    Notes: The Bscherfchia coli urinary tract Isolate C1212 contains two pyelonephritis-associated pili (pap) DNA sequences designated here as pap-17 and pap-21. Each of these pap sequences encodes antigenically-distinct pilin monomers, pllin-17 and pilin-21, respectively. Most individual strain C1212 cells isolated from a single bacterial colony expressed pilin-21. Only a small fraction (5%) of strain C1212 cells expressed pilin-17. Most of the latter population simultaneously expressed pilin-21, but a low percentage of cells expressed pill composed of pilin-17 alone. In contrast, almost every E. coli K-12 cell containing multicopy pap-17 expressed pilin-17 at the ceil surface. These results Indicated that the regulation of pilin-17 expression observed for strain C1212 was lost when pap-17 was in the multicopy state. Transfer of pap-17 to a single copy vector resulted in a pilin-17 expression frequency lower than strain C1212 (1%). Using E. coli K-12 containing single copy pap-17, we found that the frequency of piiin-17 expression increased about 15-foid when pap-21 was present in multiple copies in trans. Subcloning of pap-21 showed that a 2.2 kilobase-pair DNA sequence adjacent to, but not including, the pilin-21 structural gene was sufficient for activation of pilin-17 expression.

    Notes: Common epitopes accessible to antibody on purified macromolecules or structurally altered gonococci may not be accessible to antibody when those macro-molecules are in their native state on the surface of intact organisms. To determine the immunologic accessibility of cyanogen bromide fragment 2 (CNBr2), a portion of the gonococcal pilin molecule that is common to all gonococcal strains on the surface of viable gonococci, probes composed of specific CNBr2 antibodies linked to gold spheres were manufactured. When whole piliated gonococci were exposed to these anti-CNBr2 immunological probes and examined using transmission electron microscopy, no significant marking of native pili was evident. These probes, however, detected CNBr2 in purified form. The epitopes encompassed within the CNBr2 portion of pili appear to be inaccessible to anti-CNBr2 probes within native gonococcal pili.

    Notes: Abstract Blastocysts and late gestation stages of the marsupial mouse, Antechinus stuartii, were examined cytologically and electrophoretically to investigate X chromosome activity during embryogenesis. A late replicating X chromosome was identified in the protoderm cells of female unilaminar blastocysts and in the cells of embryonic and extra-embryonic regions of older blastocysts. Sex chromatin bodies were also observed in female bilaminar and trilaminar blastocysts. The X linked enzyme α-galactosidase showed no evidence of paternal allele expression in the extra-embryonic region of bilaminar blastocysts or in the yolk sac and embryonic tissue of known heterozygotes. It is concluded that the late replicating X chromosome is paternal in origin and that unlike the laboratory mouse, X inactivation is not correlated with cell differentiation in Antechinus.

    Notes: Abstract Electrophoretic data for 15 enzymes encoded by 18 loci were analysed in samples of larval lampreys from two southern hemisphere species (Geotria australis and Mordacia mordax). Low values for Nei genetic distance coefficients for all loci in populations of G. australis from Western Australia, Tasmania and New Zealand (0.0013 to 0.0058) suggest that adults may intermix over distances greater than 4000 km. The average heterozygosity for four populations of G. australis (0.086) was much greater than for two populations of the far less widely distributed M. mordax (0.021).

    Notes: Abstract Nucleolar organiser regions (NORs) were examined in over 90% of the species of Australian ground frogs (familiy Myobatrachidae), including representatives from all twenty currently recognised genera and the three subfamilies. Throughout the family, location of the NOR within the karyotype showed considerable variation yet karyotype morphology showed uniformity. The precise mechanism(s) whereby variation in NOR location evolved while karyotype morphology was unchanged remains uncertain. Comparison of the two major subfamilies showed that the Limnodynastinae had a greater diversity of NOR location than the Myobatrachinae. The limnodynastine genus,Heleioporus, was the only one to show multiple NOR sites in several species. NOR location was particularly stable within most polytypic genera. Differences in NOR location within the remaining polytypic genera (Heleioporus, Limnodynastes, Neobatrachus, Philoria andRanidella) pointed to taxonomic discriminations that were generally consistent with recent proposals based on other criteria.

    Notes: Abstract The immunoaccessibility of the H8 antigen was examined as was the functionality of H8 specific monoclonal antibody (McAb 10). Immunoaccessibility of H8 varied among individual gonococci grown in vitro; the overall degree of variability was strain dependent. The H8 antigen was detected on the surface of most but not all gonococci found in urine sediment from an infected volunteer. The H8 specific McAb 10 was not bactericidal for strains of serum resistant gonococci; in the serum sensitive strain, an increase of approximately .5 logs above the bactericidal activity of antibody depleted normal human serum was observed. It is possible that the apparent paradox of the common, immunogenic, antigenic surface constituent H8 may be resolved in part by variation in immunoaccessibility within and among gonococcal strains.

    Notes: Abstract We show that the discrepancies among the various measurements of the radial velocity curve of the K5 V star in the dwarf nova SS Cyg have been caused by (1) poor choices of lines for measuring its velocity and (2) large distortions in its velocity caused by heating from the white dwarf and its accretion disk. The correctK velocity of the center of mass of the K5 V star is 158±3km s −1.

    Notes: Abstract SW UMa is an unusual dwarf nova in several respects. Recent observations by Robinsonet al. (1986) have established that SW UMa is an SU UMa system, while Shafter, Szkody, and Thorstensen (1986) have presented evidence suggesting that SW UMa may be a DQ Her system as well. If the DQ Her classification is confirmed then SW UMa will be the only system to have this dual classification. Finally, time resolved spectroscopy of SW UMa has revealed the presence of an S-wave component in the Hα emission which has an unexpected phasing relative to the line wings.

    Notes: A safer and more efficient method of mixing, granulating and drying (MIGRAD) highly energetic (UN 1.1) pyrotechnic composition will be reported. The process uses a MIGRAD mixer/granulator that was tailored to mix, granulate and dry a pyrotechnic material (20-40 lb) within a single mixing chamber. Features of the process include remote loading and unloading, reduced solvent, shortened mixing cycle (40 min), infinitely variable speed control of mixing and granulating impellers, built-in state-of-the-art fire detection/suppression (UV-50 ms, IR-10 ms), explosion venting, heated-chamber, vacuum drying, remote/programmable controls, and remote mixer cleanup using energy of the mixer. The mixing process will be evaluated for preparation of some 24 pyrotechnic compositions for five Arsenals/Ammo Loading Plants. Hazards Classification Safety Data Sheets were prepared for the 24 pyrotechnic compositions and will be reported along with resultant physical properties of the completed compositions.
