    Description / Table of Contents: Zusammenfassung Die Beziehungen zwischen der zeitlichen Veränderung der Charakteristika von Magmenquellen und der Öffnung des Japanischen Meeres werden anhand von Sr-Isotopendaten tertiärer und quartärer basaltischer Gesteine diskutiert. Basaltische Gesteine von der Grabenseite und aus der Übergangszone ergaben initiale87Sr/86Sr Verhältnisse (Sri-Verhältnisse) von 0.70411–0.70546 und lassen keine zeitabhängige Änderung erkennen. Basaltische Besteine aus dem Back-Arc-Bereich mit Altern zwischen 29.8 und ca. 15 Ma zeigen ähnliche Sri-Verhältnisse und ebenfalls keine zeitliche Veränderung. Im Gegensatz dazu sind basaltische Gesteine aus dem Back-Arc-Bereich, die jünger als ca. 15 Ma sind, signifikant in ihren Sri-Verhältnissen (0.70396–0.70290) erniedrigt. Diese Verhältnisse liegen etwas höher als die von N-MORB. Die Sr-Isotopenergebnisse lassen vermuten, daß zumindest vor ca. 15 Ma der Herkunftsbereich der Magmen (subkontinentaler Mantel) unter dem NE japanischen Vulkanbogen chemisch angereichert war, während die Magmenquellen der jüngeren vulkanischen Gesteine des Back-Arc-Bereiches durch eine drastische Abreicherung charakterisiert sind. Die abgereicherten Magmen könnten, während der Öffnung des japanischen Back-Arc-Beckens, als Folge der Injektion abgereicherter Asthenosphäre (oder eines abgereicherten Manteldiapirs) in subkontinentalen Mantel unterhalb des Back-Arcs des NE japanischen Vulkanbogens, gebildet worden sein. Die miozänen basaltischen Gesteine des Back-Arc-Bereiches sind außerdem durch niedrigere Gehalte an LIL-Elementen, wie z.B. K2O und Rb charakterisiert. Dies wird als Hinweis auf eine erhöhte Aufschmelzungsrate in diesem Bereich im mittleren Miozän (im Zuge der gleichzeitigen Öffnung des japanischen Meeres) verstanden. Die erhöhte Aufschmelzrate im Mantel des Back-Arc-Breiches wird auf einer Erhöhung des geothermischen Gradienten infolge der Injektion von heißer Asthenosphäre zurückgeführt. Diese Injektion von Asthenosphäre könnte auch der Grund für die Aufschmelzung von Unterkruste und für die Produktion weitverbreiteter saurer miozäner Vulkanite im Back-Arc-Bereich und der Übergangszone sein.
    Notes: Summary Based on Sr isotopic data for Tertiary and Quaternary basaltic rocks from the NE Japan arc, relationships are discussed between the temporal variation of magma source characteristics and the opening of the Japan Sea. The basaltic rocks from the trench side and from the transitional zone show initial87Sr/86Sr ratios (Sri ratios) in the range of 0.70411–0.70546 but no temporal variation in Sri ratios. The back-arc side basaltic rocks with ages of 29.8 to ≈ 15 Ma have Sri ratios similar to those of the trench side and the transitional zone, and these values also show no temporal change. In contrast, the basaltic rocks from the back-arc side, with ages younger than ≈ 15 Ma, show significantly lower Sri ratios (0.70396 to 0.70290), which are slightly higher than those of N-type MORB. These Sr isotopic features may imply that at least before ≈ 15 Ma the magma source regions (the sub-continental mantle) beneath the NE Japan arc had an enriched chemical character and that after ≈ 15 Ma, the magma sources for volcanic rocks from the back-arc side show a drastic change in Sr isotopic character, from an enriched nature to a depleted one. The depleted magmas may have been formed as a result of injection of depleted asthenosphere (or of a depleted mantle diapir) into the subcontinental mantle under the back-arc side of the NE Japan arc, during the spreading of the Japan Sea back-arc basin. The middle Miocene basaltic rocks from the back-arc side are characterized by lower contents of LIL elements such as K2O and Rb compared with those from the trench side, suggesting that during the middle Miocene (syn-opening stage of the Japan Sea) the degree of partial melting may have been higher in the back-arc side mantle than in the trench side mantle. High degree of partial melting in the back-arc side mantle can be attributed to an increasing geothermal gradient in the mantle due to the injection of hot asthenosphere. This injection might also have caused the melting of the lower crust from which the voluminous middle Miocene acidic volcanics in the back-arc side and transitional zone may have been produced.

    Notes: Summary New multidentate phenyl-bonded phases (MPBPs) were synthesized and evaluated the chromatographic retention behaviour with polycyclic aromatic hydrocarbons as sample probes in high-performance liquid chromatography. The new MPBPs show different retention characteristics from the previously synthesized MPBP, designated TP. Nevertheless, the results indicate that the retention mechanism is still the same as the TP phase: the size and shape of the solute molecule can be recognized by a cavity-like space formed by the methyl groups and phenyl rings of MPBPs.

    Notes: Abstract The localization of cathepsins B, D, and L was studied in rat osteoclasts by immuno-light and-electron microscopy using the avidin-biotin-peroxidase complex (ABC) method. In cryosections prepared for light microscopy, immunoreactivity for cathepsin D was found in numerous vesicles and vacuoles but was not detected along the resorption lacunae of osteoclasts. However, immunoreactivity for cathepsins B and L occurred strongly along the lacunae, and only weak intracellular immunoreactivity was observed in the vesicles and peripheral part of the vacuoles near the ruffled border. In control sections that were not incubated with the antibody, no cathepsins were found in the osteoclasts or along the resorption lacunae of osteoclasts. At the electron microscopic level, strong intracellular reactivity of cathepsin D was found in numerous vacuoles and vesicles, while extracellular cathepsin D was only slightly detected at the base of the ruffled border but was not found in the eroded bone matrix. Most osteoclasts showed strong extracellular deposition of cathepsins B and L on the collagen fibrils and bone matrix under the ruffled border. The extracellular deposition was stronger for cathepsin L than for cathepsin B. Furthermore cathepsins B and L immunolabled some pits and part of the ampullar extracellular spaces, appearing as vacuoles in the sections. Conversely, the intracellular reactivity for cathepsins B and L was weak: cathepsin-containing vesicles and vacuoles as primary and secondary lysosomes occurred only sparsely. These findings suggest that cathepsins B and L, unlike cathepsin D, are rapidly released into the extracellular matrix and participate in the degradation of organic bone matrix containing collagen fibrils near the tip of the ruffled border. Cathepsin L may be more effective in the degradation of bone matrix than cathepsin B.

    Notes: Abstract Immunohistochemical localization of cathepsins B, D and L in the osteoclasts of rat alveolar and femoral bones was investigated by using the avidin-biotin-peroxidase complex method for semithin, 1-μm-thick cryosections. Extracellular immunoreactivity for cathepsins B and L was clearly demonstrated along the bone resorption lacunae; the intensity of the extracellular immunoreactivity of cathepsin L was stronger than that of cathepsin B. However, the intracellular immunoreactivity of both cathepsins was weak compared with that of cathepsin D. The intracellular immunoreactivity of cathespin D in the osteoclasts was clearly observed in the granules and/or vacuoles, but extracellular cathepsin D immunoreactivity was either negligible or not detected along the resorption lacunae. In the adjacent sections stained with anti-cathepsin L or D, extensive extracellular deposition of cathepsin L was found along the bone resorption lacunae, with or without osteoclasts, although the intracellular reactivity of cathepsin L was weak. This is the first morphological study in which cathepsins B and L have been demonstrated to be produced in the osteoclasts and extensively secreted into resorption lacunae, and in which cathepsin D was found to be present in the cells but scantily secreted into the lacunae. These findings suggest that cathepsins B and L directly and effectively participate in the degradation of the bone matrix.

    Notes: Summary We performed immunocytochemical localization of cathepsin D in osteoclasts of the proximal growth plate of the rat femurs using both the avidin-biotin-peroxidase complex method for cryo-semi-thin (1 μm) sections and the colloidal gold-labeled IgG method for K4M ultra-thin sections. At the light microscopic level, cathepsin D immunoreactivity in the osteoclasts appeared at the vesicles, granules, and/or small vacuoles. They were distributed throughout the cytoplasm of each cell and were relatively numerous close to the bone surface. This antigen could not be detected at the eroded bone surface. As for other cells, immunoreactivity was seen only in the lysosomes of osteoblast-like cells. Immunoreactivity in the osteoclasts was stronger and greater in the density and number than in osteoblast-like cells. At the electron microscopic level, osteoclasts with well-developed ruffled border possessed numerous cathepsin D-containing lysosomes, vacuoles, and coated vesicle-like structures. Cathepsin D-containing lysosomes fused with cathepsinnegative vacuoles and formed large secondary lysosomes. Osteoclasts with poorly developed ruffled border possessed fewer cathepsin D-containing lysosomes than those with well-developed ruffled border. No immunogold particles were seen in vacuole-like channel expansions of the ruffled borders, between the channels of the ruffled borders, or on the eroded bone surface. These findings demonstrate that osteoclasts contain a large amount of cathepsin D. They suggest that cathepsin D is necessary for osteoclastic bone resorption, that it plays an indirect rather than direct role.

    Notes: Abstract The ion-exchange reaction of lysozyme with carboxymethyl Sephadex C-25 was followed by conductivity change as a function of time just after the rapid mixing of the protein solution with the Sephadex suspension. A single relaxation process was observed; the conductivity increased exponentially with time in the 100 s scale. In this process, protons were released from the Sephadex C-25 in the same time scale. The relaxation process slowed down with an increase in the lysozyme concentration, but it quickened upon the addition of HCl. On the other hand, the ζ potential on the Sephadex C-25 surface changed from a negative value to a positive one with an increase in the amount of lysozyme adsorbed on the surface. On the basis of these data, the relaxation process was attributed to the ion-exchange reaction of lysozyme with several protons of carboxymethyl groups of the Sephadex.

    Notes: Abstract For the formation of recombinants involving closely linked markers, two distinct processes play a role. The recombinational interaction between homologous DNA molecules results in the presence of heteroduplex DNA joining the parental components of the recombinant. The presence of markers distinguishing the parents in the region of heteroduplex DNA can result in base pair mismatches. The post recombination repair of such mismatches can contribute to the separation of closely linked markers. The processes responsible for such repair also play roles in mutation avoidance. The specificities, functions and contribution to the formation of recombinants for closely linked markers of the processes inEscherichia coli are described.

Pentatomidae) entre en diapause imaginale en jours courts, lorsqu'il s'alimente avec des feuilles ou des graines de Brassica napus L. En jours longs, les adultes élevés sur feuilles se reproduisent; par contre, élevés sur graines, ils entrent en diapause, bien que leur alimentation ait été meilleure que sur feuilles à en juger par la mortalité et le gain de poids pendant le dernier stade larvaire. On suggère que E. rugosa utilise l'état de la plante hôte comme moyen de régler l'induction de la diapause.
    Notes: Abstract Eurydema rugosa Motschulsky (Heteroptera: Pentatomidae) entered adult diapause under a short-day photoperiod, whether the food was leaves or seeds of Brassica napus L. Under a long-day photoperiod, adults raised on leaves became reproductive. However, adults raised on seeds entered diapause even under a long-day photoperiod, although they were nutritionally better than leaves in respect of mortality and weight gain in the nymphal period. It is suggested that E. rugosa uses the status of the host plant as a cue for timing the diapause induction.

    Notes: Abstract The effect of cis-propenylphosphonic acid (PPOH), a structural analoge of ethylene, on flower wilting and ethylene production was investigated using cut carnation flowers which are very sensitive to ethylene. Wilting (petal in-rolling) of the flowers was delayed by continuously immersing the stems in a 5–20 mM PPOH solution. In addition, the continuous treatment with PPOH markedly reduced autocatalytic ethylene production of the petals accompanying senescence. This reduction of autocatalytic ethylene production was considered responsible for the inhibitory effect of PPOH on flower wilting. The inhibitory activity of trans-propenylphosphonic acid (trans-PPOH), on both flower wilting and the autocatalytic ethylene production accompanying senescence was markedly lower than that of PPOH, suggesting that PPOH action is stereoselective. PPOH may be of interest as a new, water-soluble inhibitor of wilting and autocatalytic ethylene production in cut carnation flowers.
