    Notes: Abstract A tuber disc transformation and regeneration system was devised for potato (Solanum tuberosum). Tuber discs were found to be the most morphogenetic organ on a medium previously optimised for tomato regeneration. Shoot regeneration from tuber discs was rapid and transformed as shown by nopaline assays and Southern blot analysis. The ease and speed of the tuber disc method will allow for the increased use of this commercially important plant in transformation studies.

    Notes: Summary Derivatives of potato (Solanum tuberosum cv.'s ‘Maris Bard’ and ‘Desiree’) transformed with disarmed T-DNA from genetically engineered Agrobacterium tumefaciens strains were isolated. The transformed plants were recovered from shoot-forming tumours induced by infection of wounds with mixedcultures of shoot-inducing A. tumefaciens strains T37 and either Agrobacterium strain LBA1834(pRAL1834), (Hille et al. 1983) or LBA4404(pBIN6; pRAL4404), (Bevan 1984). Two small-scale feasibility experiments gave at least four ‘Maris Bard’ plants transformed with pRAL1834 T-DNA and two ‘Desiree’ plants with pBIN6 T-DNA. The transformed ‘Maris Bard’ plants were morphologically abnormal and highly aneuploid. This was probably an unfortunate side-effect of a tissue culture-step introduced to promote the efficiency of shoot regeneration. The transformed ‘Desiree’ plants, in contrast, were isolated without promoting additional shoot-growth. They were morphologically normal, contained 47 and the euploid 48 chromosomes per cell respectively and had improved growth on media containing kanamycin.

    Notes: Abstract. The expression of many genes encoding transcriptional activators in prokaryotes and eukaryotes is upregulated through positive feedback activation. During positive feedback activation, a transcriptional activator binds to its own promoter and thus increases its own expression as well as the expression of its target genes. In the simplest case, increased levels of the transcriptional activator can be directly correlated with increased expression of its target genes. In this study, we present a gene expression system, designated feedforward loop (FFL) system, which makes use of this kind of positive feedback regulation for the expression of plant transgenes. We show in transient and stable transformation experiments that such a system is functional and that it can be used to obtain high level gene expression in plants. We also provide evidence that the transgene is ubiquitously expressed when using the FFL system. Finally, we discuss the possibilities of using FFL gene expression systems for applications in plant molecular genetics and biotechnology.

    Notes: Abstract Polygalacturonase (PG) is the major cell wall degrading enzyme of tomato fruit. It is developmentally regulated and is synthesised de novo in ripening fruit. Genomic clones encoding a PG gene of tomato (Lycopersicon esculentum Mill cv. Ailsa Craig) have been isolated, mapped and sequenced. The sequence of the protein-coding region is identical to that of a PG cDNA [20]. Comparison of the cloned restriction fragments with genomic Southern data suggests that there may only be one gene for PG per haploid genome. The PG gene, which covers approximately 7 kb, is interrupted by 8 intervening sequences ranging in size from 99 bp to 953 bp. The transcription start point was identified by S1 mapping and primer extension analysis. About 1.4 kb of 5′ flanking DNA has been sequenced. This contains putative TATA and CAAT boxes and also direct repeat sequences. A transcriptional fusion has been constructed between the putative 1.4 kb promoter fragment and the chloramphenicol acetyl transferase (CAT) gene. Constructs containing this gene have been transferred to tomato using binary vectors. Regenerated transgenic plants express CAT in ripe tomato fruit, but not in unripe tomatoes, leaves, or roots.

    Description: Colloidal matter exhibits unique collective behaviors beyond what occurs at single-nanoparticle and atomic scales. Treating colloidal particles as building blocks, researchers are exploiting new strategies to rationally organize colloidal particles into complex structures for new functions and devices. Despite tremendous progress in directed assembly and self-assembly, a truly versatile assembly technique without specific functionalization of the colloidal particles remains elusive. We develop a new strategy to assemble colloidal matter under a light-controlled temperature field, which can solve challenges in the existing assembly techniques. By adding an anionic surfactant (that is, cetyltrimethylammonium chloride), which serves as a surface charge source, a macro ion, and a micellar depletant, we generate a light-controlled thermoelectric field to manipulate colloidal atoms and a depletion attraction force to assemble the colloidal atoms into two-dimensional (2D) colloidal matter. The general applicability of this opto-thermophoretic assembly (OTA) strategy allows us to build colloidal matter of diverse colloidal sizes (from subwavelength scale to micrometer scale) and materials (polymeric, dielectric, and metallic colloids) with versatile configurations and tunable bonding strengths and lengths. We further demonstrate that the incorporation of the thermoelectric field into the optical radiation force can achieve 3D reconfiguration of the colloidal matter. The OTA strategy releases the rigorous design rules required in the existing assembly techniques and enriches the structural complexity in colloidal matter, which will open a new window of opportunities for basic research on matter organization, advanced material design, and applications.
