    Notizen: As has been previously reported one surge in cytosolic calmodulin is produced between 4 and 12 h after a partial hepatectomy. Moreoever, a surge in cytosolic cyclic AMP and another in cyclic AMP-dependent protein kinase activity can be detected during the late period of the prereplicative phase of liver regeneration after a partial hepatectomy. It is known that these three surges are involved in triggering DNA synthesis.By kinetic studies and by injecting transcription (actinomycin D) and translation (cycloheximide) inhibitors into hepatectomized rats we have demonstrated that the cyclic AMP-dependent protein kinase surge is produced by new synthesis of the enzyme. In thyroparathyroidectomized rats subjected to a partial hepatectomy the calmodulin surge was similar to that observed in normal hepatectomized rats whereas the surge in cyclic AMP-dependent protein kinase activity was strongly decreased suggesting that the cyclic AMP-dependent protein kinase surge is not generated by the previous surge in calmodulin.

Population genetics studies were carried out in Galicia (NW Spain) and two new ORM variants were found.

    Notizen: Sustained oscillations have been observed in continuous cultures of Saccharomyces cerevisiae. These oscillations appear spontaneously under aerobic conditions and may constitute a severe limitation for process control. We have found that oscillations arise only in a well defined range of dilution rates and dissolved oxygen values. The period of the oscillations is related, but not equal, to the mass doubling time, and shows a relation ship with both the parent cells and daughter cells generation times. At high dilution rates two oscillatory regimens, with different periods, are observed. The analysis of the budding index shows a marked degree of synchronization of the culture, however significant differences, both in phase and in amplitude, are ob served if the budding index of parent cells and of daughter cells are considered separately. The complex changes of the cell population are clearly demonstrated by the continuous and periodic modification of both cell volume distributions and protein distributions. Ethanol is always accumulated before the drop of dissolved oxygen concentration and one of the peaks of budding index. We propose a model that explains the insurgence of these oscillation as a consequence of changes in cell cycle parameters due to alternate growth in glucose and in ethanol.

    Notizen: Cellulose samples from cotton and wood pulps with varying low degrees of crystallinity (mechanically decrystallized) were studied. The influence of initial cellulose crystallinity on sugar yield after enzymatic hydrolysis was determined by two different methods. As expected, samples with low crystallinity were much more accessible to enzymatic attack and glucose yields were higher than were samples of high initial crystallinity. Hydrolysis of cellulose seems more dependent on cellulose crystallinity than on the source of cellulose. It is known that decrystallized or amorphous cellulose can recrystallize under proper conditions, e.g., during acid hydrolysis. The data reported here also reveal some recrystallization during enzymatic hydrolysis which probably occurs simulataneously with a selective enzymatic attack on the amorphous regions of cellulose. In all cases, the amorphous celluloses recrystallized in the original lattice form, that of native cellulose.

    Notizen: Method of flow cytometric analysis have recently been developed that make it possible to obtain segregated data on a single cell basis. In particular, it has been previously demonstrated that protein distributions obtained by flow cytometry give information about the law of growth of the cell population and the law of growth of the single cell; thus these distribution show how the microbial population is actually growing at the moment of the analysis and may yield more accurate and predictive information. We have extended the analysis of protein distribution and cell volume distribution to continuous cultures of Saccharomyces cerevisiae growing in a glucose-limited chemostat. We have found that: (1) to each dilution rate corresponds a given protein and volume distribution that does not change with time in steady state cultures; (2) there is a good proportionality between the average cell volume and the average protein content; (3) the protein distribution obtained can be easily analyzed with the model of growth of yeast previously developed in our laboratory; (4) the analysis of perturbed states shows that both protein distribution and volume distribution change very quickly; thus they are very sensitive parameters and can be used for monitoring and controlling industrial fermentation.

    Notizen: The influence of the following factors on the measurement of yeast concentration in liquid hydrocarbon fermentations was studied: surfactant, type (Tween 20, Tween 80, and G 3300), yeast concentration (0.215 to 0.272 and 3.37 to 4.08 g/liter, as dry matter), oil concentration (8.32 and 61.6 g/liter), surfactant, concentration (0.515, 1.030, 1.545, and 2.060 g/liter), and time of contact of the surfactant with the oil-yeast-aqueous medium mixture (2 and 10 min). The statistical significance of the obtained results was determined.

    Notizen: Three anaerobic fluidized bed reactors at 37°C were utilized to observe the effects of startup and operational procedures on biomass attachment. Using a meat-based synthetic waste and stepped-loading regime, the influences of synthetic polymer addition and maintenance of anaerobiosis during startup were investigated. Subsequently, increasing bed expansions were applied to assess shear effects. Synthetic polymer addition enhanced biomass retention but did not improve process performance. Maintenance of a reduced environment ameliorated fluctuating process parameters during start up and aided biomass retention and substrate removal. A bed expansion of 5% was detrimental to biomass attachment and COD removal but system stability was maintained at expansions between 10% and 30%. Startup was achieved in 56 days. Anaerobiosis appeared to enhance the initial evolution of a stable, well-adapted microbial population, whereas polymer addition interfered with this. Moderate bed expansions had negligible effects on attachment and performance.

    Notizen: This work shows that the enzyme membrane reactor offers the opportunity to carry out the enzymatic regeneration of ATP providing continuous operation with high performance. In this system, the coenzyme is immobilized on a water-soluble polymer. These high-molecular weight derivates are entrapped within an ultrafiltration membrane together with the enzymes for production of regeneration. Several polymer derivatives of ATP have been prepared for this immobilization technique. Coenzymatic activity of these derivatives was studied with several enzymes for both ATP-requiring and ATP-regenerating reactions. PEG-N6-aminohexyl-ATP was selected as the appropriate coenzyme for operating the enzyme membrane reactor. Acetate kinase was the only enzyme providing enough activity for regeneration. Production of glucose-6-phosphate is cited as the first example. The kinetics of acetate kinase and hexokinase were examined and used to choose the operating conditions of the process. The process operated continuously for more than 1 month. With a mean conversion of 80%, the space-time yield amounted to 348 g glucose-6-phosphate/L d. The cycle number of ATP was estimated as 20, 000 mol/mol. With the continuous production of γ-glutamylcysteine and NADP, the feasibility of the system was proven.
