    Notes: Abstract  Vertebrate Pax-6 and its Drosophila homolog eyeless play central roles in eye specification, although it is not clear if this represents the ancestral role of this gene class. As the most ”primitive” animals with true nervous systems, the Cnidaria may be informative in terms of the evolution of the Pax gene family. For this reason we surveyed the Pax gene complement of a representative of the basal cnidarian class (the Anthozoa), the coral Acropora millepora. cDNAs encoding two coral Pax proteins were isolated. Pax-Aam encoded a protein containing only a paired domain, whereas Pax-Cam also contained a homeodomain clearly related to those in the Pax-6 family. The paired domains in both proteins most resembled the vertebrate Pax-2/5/8 class, but shared several distinctive substitutions. As in most Pax-6 homologs and orthologs, an intron was present in the Pax-Cam locus at a position corresponding to residues 46/47 in the homeodomain. We propose a model for evolution of the Pax family, in which the ancestor of all of the vertebrate Pax genes most resembled Pax-6, and arose via fusion of a Pax-Aam-like gene (encoding only a paired domain) with an anteriorly-expressed homeobox gene resembling the paired -like class.

    Notes: Abstract Apolipoprotein H (apoH, protein; APOH, gene) has been implicated as a necessary cofactor for the binding of certain autoimmune antiphospholipid antibodies to anionic phospholipids. APOH exhibits genetically determined structural polymorphism with the occurrence of four alleles. Recently three IgG1k monoclonal antibodies (mAb) to human apoH, designated 3G9, 1B4, and 3D11, have been produced. The mAb 3D11 does not recognize apoH bound to anionic phospholipids in contrast to mAb 3G9 and 1B4, which recognize free and phospholipid-bound apoH. In this investigation we have determined the reactivity of the three mAb with four APOH allele products and the binding ability of these allele products with anionic phospholipids. The mAb 3G9 and 1B4, like the polyclonal anti-apoH, were equally reactive with all four allelic products, but the 3D11 recognized only the APOH *3 allele product. In the 159 APOH *3 carriers tested from five ethnic groups, the reactivity of mAb 3D11 was observed with all the Chinese but none of the African blacks. For the U.S. whites and Polynesians 89% and 75%, respectively, of the APOH *3 allele products were recognized by 3D11, while 87% of the U.S. blacks with this allele had no 3D11 reactivity. These data show that the APOH *3 allele, originally identified as a single entity by the polyclonal anti-apoH, is heterogeneous with at least one distinct variation based on mAb 3D11 reactivity. Our data also demonstrate that the apoH from certain homozygous APOH *3 individuals is unable to bind to anionic phospholipids. Such ethnic-specific apoH variations could play a significant role in the binding properties of autoimmune antiphospholipid antibodies to anionic phospholipids.

    Notes: Abstract. The sensitivity of the strength-duration (S-D) relationship to changes in the parameters describing the sodium channel of mammalian neuronal membrane was determined by computer simulation. A space-clamped patch of neuronal membrane was modeled by a parallel nonlinear sodium conductance, linear leakage conductance, and membrane capacitance. Each parameter that governs the activation (m) and inactivation (h) variables of the sodium channel was varied from −50% to +50% of its default value, and for each variation a S-D relationship was generated. Individual changes in six of the eleven parameters (α m A, α m D, α h A, β m A, β m B, and β h B) generated substantial changes in the rheobase current and chronaxie time (Tch) of the model. Changing the parameter values individually did not correct for the model's failure to generate excitation after the release from a long duration hyperpolarization (anode break excitation). Scaling a combination of five parameters (α m A, α m B, α h A, β m A, and β h B) by an equal amount produced a model that generated anode break excitation and increased Tch, but also decreased the amplitude of the action potential. To reproduce the amplitude of the action potential, the maximum sodium conductance and sodium Nernst potential were increased. These modifications generated a model that had S-D properties closer to experimental results, could produce anode break excitation, and reproduced the action potential amplitude.

    Notes: Although there has been considerable effort placed on measuring supply chains in order to assess their performance, these techniques have been found to be time and cost focused, aimed at coping with rapid change. This approach tends to have a short-term outlook. Work on greening supply chains is much longer-term in outlook. Is information intensive and biased towards the supply side? These two mindsets appear to be diverging, developing in conflicting directions. This is an alarming prospect for the environment, which has no place in future supply chain performance measurements, thus running the risk of being increasingly side-lined; and for performance measurements, which is unconcerned with longer-term sustainability in terms of the environment. The case is made to amalgamate the advantages of both schools of thought to allow long-term views to be represented by short-term performance measurement.

    Notes: β-Phenylethylamine oxidase from the Gram-positive bacterium Arthrobacter globoformis has been crystallized as three crystal forms. Two belong to space group C2 and one to space group P212121, respectively. The unit-cell volumes are consistent with one infunit of 70 644 Da per asymmetric unit for the two monoclinic forms, and with two infunits per asymmetric unit for the orthorhombic crystals. Three-dimensional intensity data have been recorded to 2.8Å resolution for one of the monoclinic crystal forms and to 3Å, resolution for the orthorhombic crystal form.

    Notes: : The Buffalo River is a tributary to the Mississippi River in west-central Wisconsin that drains a watershed dominated by agricultural land uses. Since 1935, backwater from Lock and Dam 4 on the Mississippi River has inundated the mouth of the Buffalo's valley. Resurveys of a transect first surveyed across the lake in 1935 and cesium-137 dating of backwater sediments reveal that sedimentation rates at the Buffalo's mouth have remained unchanged since the mid-1940s. Study results indicate that sediment yields from the watershed have persisted at relatively high levels over a period of several decades despite pronounced trends toward less cultivated land and major efforts to control soil erosion from agricultural land. The maintenance of sediment yields is probably due to increased channel conveyance capacities resulting from incision along some tributary streams since the early 1950s. Post-1950 incision extended the network of historical incised tributary channels, enhancing the efficient delivery of sediment from upland sources to downstream sites.
