    Notes: The formation of Babesia equi sporozoites in the salivary glands of three tick species (Hyalomma anatolicum anatolicum, H. a. excavatum, Rhipicephalus turanicus) was studied by electron microscopy. The development was identical in all three vectors. On the 8th day post repletionem kinetes of B. equi had invaded alveoli of the nymphal salivary glands and were transformed to sporonts bounded by a single membrane. The sporonts were polymorphous bodies each with a highly lobed nucleus and numerous mitochondria. These stages persisted during ecdysis of the tick nymph to the adult stage. After attachment of these newly molted adults to a new host the formation of sporozoites was completed within five days. The sporonts occupied most of the infected alveolus and were extensively divided into cytoplasmic portions of various size. On the 4th day after attachment of the tick, sporozoite-anlagen, into each of which a nucleus and a mitochondrion were incorporated, appeared at the periphery of the sporonts. An apical complex with a polar ring, rhoptries, and micronemes was formed at the tip of each protruding anlage. Finally thousands of pyriform sporozoites (3.0 × 1.2 μm) filled the hypertrophied alveolus. This development is similar to sporogony in the genus Theileria.

    Notes: Summary 1. We determined the tuning properties of 66 single chemoreceptor cells from the legs of the lobsterHomarus americanus. The specificity of these (taste) cells was tested electrophysiologically in the upper range of biologically relevant stimulus concentrations (3×10−4 mol/l) with 15 compounds listed in Table 1. 2. Most of the chemoreceptors surveyed were narrowly tuned to specific test chemicals (Fig. 3). The best stimuli for these chemoreceptors were glutamate (Glu), ammonium chloride (NH4), betaine (Bet) and hydroxyproline (OH-Pro) (Fig. 4). Cell populations were defined by their best stimulus. Populations of cells responding best to Glu and NH4 were extremely narrowly tuned; populations responding best to Bet and OH-Pro were less narrowly tuned (Fig. 5, Table 2). 3. Each cell population showed a range of response specificities, from cells responding to only one compound to cells responding more broadly to other compounds. Within each population cells were arranged by their tuning breadth (H). Cells that shared a best stimulus but responded also to other compounds showed no consistency for the next best stimulus (Fig. 6). This suggests that even cells responding to the same stimulus do not share the same receptor complement. 4. The response patterns to Glu, NH4 and Bet elicited across all the chemoreceptor cells surveyed can be distinguished statistically as different from each other and from all others tested. Similarly, OH-Pro can be distinguished from all other stimuli except glycine and taurine (Figs. 8 and 9). 5. Systematic prior exposure to all 15 test compounds (corresponding to 21 stimulus exposures) at our stimulus concentrations reduced the absolute sensitivity of Glu best cells but did not change the slope of their dose-response functions (Fig. 10) with respect to earlier results (Derby and Atema 1982a). 6. We suggest that extremely narrowly tuned cells in lobster legs may function to detect key compounds as feeding stimuli over a large dynamic range of mixture concentrations such as found in nature. These cells may not be affected as much as broadly tuned cells by ambient backgrounds of cross-adapting compounds. In addition, both narrowly tuned and broadly tuned cells may participate in across fiber patterns to encode the quality of naturally occurring stimulus mixtures.

    Notes: Summary When deplasticized Epon sections were treated with endo- and/or exopeptidases prior to incubation with antibodies, the neuropeptide immunoreactivity of secretory nerves was often altered in a predictable way. Cleavage of neurosecretory material in octopus nerves by trypsin and carboxypeptidase-B enhanced enkephalin-like immunoreactivity, while Molluscan neuropeptide-like immunoreactivity was prevented by tryptic cleavage. The enzyme effects indicated the occurrence of a heptapeptide (Tyr-Gly-Gly-Phe-Met/Leu-Arg-Phe) that contains both the enkephalin and the Molluscan neuropeptide sequence. Vasopressin terminals of the rat neurohypophysis, which presumably contain enkephalin precursor sequences, exhibited enkephalin-like immunostaining after tryptic cleavage. ACTH/β-endorphin cells of the rat intermediate pituitary, which synthesize the enkephalin sequence at the N-terminus of β-endorphin, exhibited enkephalin-like immunoreactivity when sections were treated with α-chymotrypsin or trypsin, but not after incubation with leucine-aminopeptidase or carboxypeptidase-B. Enkephalin-like immunostaining could not be induced in any way in ACTH/β-endorphin cells of the anterior pituitary. Enzymatic cleavage may give additional information in immunocytochemical localization studies on neuropeptide sequences in secretory nerves and hormonal granules.
