    Notizen: This paper presents a model designed to help determine the number of clerks or servers in a retail queuing situation. The model demonstrates a realistic treatment of the effect of waiting time on customer demand and revenues by treating the average waiting time as a form of price. Thus, the demand rate and service level are interrelated. The number of servers is obtained by balancing the cost of additional servers against the revenues and profits arising from greater demand. An example is shown, using data obtained from a supermarket, to demonstrate the insight gained from applying the model. Some extensions and research possibilities are considered, including a treatment of ancillary activities. An appendix examines a continuous version of the model which provides additional insight into the character of the model.

    Notizen: Since Skinner's [40] landmark article depicting the manufacturing function as the “missing link” in corporate strategic processes, a portion of the blame for inferior performance in many firms has been attributed to the subordinate strategic position of manufacturing. It has been argued that part of the solution to misalignments between the capabilities possessed by manufacturing and the requirements dictated by customers is for manufacturing to take a more proactive stance. However, little research has been reported which examines manufacturing proactiveness empirically. In this paper, we address this gap by developing an operational definition of manufacturing proactiveness and testing empirically whether a link exists between proactiveness and performance based on data collected from a sample of manufacturers.Based on the manufacturing strategy literature, we identify two major dimensions of manufacturing proactiveness: (1) the degree of manufacturing's involvement in the strategic processes of the business unit; and (2) the degree of commitment to a long-term program of investments in manufacturing structure and infrastructure aimed at building capabilities in anticipation of their need. We develop reliable scales for measuring each of the dimensions of proactiveness and use the data to provide evidence of a clear link between manufacturing proactiveness and business performance. We show that investments in structural programs coupled with either high levels of manufacturing involvement in strategic processes or planned investments in infrastructural programs correlate with higher than average performance.

    Notizen: The Incl1 conjugative plasmid Collb-P9 carries a psiB gene that prevents induction of the SOS response in host bacteria. This locus is located 2.5 kb downstream of the ssb (single-stranded DNA-binding protein) gene in the leading region. This portion of Collb is strikingly similar to part of the leading region of the otherwise distinct F plasmid. Expression of psiB and ssb is increased when the host cell is exposed to an SOS-inducing treatment or the Collb transfer system is derepressed. Moreover, expression of both genes on a derepressed plasmid is strongly enhanced in conjugatively infected recipient cells. Carriage of the psiB gene by Collb is shown to prevent a low level of SOS induction following conjugation. Plasmid ssb and psiB genes may function to promote installation of the replicon in the new cell.

    Notizen: The β amyloid protein found in extracellular deposits in Alzheimer's disease (AD) is heterogeneous at its C-terminus; proteins ending at residues 40, 42, and 43 have been identified in neuritic deposits, while protein in vascular amyloid appears to end at residue 39 or 40. Studies of synthetic β proteins (β1–39, β1–40, β1–42), and model peptides (β26–39, β26–40, β26–42, β26–43) demonstrate that amyloid formation is a nucleation-dependent phenomenon. Peptides ending at residues 39 or 40 were kinetically soluble for hours to days, while peptides ending at residues 42 or 43 aggregated immediately; all eventually reached similar thermodynamic solubility. The kinetically soluble variants could be seeded with the kinetically insoluble variants. The secondary structure of β26–39 fibrils was different from that of β26–42 fibrils, however, seeding β26–39 with β26–42 produces mixed fibrils with structure similar to β26–42. These results suggest that neuritic plaques may be seeded by their minor component; this may determine the structure and properties of amyloid in AD.

    Notizen: Abstract A genomic DNA library was constructed from a pathogenic strain of Aspergillus fumigatus using the cosmid vector pCosAX. Cosmid clones with homologies to the rodA, brlA, fluG, flbA or trpC genes from A. nidulans were isolated from the library. Each A. fumigatus clone was used to complement strain of A. nidulans with a mutation in the homologous gene. A spontaneous white spored strain of A. fumigatus was isolated. The mutation was complemented by transforming the strain with a plasmid containing the wA gene from A. nidulans. The results from these experiments indicate a significant degree of structural and functional homology between genes from the organisms. These findings indicate the potential to exploit the methods and information available from A. nidulans to address questions related to human disease caused by A. fumigatus and the ability to use A. nidulans as a surrogate genetic host for characterizing A. fumigatus gene function.
