Notes: Summary Intact and castrated juvenile male rainbow trout (Salmo gairdneri) were treated with testosterone and gonadotropic hormone (GTH) to determine the maturational effects of these hormones on the GTH-cells. Electron-microscopic studies of the GTH-cells revealed that GTH and testosterone in intact animals, and testosterone in castrated fish, caused GTH-cell maturation: These cells now displayed the same appearance as GTH-cells in adult trout, including the presence of globules, a well-developed Golgi apparatus and rough endoplasmic reticulum, all of which were absent in GTH-cells of control animals. Animals with stimulated GTH-cells also had an increased GTH content of the pituitary; release of GTH could not be demonstrated. Animals treated with GTH exhibited an accelerated development of the testes, resulting in complete gametogenesis and elevated plasma testosterone levels. These results indicate that exogenous steroids as well as endogenous gonadal steroids can stimulate the full development of GTH-cells and accelerate GTH synthesis. The significance of this stimulating effect of the gonadal hormones with respect to the development of the brain-pituitary-gonadal axis and the onset of puberty is discussed.

Notes: Summary Effects of steroids on the accumulation of glycoprotein gonadotropin (GTH) in pituitaries of juvenile trout were investigated by means of scanning cytophotometry applied to immunocytochemical preparations, and with the use of a radioimmunoassay. Effects on other aspects of GTH-cell activity were analyzed by measuring the size of the gonadotrops and their nuclei. Progesterone added to aquarium water and methyltestosterone incorporated into the food showed a pronounced stimulatory effect on the accumulation of GTH. To a lesser extent, treatment with cortisol, cortisone, and desoxycorticosterone acetate administered to aquarium water, and 11β-hydroxy-androstenedione added to the food resulted in an increase of the hypophysial content of GTH. Steroids stimulating the accumulation of GTH in the pituitary also exhibited a positive effect on GTH-cell activity as indicated by an increase in the size of gonadotropic cells. Progesterone incorporated into the food did not influence the GTH-content and the GTH-cell activity. It is suggested that the route of administration of an exogenous steroid is essential for its effect on GTH cells in trout. Comparison of GTH values reveals an excellent correlation between the data from the radioimmunoassay and those from the corresponding densitometric measurements. No correlation was observed between values of morphometrically determined GTH-cell activity and the densitometric values reflecting hypophysial GTH content.

Description / Table of Contents: X-Ray Stress Measurement on Austenitic Steels - A Possibility of Predicting Fatigue DamageBased on the results found for fatigue tests on a ferritic steel combined with x-ray stress analysis similar investigations were carried out for an austenitic stainless steel. For that reason the Wöhler tests were interrupted after defined load cycle numbers and the residual stress and the half value breadth of the x-ray peaks were determined in the groove of the samples. The investigation has shown correlations between residual stress, load amplitudes and load cycle numbers. These relations in connection with a dependence of the breadth of the x-ray peaks from the load cycle number have been used for a non-destructive testing model of predicting the life of samples under alternating load.

Description / Table of Contents: The Effect of Moisture on the Mechanical Properties of Phenolic Moulded MaterialsIn order to use thermosetting moulding material it is necessary to know how the mechanical properties are changed by absorption of water. Therefore, multiphase systems based on phenolic resin, with varying quantities of wood flour and/or powdered minerals as fillers, were examined. The relationship between moisture absorption and the change in Young's modulus, flexural strength and the strain of the outer fibre at fracture was determined. It could be shown that the increase in flexural strength as water absorption began is a result only of the swelling stress.

Description / Table of Contents: The Swelling Behaviour of Thermosetting MaterialsThe moisture absorption of moulded materials can endanger a construction because of the change in volume of various parts. Therefore it is necessary to know the maximum possible swelling of the moulded materials employed at different relative humidities. The swelling of modified phenolic resin systems are described. Wood flour (which absorbs moisture) and powdered minerals (which in this case can be considered as inert) were used as fillers.Different methods have been developed to calculate the maximum possible swelling after a relatively short testing time (ca. 1000 h).

Description / Table of Contents: Investigations of the Local Composition of Glass Fibers with the Help of the Scanning Electron MicroanalysisVarious glass-fiber types show different chemical resistance in the surrounding of aggressive media. Through the influence of inorganic acid or alkali media certain elementary glass fibres can be leached. This effect can be seen optically in some kinds of glass-fibers. Other glass-fiber types show no optical change in the surface, although there is an aggressive attack Now it is possible to understand the leaching process in glass-fibers with the help of the scanning electron microanalysis. In that way corrosion procedures of various glass-fiber types can be better described.

Notes: Glass capillary gas chromatography has been investigated as a method for the analysis of nitrated polycyclic aromatic hydrocarbons (nitro-PAH) using both splitless and on-column injection techniques. The nitro-PAH showed good chromatographic performance and thermal stability under the GC conditions used. Retention times and response factors of several nitro-PAH were compared to those of conventional PAH.Simultaneous flame ionization and thermionic nitrogen selective detection was used to facilitate identification of nitro-PAH in complex samples. The feasibility of the method is demonstrated on two samples of commercial carbon black. Besides 1-nitropyrene several isomeric dinitropyrenes have been identified.
