    Notes: The coding sequence of the cyanogenic α-hydroxynitrile lyase gene of Manihot esculenta Crantz (cassava) was cloned in the plasmid vector pMal-c2 and expressed in Escherichia coli strain JM105. DNA sequencing showed that the recombinant plasmid contained the same sequence as the cDNA clone pHNL10. Peptide sequencing of the recombinant protein showed that the N-terminus was heterogeneous, with either four or six additional amino acid residues compared with the native protein. Circular dichroism spectra indicated similar secondary structure contents for both proteins. Enzyme assays showed that specific activity of native and recombinant proteins were 0.24 and 0.26 mmol CN-/mg/min, respectively; that both proteins had optimal activity at 40°C and pH 5.5; and that both proteins were inhibited by the serine protease inhibitor phenyl-methane sulfonyl flouride (PMSF). Isoelectric focusing of native and recombinant protein revealed multiple isoforms for both proteins; the recombinant protein had a more basic mean isoelectric point (pl) (5.1) than the native protein (4.5). © 1997 John Wiley & Sons, Inc.     Description: The climate signal imprinted in the snow isotopic composition allows to infer past climate variability from ice core stable water isotope records. The concurrent evolution of vapor and surface snow isotopic composition between precipitation events indicates that post-depositional atmosphere-snow humidity exchange influences the snow and hence the ice core isotope signal. To date, however, this is not accounted for in paeleoclimate reconstructions from isotope records. Here we show that vapor-snow exchange explains 36% of the summertime day-to-day δ18O variability of the surface snow between precipitation events, and 53% of the δD variability. Through observations from the Greenland Ice Sheet and accompanying modeling we demonstrate that vapor-snow exchange introduces a warm bias on the summertime snow isotope value relevant for ice core records. In case of long-term variability in atmosphere-snow exchange the relevance for the ice core signal is also variable and thus paleoclimate reconstructions from isotope records should be revisited.

    Notes: Bench scale packed columns were used to study the kinetic behavior of biological denitrification. After tracer studies were run to determine flow characteristics, steady state nitrate profiles were determined. A first-order rate expression with an Arrhenius temperature dependence is an adequate model of the reaction for nitrate-nitrogen concentrations of 〈 100 mg/l and temperature of 5° to 20°C. The rate constants are proportional to the packing surface area.

    Notes: Infeasible path optimization algorithms have been effective with equation-solving or simultaneous modular process simulators. Using these simulators, successive quadratic programming is applied to large nonlinear programs. However, the algorithms are not directly applicable to commercial simulators of the sequential modular type.Here an infeasible path algorithm is presented which can easily be integrated with most commercial process simulators. The optimization problem remains small (10 or 20 variables) and the user merely replaces the convergence block with an “optimization” block.A simple process example demonstrates the operation, effectiveness, and potential of this algorithm.
