    Notes: Summary An assay has been developed for quantitating the reassociation of cortical secretory vesicles (CVs) with fragments of sea urchin egg plasma membrane attached to glass slides (PM lawns). Binding ofS. pupuratus CVs to homologous PM lawns increased with time and CV concentration. The observation that CV binding was blocked by chymotrypsin digestion of the PM fragments suggested that a PM protein(s) is required for reassociation. The possibility that the extent of CV lysis that occurred during CV preparation (15.4±3.8% as assessed by ovoperoxidase assay) influenced reassociation was investigated by determining the effect of CV content proteins (isolated as fertilization product) on binding. Various concentrations of fertilization product (up to equivalent amounts of fertilization product and CV protein) had no effect on CV binding. The specificity of binding was investigated by assessing the ability of CVs to bind to PM lawns prepared from human red blood cells, and by determining the ability of heterologous vesicles to bind to egg PM fragments. PM lawns from HRBCs did not support CV binding; however, PM lawns prepared from the eggs of several species of sea urchin did bindS. pupuratus CVs. Vesicles from a partially purified preparation of yolk platelets bound to egg PM lawns with low efficiency (1/7 that of CVs), but immunofluorescence analysis with an anti-hyalin monoclonal antibody demonstrated that 74±9% of the bound vesicles were CVs that contaminated the yolk platelet preparation. Dioleoylphosphatidyl choline liposomes were also unable to bind to egg PM lawns. These results are consistent with hypothesis that CV binding to egg PM lawns is a specific, protein-mediated event.

    Notes: Single-photon dissociation of (CH3)2Zn has been examined at 248 nm in the gas phase. The relative yields of CH3Zn and Zn as a function of buffer gas pressure were determined by laser-induced fluorescence. With He as the buffer gas, the yield of CH3Zn increases from ∼20% at very low pressure to ∼65% at 500 Torr. The yield of Zn atoms correspondingly drops over the same pressure range. This behavior is due to the increasing probability for collisional stabilization of the primary photoproduct, vibrationally hot CH3Zn, as the buffer gas pressure increases, thereby quenching its subsequent thermal dissociation to CH3 and Zn. Information on the nascent vibrational energy distribution of the CH3Zn photoproduct is obtained by examining the probability of CH3Zn stabilization vs buffer gas pressure.

    Notes: The effects of reaction and transport kinetics on deposition resulting from laser-induced gas-phase photodissociation were investigated using numerical and analytical models. Deposition rates and deposit profiles for a laser beam focused onto a substrate in a chamber were determined assuming production of metal atoms in the gas phase by a single-photon dissociation mechanism and free-molecular and diffusive transport to chamber surfaces. The predictions of the model computations were compared to experimental and theoretical results from the literature. The effect of total pressure on deposition rates and profiles depends strongly on the sticking coefficient. With a sticking coefficient of unity, deposition rates and profiles do not depend strongly on the total pressure or transport regime. However, when the sticking coefficient is 0.01, the deposit profile flattens dramatically at pressures below about 1 atm. The conditions and consequences of gas-phase reactant depletion due to diffusional limitations were also demonstrated. The extent of gas-phase reactant depletion can be predicted by a single dimensionless group; however, transport of the reaction product must also be considered to predict deposition rates. At high laser power or total pressure, reactant depletion leads to reduced deposition rates and flatter deposit profiles, and severe depletion can produce volcano-shaped deposits.

    Notes: The gas-phase photodissociation of (CH3)2Zn, (C2H5)2Zn, and (n-C3H7)2Zn has been examined at 248 nm using laser-induced fluorescence to detect the monoalkyl zinc radical and zinc atom photoproducts. For each compound, the monoalkyl zinc radical is the primary photoproduct and is formed sufficiently hot that it spontaneously dissociates to an alkyl radical and a Zn atom without absorption of a second photon. Photodissociation was examined in the presence of He buffer gas to measure the probability of quenching the secondary spontaneous dissociation of the monoalkyl zinc species. For all three dialkyl zinc compounds, the probability of quenching the secondary dissociation step increases substantially over the He pressure range of 0–400 Torr. The quenching probability vs He pressure was fit using RRKM theory in conjunction with a time-dependent master equation, treating the nascent vibrational energy distribution of the monoalkyl zinc product as an adjustable function. The quenching data for C2H5Zn and n-C3H7Zn can be fit only if it is assumed that these species are formed with a hot, narrow vibrational energy distribution, much narrower than that predicted by phase-space theory. A dissociation mechanism involving crossover from an optically prepared singlet state to a repulsive triplet state is proposed to explain this observation. Spontaneous dissociation of CH3Zn is quenched much more strongly by He than is calculated using any reasonable vibrational energy distribution function for CH3Zn. This is attributed to the inapplicability of RRKM theory to reactions involving very low-state-density molecules like CH3Zn.

    Notes: Undeveloped and unbaked paterns or latent images (LI) in polymethylmethacrylate (PMMA) have been observed and measured using different measurement techniques. Results from two common-path baseband optical techniques have been compared with the results of profiling LI using an atomic force probe. These results indicate that 5 A(ring) detection limits in optical path length measurements are easily achievable at near video rates using simple optical systems.

    Notes: Abstract Agroecosystems contain about 12% of the terrestrial soil C and play an important role in the global C cycle. We describe a project to evaluate the degree to which management practices can affect soil C in agroecosystems. The objectives of the project are to determine if agricultural systems can be managed to conserve and sequester C and thereby reduce the accumulation of CO2 in the atmosphere, and to provide reference datasets and methodologies for agricultural assessments.
