    Notes: Summary Two experiments were conducted to establish the effect of dieldrin on survival of chickens during severe dietary restriction. White Leghorn hens were fed rations containing 0, 1, 5, 10, and 20 ppm of dieldrin for 12 weeks. At this time, dieldrin had accumulated in the body fat in direct proportion to the amount fed. During subsequent feed restriction, survival of hens fed 10 or 20 ppm dieldrin was significantly shorter than that of hens fed 0, 1, or 5 ppm dieldrin. Dieldrin concentration in blood declined during the first and second weeks following feed removal, then it increased. Blood sampling was terminated at the end of 4 weeks because nearly all hens had died, but dieldrin concentration in the blood had not increased appreciably over the concentration observed in blood at the time that feed was removed.

    Notes: Summary Rapid auxin-induced cell expansion in artichoke tuber slices is obtained by aerating the slices in water (“aging”) prior to auxin treatment. 5-fluorouracil (5-FU), an inhibitor of ribosomal RNA synthesis in plant cells, markedly inhibits auxin-induced growth only if present in the pre-growth aging period. Autoradiographic studies show that 5-FU given in the aging and/or growth periods reduces the incorporation of RNA precursors into the cytoplasm. Pulse-chase experiments suggest that the reduced cytoplasmic incorporation is in large part due to decreased stability of ribosomal rNA, as nucleolar and chromatin label are only slightly depressed at the end of the pulse. Though the nucleoli continue to incorporate RNA precursors following 5-FU treatment, they lack a distinct granular zone, and appear as homogeneous fibrillar structures under the electron microscope. 5-FU has a parallel inhibitory effect on growth and protein synthesis as shown by 3H-leucine studies during the growth period. Electron-microscope studies show that treatment with 5-FU causes decreased numbers of ribosomes and rough endoplasmic reticulum. The results suggest that the ribosomes and rough endoplasmic reticulum formed during aging are important in obtaining subsequent rapid auxin-induced expansion. The new ribosomes serve in part to replace pre-existing ribosomes present at the time of excision, which from electron microscopic evidence from 5-FU treated tissue, appear to slowly disappear.

    Notes: Summary In the dormant embryonic wheat coleoptile the nucleolus is almost a purely fibrillar structure and numerous large blocks of heterochromatin are present in the nucleus. Following germination the nucleoli develop distinct granular and fibrillar zones which become intricately intermingled, and the nuclear heterochromatin disperses. What appear to be perichromatin granules have been noted in coleoptile nuclei. When these dark grown coleoptiles cease division growth the nucleoli once again assume a largely fibrillar structure, and show low incorporation of3H-uridine. Associated with these latter changes at the cessation of meristematic growth there is a dilution of cytoplasmic ribosomes. The cytoplasm up until this time is densely packed with ribosomes. This study, taken together with other reports, suggests that characteristic nucleolar structural changes and reduced nucleolar activity are often associated with the differentiation of plant cells, following the cessation of meristematic growth.

    Notes: Scanning electron micrographs were made of the surfaces of silicone rubber finger joint prostheses which had been subjected to 107 bending cycles at 37°C in bovine serum. Preferential attack at the regions of highest stress and the formation of microcracks of length 10μ and less were observed. By way of analogy with the corrosion fatigue phenomenon in metallic alloys, it is suggested that these may be initiation sites for the mechanical failures which have been observed, and that the acceleration of lipid absorption by tensile stress provides potential mechanisms for both initiation and accelerated propagation of fatigue cracks. It is demonstrated as a consequence of mechanochemical thermodynamics that any constituent of the body fluids (e.g. lipids) which can swell the elastomer will in fact be preferentially absorbed at the point of highest tensile stress, i.e., the crack tip, and that swelling will be accelerated by the applications of tensile stress.

    Notes: Heat vulcanized silicone elastomer (Silastic-Dow) has been in clinical use as a prosthetic replacement material for over a decade. When subjected to operating stress in body fluids, as in cardiac poppet-valves, failure has occurred. Fractures of silastic finger-joint prosthesis have also been experienced.Chemical and fractographic analyses of such failed finger-joint prostheses raise the suspiciaon of failure. Severe discoloration of the implant at the point of maximal stress concentration, the presence of significant amounts of triglyceride and cholesterol on the fractured prostheses, and the pattern of fracture markings indicate that fracture was due to a combination of chemical surface deterioration and stress concentration.Study of the method of surgical insertion of these prostheses suggests failure to completely excise the volar plate and other palmar joint structures could create a step off and sufficiently increase the stress in the bending element of the device to cause fracture. In vitro cyclic testing at 37°C in serum failed to cause fracture of these devices after 10 million cycles, but did produce gradual discoloration of the entire prosthesis which began and was most noticeable at the point of bending. Although the prostheses adsorb lipid, which may produce surface deterioration, it is anticipated that properly inserted, the service life of these devices should be more than adequate for full clinical application.
