet etching, thermodynamic conditions for garnet growth, textural relationships between faceted garnets and authigenic and detrital phases, and distribution of faceted garnets in the subsurface, this paper provides conclusive proof that faceted garnet surfaces form as a result of dissolution, not overgrowth.

    Notes: This work was conducted in an attempt to elucidate some of the effects of nutrition on the incidence of witchweed (Striga asiatica (L.) Kuntze) in maize (Zeu mays L.).In an N, P, K field trial N applications resulted in up to 93% reduction in the incidence of witchweed. Maize height was unaffected by N and decreases in the incidence of Striga could not be attributed to the effects of host-parasite competition. Although plant height was 10% lower on zero-P plots, P did not have a significant effect on Striga and the beneficial effects of N were equally evident on both high and low-P plots.Potassium applications had the opposite effect to N. While the main effect of K was not quite significant at the 5% level of probability, an N × K interaction was evident, and in the absence of applied N, K applications led to a more than 4-fold increase in the incidence of Striga. As with N, K did not influence plant height and it seems unlikely that competition effects were responsible.

    Notes: The objectives of the study were to estimate the genetic components of variance, predict the response to selection and to evaluate observed response to selection using two populations namely, PSB 7 and PSB 3, of pearl millet (Pennisetum americanum [L.] Leeks). The observed response was studied using four methods of selection based on single and multitrait criteria in three types of families, namely S1, half-sib (HS) and full-sib (FS) families. One hundred SI, HS and FS families each were developed in PSB 7 and PSB 3. Plants within a population were pollinated with bulk pollen of other population to develop HS families. Crosses were made between a pair of plants using one plant from each of the two populations to develop PS families. Data were recorded on days to flower, plant height, tiller number, spike length, spike girth, seed yield per plant and spike weight. Additive genetic variance estimates were significant for all traits. The estimates of dominance and additive × location and dominance × location interaction components were not significant, except additive × location component in PSB 7 for seed yield and spike weight. The genotypic component of variance among S1 families and S1 × location interactions were significant except S1 × location interactions in PSB 7 for days to flower, spike length and spike girth.Twenty-four strains were developed by intermating superior families identified on the basis of four selection methods in each of the six types of families.The selection methods were high seed yield (YS) per se; index selection (IS) for high tiller number and long and thick spike; truncation selection (TS) for high yield among the families having days to flower and plant height lower than the population mean: and visual selection (VS) after completion of flowering for good vigour, uniformity in plant height and good inflorescence attributes. YS and IS were found to be superior to TS and VS with respect to observed genetic gains for seed yield. That advance was higher in selection among SI families as compared with FS and HS families. In general, a good agreement was noted between observed and expected responses. Expected genetic gain from FS reciprocal (R) recurrent selection was higher than the average gain from HS-R selection.

    Notes: South of the Main Mantle Thrust in north Pakistan, rocks of the northern edge of the Indian plate were deformed and metamorphosed during the main southward thrusting phase of the Himalayan orogeny. In the Hazara region, between the Indus and Kaghan Valleys, metamorphic grade increases northwards from chlorite zone to sillimanite zone rocks in a typically Barrovian sequence. Metamorphism was largely synchronous with early phases of the deformation. The metamorphic rocks were subsequently imbricated by late north-dipping thrusts, each with higher grade rocks in the hanging wall than in the footwall, such that the metamorphic profile shows an overall tectonic inversion. The rocks of the Hazara region form one of a number of internally imbricated metamorphic blocks stacked, after the metamorphic peak, on top of each other during the late thrusting. This imbrication and stacking represents an early period of post-Himalayan uplift.

    Notes: Abstract Sclerotinia sclerotiorum isolates B24 (virulent) and SS41 (hypovirulent) possess oxalate decarboxylase. Production was regulated by composition and pH of culture medium and required the presence of oxalate or its precursor, succinic acid, as inducers. Mycelia of both isolates contain equivalent amounts of enzyme.
