    Description: Despite the progress made in DNA sequencing over the last decade, reconstructing telomere-to-telomere genome assemblies of large and repeat-rich eukaryotic genomes is still difficult. More accurate basecalls or longer reads could address this issue, but no current sequencing platform can provide both simultaneously. Perennial ryegrass (Lolium perenne L.) is an example of an important species for which the lack of a reference genome assembly hindered a swift adoption of genomics-based methods into breeding programs. To fill this gap, we optimized the Oxford Nanopore Technologies’ sequencing protocol, obtaining sequencing reads with an N50 of 62 kb—a very high value for a plant sample. The assembly of such reads produced a highly complete (2.3 of 2.7 Gb), correct (QV 45), and contiguous (contig N50 and N90 11.74 and 3.34 Mb, respectively) genome assembly. We show how read length was key in determining the assembly contiguity. Sequence annotation revealed the dominance of transposable elements and repeated sequences (81.6% of the assembly) and identified 38,868 protein coding genes. Almost 90% of the bases could be anchored to seven pseudomolecules, providing the first high-quality haploid reference assembly for perennial ryegrass. This protocol will enable producing longer Oxford Nanopore Technology reads for more plant samples and ushering forage grasses into modern genomics-assisted breeding programs.

    Description: Past changes in ocean 14C disequilibria have been suggested to reflect the Southern Ocean control on global exogenic carbon cycling. Yet, the volumetric extent of the glacial carbon pool and the deglacial mechanisms contributing to release remineralized carbon, particularly from regions with enhanced mixing today, remain insufficiently constrained. Here, we reconstruct the deglacial ventilation history of the South Indian upwelling hotspot near Kerguelen Island, using high-resolution 14C-dating of smaller-than-conventional foraminiferal samples and multi-proxy deep-ocean oxygen estimates. We find marked regional differences in Southern Ocean overturning with distinct South Indian fingerprints on (early de-)glacial atmospheric CO2 change. The dissipation of this heterogeneity commenced 14.6 kyr ago, signaling the onset of modern-like, strong South Indian Ocean upwelling, likely promoted by rejuvenated Atlantic overturning. Our findings highlight the South Indian Ocean’s capacity to influence atmospheric CO2 levels and amplify the impacts of inter-hemispheric climate variability on global carbon cycling within centuries and millennia.

    Description: Bulk sediment δ15N records from the eastern tropical Pacific (ETP) extending back to the last ice age most often show low glacial δ15N, then a deglacial δ15N maximum, followed by a gradual decline to a late Holocene δ15N that is typically higher than that of the Last Glacial Maximum (LGM). The lower δ15N of the LGM has been interpreted to reflect an ice age reduction in water column denitrification. We report foraminifera shell‐bound nitrogen isotope (FB‐δ15N) measurements for the two species Neogloboquadrina dutertrei and Neogloboquadrina incompta over the last 35 ka in two sediment cores from the eastern equatorial Pacific (EEP), both of which have the typical LGM‐to‐Holocene increase in bulk sediment δ15N. FB‐δ15N contrasts with bulk sediment δ15N by not indicating a lower δ15N during the LGM. Instead, the FB‐δ15N records are dominated by a deglacial δ15N maximum, with comparable LGM and Holocene values. The lower LGM δ15N of the bulk sediment records may be an artifact, possibly related to greater exogenous N inputs and/or weaker sedimentary diagenesis during the LGM. The new data raise the possibility that the previously inferred glacial reduction in ETP water column denitrification was incorrect. A review of reconstructed ice age conditions and geochemical box model output provides mechanistic support for this possibility. However, equatorial ocean circulation and nitrate‐rich surface water overlying both core sites allow for other possible interpretations, calling for replication at non‐equatorial ETP sites.
    Description: Plain Language Summary: The 15N/14N ratio of sediments provides information on the past marine nitrogen (N) cycle through the production of N‐bearing organic matter in the surface ocean and its burial in the sediments. Previous measurements of the sedimentary 15N/14N ratio in the eastern equatorial Pacific (EEP) indicate lower values during the last ice age compared to the Holocene (the current warm period). This has been interpreted to reflect an ice age reduction in the oceanic N loss process known as “denitrification” that occurs between 200 and 500 m depth in this region of the ocean. However, the 15N/14N ratio measured on the whole sediment can be biased by biological and chemical processes in the sediments and by foreign N inputs. To avoid these complications, we measured the 15N/14N ratio of organic N embedded in the calcite shell of unicellular zooplankton (foraminifera) in two sediment cores from the EEP. We found similar foraminifera‐bound 15N/14N ratios during the last ice and the Holocene. This may argue against the long‐held interpretation of a reduction in denitrification during the last ice age. However, the oceanographic setting of these equatorial cores leaves open alternative interpretations, calling for further work at other eastern tropical Pacific sites.

    Notes: Zusammenfassung Die Chemie der Polymyxine, einer Klasse von basischen Polypeptid-Antibiotika, begann 1954, als durch Gegenstromverteilung erstmals ein definierter Vertreter, das Polymyxin B1 in reiner Form isoliert werden konnte (L. C.Craig). Partialhydrolyse mit Mineralsäuren führte zum Schluss, dass es sich um Cyclohepta- oder Cyclooctapeptide mit Seitenketten handelt, dieα, γ-Diaminobuttersäure (Dab) enthalten (W.Hausmann). Amidartige Verknüpfung der Seitenkette mit einer Fettsäure [(+)-6-Methyloctansäure (MOA) oder 6-Methylheptansäure (IOA)] (S.Wilkinson) verleiht diesen Antibiotika den Charakter von Invertseifen. Sie sind besonders gegen gramnegative Erreger wirksam. Schwierigkeiten bereiteten die Aufklärung der Verknüpfungsweise der Seitenkette (α- oderγ-) und die Beantwortung der Frage, ob das Molekül nebend-Phenylalanin in der Ringsequenz noch einend-α, γ-Diaminobuttersäurerest in Nachbarschaft zur Fettsäure in der Seitenkette enthält. Synthetische Versuche mitd-α, γ-Diaminobuttersäure an dieser Stelle führten zu hochaktiven Produkten, die aber mit natürlichem Polymyxin B1 nicht identisch waren. Entscheidende Fortschritte wurden mit dem bakteriellen Enzym Nagarse (T.Suzuki) erzielt, das schrittweise die Seitenkette bis zum Ringpeptid abbaut. Dabei ergab sich, dass den Polymyxinen die allgemeine Struktur eines Cycloheptapeptides mitα-verknüpfter Seitenkette zukommt (Figur 4). Die Polymyxine B1 E1 (Colistin A) sowie Circulin A unterscheiden sich voneinander nur durch eine Variation in der gleichen Dipeptidsequenz des 7-gliedrigen Ringes. Die im Polymyxin B1 vorhandene Dipeptidsequenzd-Phe-l-Leu ist in Polymyxin E1 (Colistin A) durchd-Leu-l-Leu und in Circulin A durchd-Leu-Ll-Ile ersetzt. Im Polymyxin D1 ist neben dem Ersatz der entsprechenden Sequenz durchl-Leu-l-Thr noch einα, γ-Diaminobuttersäurerest der Seitenkette durch eind-Serin ausgetauscht. Die entsprechenden Verbindungen mit dem Index 2 unterscheiden sich von denjenigen mit dem Index 1 durch einen Austausch der (+)-6-Methyloctansäure durch 6-Methylheptansäure. Die Struktur von Polymyxin B1 E1 und Circulin A konnte durch Totalsynthese gesichert werden (K.Vogler). Weitere Fortschritte in der Erforschung der Natur der noch unbekannten Vertreter sind in Kürze zu erwarten.
