    Notes: The exotoxin pertussis toxin (PT) produced by virulent Bordetella pertussis bacteria is regarded as the main virulence factor of the organism and held responsible for most of its pathological effects. Identification of functional sites on PT would greatly facilitate site-specific detoxification and thus also the development of a new vaccine. For the investigation of structure-function aspects of PT we have prepared and characterized eleven monoclonal antibodies (mAbs) (UB-A1, UB-A2, UB-A10, UB-B7, UB-B12, UB-D4, UB-D7, UB-D10, UB-F7, UB-G1, and UB-G12) directed at the native toxin. Only UB-B12 and UB-D10 recognized PT in Western blotting indicating that most of the mAbs were directed against conformational epitopes. The mAbs were assayed for their ability to interfere with the binding of PT in model receptor systems like a solid phase binding assay using fetuin as receptor moiety, hemagglutination of chymotrypsin-sensitized goose erythrocytes, and the PT-mediated induction of the clustered growth pattern (CGP) of Chinese hamster ovary (CHO) cells. Five of the eleven mAbs (UB-A1, UB-A2, UB-B7, UB-B12, and UB-D7) interfered with the binding of PT to fetuin on solid phase and with PT-mediated hemagglutination. UB-A2, UB-B7, and UB-B12 also inhibited the induction of the clustered growth pattern of CHO-cells. This indicates that the determinants recognized by these mAbs are associated with the formation of the carbohydrate recognition sites of PT. Thus, the monoclonal antibodies described in this study will be valuable tools in the further analysis of the structure-function relationship of pertussis toxin with respect to receptor recognition and binding.

    Notes: The adherence mechanisms of enteropathogenic Escherichia coli (EPEC) to epithelial cells are still not understood. To study the molecular basis of the diffuse adherence (DA) phenotype exhibited by diarrhoeagenic E. coli expressing classical EPEC serotypes we investigated strain 2787 (O126:H27) isolated from a case of infantile diarrhoea. A 6.0 kb plasmid-derrved DNA fragment mediates the DA phenotype and encodes the 100 kDa adhesin protein AIDA-I (adhesin involved in diffuse adherence). Sequencing of the entire fragment revealed two open reading frames which encoded proteins of 45 kDa and 132 kDa, respectively. The 132 kDa protein has been identified as an AIDA-I precursor protein. After cleavage of the signal sequence further processing at the C-terminus of the 132 kDa precursor leads to the mature ∼100 kDa AIDA-I. While the exact function of the cytopiasmic 45 kDa protein is not known, preliminary evidence indicates that it is necessary for the correct maturation of AIDA-I. The AIDA-l precursor exhibits significant homology with the virG(icsA) protein of Shigella flexneri which seems to be involved in the intercellular spread of invasive Shigella organisms.

    Notes: Liquid Crystals with Aliphatic Groups between Cyclohexane Rings - Synthesis of 1,4-Bis(4-n-alkylcyclohexyl)butanes and Esters of 4-[4-(4-n-Alkylcyclohexyl)-butyl]cyclohexanolsThe synthesis of 1,4-bis-(4-n-alkylcyclohexyl) butanes (1), esters of 4-[-(4-n-alkylcyclohexyl)butyl]cyclohexanols (2) and some analogous aromatic compounds are described. The thermal stability of mesophases of these compounds are in the order of the analogous ethylene-bridged liquid crystals. The reason for this result seems to be the combination of molecular rigidity and conformational flexibility.

    Notes: The chain stiffness of a methoxypropyl-substituted poly[1.1.1]propellane in solution is investigated by static and dynamic light scattering. The Kuhn statistical segment length is determined as lk=150Å utilizing the Kratky-Porod wormlike chain model. It is, however, not yet clear whether the derived Kuhn length characterizes the chain stiffness of a perfect polypropellane chain or rather originates from “kinks” in the polymer backbone due to the presence of possible side reactions during polymerization.

Treatment of these two compounds with 85% tetrafluoroboric acid. diethylether adduct yields di-cyclo-hexoyl- (1b) and diadamant-1-oylphosphine (1c). In nmr spectroscopic studies 1b over a range of 203 to 343 K, a strong temperature dependence of the keto-enol equilibrium is found; thermodynamic data characteristic for the formation of the enol tautomer (ΔH0 = -4.3 kJ. mol-1; ΔS0 = -9.2 J. mol-1. K (-1) are compared of 1,3-diketones.The enol tautomer of diadamant-1-oylphosphine (E-1c) as obtained from a benzene solution in thin colourless plates, crystallizes in the monoclinic space group P21/c {a = 722.2(2); b = 1085.5(4); c = 2434.8(5) pm; ß = 96.43(2)° at -100 ± 3°C; Z = 4}. An X- ray structure analysis (Rw = 0.033) shows bond lengths and angles to be almost identical within the enolic system (P—C 179/180; C—O 130/129; C—C(adamant-1-yl) 152/153 pm; C—P—C 99°; P—C—O 124°/124°; P—C—C 120°/120°; C—C—O 116°/116°. The geometry of the very strong, but probably asymmetric O‥H‥O bridge is discussed (O—H 120/130, O‥O 245 pm).

Nach den Ergebnissen der Röntgenstrukturanalyse (Rw=0,033) weist das Molekül in beiden des Hälften des Enolrings nahezu identische Bindungslängen und -winkel auf P—C 179/180; C—O 130/129; C—C(Adamant-1-yl) 152/153 pm; C—P—C 99°; P—C—O 124°/124°; P—C—C 120°/120°; C—C—O 116°/116°. Die Geometrie der sehr starken, aber vermutlich asymmetrischen O‥H‥O-Brücke wird diskutiert (O—H 120/130; O‥O 245 pm).

    Notes: A specific and sensitive method for the determination of ethylenediamine in human plasma is described. The aqueous sample is treated with m-toluoyl chloride, yielding the N,N′-bis(m-toluoyl) derivative of the diamine, which is extracted into dichloromethane and quantitated after “on column” methylation by capillary gas liquid chromatography with alkali flame ionization detection. The corresponding derivative of putrescine serves as internal standard. The assay is reproducible and calibration curves are linear over the concentration range 0.05 to 10 μg · ml-1. The lower detection limit is about 10 ng · ml-1. The structures of the compounds of interest eluting from the capillary column are examined by gas liquid chromatography/mass spectrometry. The assay has been applied to the analysis of ethylenediamine in plasma following the administration of aminophylline and ethylenediamine in a cross-over study to patients with bronchopulmonary diseases. The method also proves suitable for measuring other primary and secondary amines and diamines in aqueous solutions by gas liquid chromatography.
