    Notes: Abstract Biodegradable microspheres containing a lipophilic muramyl dipeptide, MDP-B30, were prepared from a L-lactic acid–glycolic acid copolymer. The effect of precoating the microspheres with water-soluble polymers including proteins on the antitumor activity of mouse peritoneal macrophages (Mφ) was investigated. Macrophages activated by phagocytosis of the microspheres exhibited growth inhibitory activity toward Meth-A tumor cells. The activity correlated with the extent of Mφ phagocytosis of the microspheres. Mφ phagocytosis was greatly augmented by gelatin precoating of the microspheres, resulting in a significant increase of in vitro antitumor activity of Mφ by the microspheres. However, potentiation of Mφ activity by gelatin precoating was minimal after intraperitoneal injection of the microspheres, but cross-linking of the coated gelatin with glutaraldehyde afforded potentiation of the antitumor activity in vivo.

    Notes: Abstract In order to evaluate an indomethacin poly vinyl alcohol (PVA) hydrogel for rectal administration, the in vitro release characteristics of indomethacin from the hydrogel and indomethacin plasma concentrations after rectal administration were examined. The PVA hydrogel containing indomethacin was prepared by a low-temperature crystallization method. The release of indomethacin from the PVA hydrogel agreed with the Fickian diffusion model for 10 hr. Rectal administration of indomethacin hydrogels to rats yielded high indomethacin plasma concentrations, without producing a sharp peak, and a sustained-release effect. In dogs, the indomethacin hydrogel produced a similar sustained-release effect; however, the indomethacin plasma concentration was relatively low compared with that of an indomethacin suppository.

    Notes: Abstract Gelatin microspheres with a diameter less than 2 µm were synthesized by means of cross-linking with glutaraldehyde. When the microspheres were subjected to degradation in phosphate-buffered saline solution containing collagenase, the digestion of microspheres was found to decrease with increasing cross-linking. Interferon was incorporated in the microspheres at a high trapping efficiency, and the rate of interferon release from the microspheres was regulated by the extent of cross-linking with glutaraldehyde. Gelatin microspheres incorporating interferon-α were readily phagocytosed by macrophages, regardless of the extent of cross-linking, and the phagocytosed microspheres were observed to be degraded gradually in the interior of macrophages, resulting in the slow release of the incorporated interferon in the cells.

    Notes: To study the dynamics of adsorption and desorption of plasma proteins, bovine serum albumin(BSA), γ-globulin, fibrinogen, and plasma fibronectin(FN) were labeled with131 I and phosphate buffered solutions of the single proteins and of the mixed proteins were used for the in vitro adsorption and desorption study. When the proteins in single concentration were first adsorbed onto a polyethylene tube and desorbed after 60 min adsorption by changing the flowing liquid from the protein solution to a buffered saline without any protein, FN molecules were found to remain adsorbed so strongly on the tube surface that their desorption did not take place even after washing with the flowing buffer. On the contrary, BSA was rather readily desorbed from the tube surface upon continued washing. A similar result was obtained when the protein pre-adsorbed on a polyethylene film was exposed to the solution of the same but unlabeled protein; the adsorbed FN was not replaced, while the BSA solution expelled the adsorbed BSA molecules from the polyethylene surface. Adsorption and desorption behaviors of γ-globulin and fibrinogen were intermediate between those of BSA and FN. When a mixture of BSA and FN was allowed to flow into the polyethylene tube at a concentration of 0.05 mg ml-1 for each protein, BSA adsorption took place more rapidly and significantly than FN in the initial stage of adsorption but was decreased after passing a maximum, whereas FN was increasingly adsorbed onto the polyethylene surface with time and finally surpassed BSA in the adsorption amount.

    Notes: Oxidation of a polyethylene (PE) surface by corona discharge and the subsequent graft polymerization of acrylamide (AAm) were studied. The maximum amount of peroxides introduced by corona treatment at a voltage of 15 kV was about 2.3 × 10-9 mol cm-2. The decomposition rate of peroxide and the dependence of graft amount on the storage period of the corona-treated PE films showed that there were several kinds of peroxides, the labile one being mainly responsible for the initiation of graft polymerization. When the corona-treated film was brought into contact with a deaerated aqueous solution of AAm, graft polymerization took place more strongly with the treatment time, but was reduced after passing a maximum. Although the x-ray photoelectron spectroscopic analyses of the corona-treated PE films showed homogeneous oxidation of the outer polymer surface by corona discharge, optical microscopy on the cross section of the grafted film revealed the graft polymerization to be limited to a very thin surface region.

    Notes: To improve the low water wettability of poly(ethylene terephthalate) (PET), graft polymerization of acrylamide (AAm) by UV irradiation was performed onto the surface of a PET film with the simultaneous irradiation method without using a photo sensitizer. The PET film immersed in a 10 wt % deaerated aqueous solution of AAm was found to become highly hydrophilic upon UV irradiation. Optical microscopy on cross sections of grafted films showed that localization of the graft polymerization was restricted to a thin surface region of the film. Both the low concentration of polymer radicals formed by UV irradiation and the monomer penetration limited to the film surface would be responsible for localization of the grafted layer to the film surface region. Pretreatment of the PET film with benzyl alcohol was effective for enhancement of the graft polymerization. Retention of high hydrophilicity of the surface even after rigorous extraction of homopolymer and a comparative study of polymerization without UV irradiation strongly suggested that UV irradiation of the PET film under immersion in the deaerated AAm aqueous solution would lead to formation of the true graft copolymer.
