    Description: Activated epidermal growth factor (EGF) receptors induce the formation of various complexes of intracellular signaling proteins that are mediated by SRC homology 2 (SH2) and SH3 domains. The activated receptors are also rapidly internalized into the endocytotic compartment and degraded in lysosomes. EGF stimulation of canine epithelial cells induced a rapid and transient association of the SH3-SH2-SH3 protein GRB2 with dynamin, a guanosine triphosphatase that regulates endocytosis. Disruption of GRB2 interactions by microinjection of a peptide corresponding to the GRB2 SH2 domain or its phosphopeptide ligand blocked EGF receptor endocytosis; other SH2 domains that bind EGF receptors or antibodies that neutralize RAS did not.     Description: CHOP, a member of the C/EBP family of transcription factors, mediates effects of cellular stress on growth and differentiation. It accumulates under conditions of stress and undergoes inducible phosphorylation on two adjacent serine residues (78 and 81). In vitro, CHOP is phosphorylated on these residues by p38 mitogen-activated protein kinase (MAP kinase). A specific inhibitor of p38 MAP kinase, SB203580, abolished the stress-inducible in vivo phosphorylation of CHOP. Phosphorylation of CHOP on these residues enhanced its ability to function as a transcriptional activator and was also required for the full inhibitory effect of CHOP on adipose cell differentiation.     Notes: Abstract  Hydrological records collected from water gauge stations since the 1950s demonstrate that the Taihu lake level is rising. The average rate of the lake-level rise is 0.4–1.1 mm/year during the non-flood season, resulting directly from a rise in sea level. High rates of 3.0–5.0 mm/year of rise are even recorded during the wet season. This indicates increasing human activities such as reclamation, sluicing and embanking, which significantly hinder the expulsion of extra lake water to the coast shortly after a rainfall. Generally, the lake level of the western inlet is higher than that of the eastern outlet. However, the lake-level difference between the west and east has been diminished annually from ∼10–15 cm in the 1950s to 〈3 cm at the present time. During non-flood seasons, the lake-level difference even appears to be reversed, indicating a retrogression of the lake flow from east to west. It is predicted that the Taihu drainage basin will lose much of its natural water-expelling ability in the next 50 years as the sea level continues to rise, and retrogression will likely occur during the flood season in the near future.

    Notes: Abstract Several sets of plankton and hyperbenthos samples were collected from the Seine estuary in late May-early June of 1991, 1992 and 1994, and the abundance, size-frequency, horizontal and vertical distribution, and short-term fluctuations of Pleurobrachia pileus (O.F. Müller, 1776) were determined. In the Seine estuary, P. pileus was more abundant than reported for any other European shallow waters. Its depth-averaged density was 33 individuals m-3 in 1991 and 32 individuals m-3 in 1992, with a maximum density of 808 individuals m-3. The ctenophores displayed passive tidal advection and active diel vertical migration. In the outer estuary, high abundances were observed around low tide, and diel vertical migration was very pronounced. However, many individuals remained aggregated near the bottom at all times. In spring, the distribution of P. pileus coincided with the 33 and 150‰ S isohalines, with high abundances on the marine side of the estuary. It is suggested that aggregation of ctenophores near the bottom and diel vertical migration play an important role in the retention of P. pileus in the estuary. Two schemes of P. pileus life cycle in the Bay of Seine are proposed.

    Notes: Abstract. Choline is a necessary substrate of the lipid membrane and for acetylcholine synthesis. Accumulating evidence indicates that besides being a structural component, choline is also a functional modulator of the membrane. It has been shown to be a muscarinic acetylcholine receptor (mAChR) agonist and can induce a novel K+ current in cardiac cells. However, the potential role of choline in modulating cardiac functions remained unstudied despite that mAChRs are known to be important in regulating heart functions. With microelectrode techniques, we found that choline produced concentration-dependent (0.1∼10 mm) decreases in sinus rhythm and action potential duration in isolated guinea pig atria. The effects were reversed by 2 nm 4DAMP (an M3-selective antagonist). Whole-cell patch-clamp recordings in dispersed myocytes from guinea pig and canine atria revealed that choline is able to induce a K+ current with delayed rectifying properties. The choline-induced current was suppressed by low concentrations of 4DAMP (2∼10 nm). Antagonists toward other subtypes (M1, M2 or M4) all failed to alter the current. The affinity of choline (K d ) at mAChRs derived from displacement binding of [3H]-NMS in the homogenates from dog atria was 0.9 mm, consistent with the concentration needed for the current induction and for the HR and APD modulation. Our data indicate that choline modulates the cellular electrical properties of the hearts, likely by activating a K+ current via stimulation of M3 receptors.
