    Notes: Abstract The uptake of 2-ketogluconate is inducible in Pseudomonas putida: 2-ketogluconate, glucose, gluconate, glycerol and glycerate were each good nutritional inducers of this ability. 2-Ketogluconate uptake obeyed saturation kinetics (apparent K min 2-ketogluconate-grown cells was 0.4 mM). 2-Ketogluconate was transported against a concentration gradient, apparently in an unchanged state, and the process required metabolic energy, all of which indicate an active transport system. A number of independently isolated mutants with deranged activity of a common glucose-gluconate uptake system were found to be also defective in 2-ketogluconate transport. Strains unable to transport 2-ketogluconate which grew readily on glucose and gluconate were also isolated. These results suggest that 2-ketogluconate transport is governed by at least two genetic elements: one which is also required to take up glucose and gluconate and another which appears to be specific for 2-ketogluconate transport. Similarly glucose and gluconate transport appears to require at least one factor which is not necessary for 2-ketogluconate transport, as suggested by the lack of induction of the common glucose-gluconate uptake system by glycerol and glycerate, substrates which are good inducers of 2-ketogluconate uptake.

    Notes: Abstract Fructose transport was not apparently affected in a number of Pseudomonas putida strains with deranged activity of a common glucose-gluconate uptake system, indicating the existence of an independent fructose uptake system. Fructose uptake by glucose-gluconate uptake mutants was induced by fructose and obeyed saturation kinetics (apparent K m =0.3 mM). The fructose uptake system serves to transport glucose in addition to fructose. The entry of fructose into P. putida cells appears to be mediated also by the glucose-gluconate uptake system, as shown by the ability to accumulate fructose of wild type cells grown on glucose, a substrate that induces the glucose-gluconate uptake system but not the fructose uptake system. In addition, fructose was found to be an inducer of the glucose-gluconate uptake system. The physiological significance of these observations is not clear because the fructose uptake system can provide the cell with a high enough internal concentration of fructose to support maximum growth rate on this hexose, as shown by following the growth course of glucose-gluconate uptake mutants on fructose.

    Notes: The anthocyanin pigments responsible for the violet color of the tropical tuber Dioscorea tryphida was investigated. The pigments were extracted with 0.1% HCI in methanol, purified with Dowex 50W × 4 cation exchange resin in the hydrogen form and further purified by paper chromatography. The identification of the pigments was based on Rf values in different solvents, partial acid hydrolysis, sugar moiety, alkaline degradation, fluorescence under ultraviolet radiation, color reactions and absorption spectra in the visisble and ultraviolet regions. The pigments were identified as follow: peonidin 3,5-diglucoside, malvidin 3,5-diglucoside and malvidin 3,5-diglucoside-ferulic acid.

    Notes: Abstract This paper presents a model of the spatiotemporal processes of simple ganglion cells of cat's retina. The model is based on the existence of two anatomical paths in the retina: a direct path, responsible for central excitatory effects, and a transversal path, responsible for peripherical inhibitory effects. Plausible spatial weighting functions and temporal transfer functions for photoreceptors, horizontal, bipolar and ganglion cell are introduced by prudent application of comparative neurophysiology. The response of the model to point light stimuli, moving light bars and sinusoidal gratings are obtained. They are in very good agreement with experimental data.

    Notes: Summary A segment of DNA located in the region of the E. coli K12 chromosome previously identified by the Rac phenotype can function as a self-replicating plasmid. Evidence is presented that this plasmid, the oriJ plasmid, contains the origin of replication of a defective prophage postulated to be located in this chromosomal region by Low (1973). The plasmid can only be maintained in strains in which this postulated prophage has been deleted. In strains which possess the prophage selection for plasmid maintenance permits the isolation of clones containing new deletions which we postulate are the result of prophage excision.

    Notes: Summary The linkage arrangements of genes governing glucolysis in Pseudomonas putida have been determined by transductional analysis. Five genes (gdh, kgtA, kgtB, edd and eda), comprising at least three operons, are cotransducible with each other, but not with ggu (glucose and gluconate uptake) nor with genes of a known supra-operonic cluster of genes specifying enzymes of other dissimilatory pathways, nor with a biochemically uncharacterized his marker. It thus appears that P. putida may have more than one chromosomal region in which genes with dissimilatory function are clustered in a supra-operonic fashion.
