    Notes: Abstract A simple and convenient procedure that obviates the need for DNA cloning and radiolabelling is described for producing a rDNA hybridisation probe for use in bacterial strain characterisation. Bulk rRNA, comprising a mixture of 16S and 23S components, was isolated from Providencia stuartii NCTC11800 and purified by CsC1-cushion ultracentrifugation. The pure RNA was used as a template for synthesis of a single-strand copy cDNA using reverse transcriptase and random primers, with simultaneous biotin-11-dUTP labelling. EcoRI digests of chromosomal DNA from the type strain and a sample of 10 clinical isolates of P. stuartii were examined by Southern blot hybridisation on nylon membranes using the cDNA probe. All 11 strains had similar but not identical hybridisation patterns of 5–7 bands (detected colorimetrically) with sizes of about 5–19 kb. Our results indicated that the probe provided a sensitive means of detecting rRNA cistrons in P. stuartii. The method should be applicable to any group of microorganisms.

    Notes: Concentrations of cyanocobalamin (vitamin B12) ranging from 0.64-186.6 μg/100g (fresh weight-edible tissue) were determined for seven different varieties of seafoods by two radioisotope dilution (RID) methods. The correlation coefficient for 156 RID seafood assays using both techniques was 0.99 and the mean relative standard deviation for samples was 12.05 ± 5.19%. The reproducibility and comparability of the two RID assays was attributable to the uniform implementation of a rigorous extraction procedure which isolated cyanocobalamin in an aqueous phase prior to radiometric evaluation.

    Notes: Environmental effects of feedlot location and related land disposal operations can be minimized if proper knowledge of hazardous conditions are known and appropriate protective steps taken. Common guidelines often do not apply because of differing physical characteristics of local areas.Analyses of various parameters within a soil profile beneath a feedlot revealed none of the chemical constituent present in high concentrations below the 23-foot depth. In areas where shallow ground water was less than 5 feet from the surface, the ground water was found to be affected by the feedlot. Other hazardous areas in feedlot location are flood-prone areas, areas of surface bedrock, and areas of excessive slope.For land disposal operations, loading rates and frequency of application of feedlot waste should be adjusted in accordance with soil permeability, depth to ground water, and irrigation practices to minimize detrimental effects on ground-water quality.

    Notes: Water samples were collected from 34 wells in the Claiborne aquifer in southwest Georgia. Land use/cover is principally forest, wetland, or agriculture. The aquifer is exposed surficially to the northwest but dips beneath confining beds to the southeast. The Claiborne aquifer is generally a fine- to medium-grained quartz sand becoming more calcareous downdip and to the east. Well water was analyzed in the field for pH, electrical conductivity, alkalinity, hardness, and temperature, and in the laboratory for Cl, NH4, NO3, total N, total P, ortho-P, SO4, K, Na, Ca, Mg, and Fe. Hardness gradually increases from 5 mg/1 updip to 162 mg/1 downdip and bicarbonate alkalinity follows the same trend reflecting variations in the aquifer lithology. Higher than normal nitrate concentrations (4-6 mg/1) are found in ground water in the exposed updip portion of the aquifer in areas where agricultural land use for cropland is high. However, in forest-covered areas, ground water in the exposed portion of the aquifer contains nitrate concentrations similar to the confined areas of the aquifer: less than 1 mg/1. Other chemical constituents measured do not show a discernible pattern of variation. One well near a fertilizer storage facility contains nitrate concentrations exceeding the Safe Drinking Water Standards, and other wells in the agricultural portion of the outcrop belt may seasonally exceed these limits.

This paper develops a measure for software effectiveness and a computer aid for determining process structure in program module specification. The proposed method uses various criteria and a mixed integer program to decompose a system graph into program modules.

    Notes: Heat transport by large-scale groundwater movement, reported for a number of sedimentary basins, may cause sufficiently severe perturbations to the thermal regime that the patterns of near surface heat-flow density (HFD) may be very different from that of the desired deep-seated conductive HFD. However, a good interpretation of the HFD data from perturbed areas is possible when knowledge of both the thermal and hydrological regimes is available, but there are three major difficulties. First, the thermal and hydrological data are often noisy; second, the two regimes are closely coupled; third, the deep-seated, or basal HFD is a very uncertain boundary condition. In this paper, we present a method, currently in 2-D, that inverts the noisy thermal and hydrological data simultaneously, and is able to resolve uncertain boundary conditions. The temperature and hydraulic head and the thermal conductivities and intrinsic permeabilities of the subsurface materials are first parameterized with an isoparametric finite element model. The parameters are then estimated using a Bayesian type non-linear inverse method. In the finite element formulation, the boundary heat and water fluxes are linearly transformed into an equivalent nodal flow (ENF) vector, which is updated together with the estimated parameters. Another (linear) inversion of the updated ENF components gives the updated boundary fluxes, with variances. The applications of the boundary flux updating technique to the determination of the background HFD in hydrologically active areas are illustrated with numerical examples.

    Notes: The effect of 14 combinations of photoperiod, soil and air temperature, and growth substance applications on the cold hardiness of Chrysanthemum morifolium‘Astrid’ rhizomes was evaluated. Both triphenyl tetrazolium chloride and regrowth tests were used to determine the viability of the cold-stressed rhizome tissues. The rhizomes exhibited different degrees of cold hardiness under these environmental conditions. A combination of short photoperiod and low air and soil temperatures induced maximum cold hardiness. Low soil temperature accompanied by long photoperiods and warm aerial temperatures did not induce rhizome hardening, while some hardening in cool soils was evident under either short photoperiods or low aerial temperatures. Warm soils reduced rhizome hardening under the normally inductive short photoperiod-cool aerial conditions. Since the induction of rhizome hardening was dependent on the induction of the aerial organs, the involvement of translocatable hardiness promoters is indicated. Foliar applications of low levels of gibberllic acid (GA3) or abscisic acid only slightly influenced rhizome hardiness.
