    Notes: A liquid chromatography/mass spectrometry methodology is described that enables the simultaneous estimation of the individual protein-binding affinities of a mixture of compounds. This approach has proved to be robust and enables a series of molecules to be ranked according to their protein-binding affinities within 20 minutes.

    Notes: Capillary electrochromatography (CEC) was coupled with electrospray (ESI) mass spectrometry and evaluated as an on-line high resolution technique for the separation and characterization of some typical pharmaceutical compound mixtures.     Notes: Enantiomeric separation of a variety of drugs and related compounds was achieved on an (S)-naphthylethylcarbamoylated-β-cyclodextrin (S-NEC-CD) chiral stationary phase (CSP) using sub- and supercritical fluid chromatography (SFC). Compounds previously resolved on native or derivatized cyclodextrin CSPs in liquid chromatography (LC) using reversed phase or polar organic mobile phase modes could be resolved in SFC using a simple carbon dioxide/methanol eluent. Resolution of cromakalim, which is not possible on the S-NEC-CD column in LC, was readily accomplished in SFC. The importance of modifier, temperature, and pressure was assessed in relation to retention, selectivity, and resolution. The nature of the modifier and the modifier concentration were found to be crucial parameters. © 1996 Wiley-Liss, Inc. Contribution of the National Institute of Standards and Technology.     Notes: Thin films of elemental iron and nickel, plus a wide composition range of iron-nickel alloys, have been deposited by atmospheric pressure metal organic chemical vapor deposition at a growth temperature of 200°C, using iron pentacarbonyl and nickel tetracarbonyl as the source precursors. Detailed magnetoresistance measurements of the permalloy layers showed that the maximum value of magnetoresistance occurred at a composition of approximately 90 % Ni10 % Fe, and the maximum sensitivity occurred at approximately 80 % Ni20 % Fe, in accordance with literature values. In addition, both the application of an 80 mT magnetic field during deposition and the annealing of samples at temperatures higher than the growth temperature of 200°C improved the magnetic properties.

    Notes: A novel electrochemical method is described for the examination of metal-organic complexation in natural waters. The technique is based upon the comparison of square wave anodic stripping voltammetry (SWASV) ‘pseudopolarograms’ (current vs. deposition potential plots) for natural samples with those obtained for metal complexation with selected ‘model’ binding ligands at constant ionic strength. For electrochemically active zinc species in seawater, the pseudopolarogram approach allowed the estimation of thermodynamic stability constants (KML) within a stability constant window of approximately 104-106 M-1 up to ≥ 1015 M-1. For a natural rainwater sample, the pseudopolarogram indicated only one zinc class, probably a mixture of ‘free’ and/or inorganically complexed species. In an application to a coastal seawater sample, three species were identified: (1) weakly complexed, log KZnL = 4.14 ± 0.92 M-1; (2) moderately complexed, log KZnL = 7.77 ± 0.62 M-1; (3) strongly complexed, log KZnL = 11.45 ± 0.95 M-1. Results are compared to those obtained using complexometric titration with SWASV at a thin mercury film-glassy carbon rotating disk (TMF-GCRD) electrode. The latter detected a single strong complexing ligand (or ligand class), with [L′] = 15.25 ± 0.04 nM and log K′ZnL ≥ 10.9 M-1.

    Notes: The influence of aging and dietary restriction on increase in intracellular free calcium ([Ca2+]i) of CD4+ lymphocytes from Macaca mulatta was examined after stimulation with anti-CD3 mAb. We used a flow cytometric assay with the dye indo-1 and either direct or reciprocal immunofluorescent staining to dientify CD4+ cells. After stimulation with anti-CD3 mAb, intracellular free calcium responses were reduced in CD4+ lymphocytes from old male and female ad libitum fed monkeys compared to young and adult male or female monkeys. Old female monkeys had significantly lower [Ca2+]i than did old male monkeys. The reduced responses were in part related to a decreased percentage of responding cells. Dietary restrition of males over a four-year period did not alter [Ca2+]i response compared to ad libitum fed male monkeys. Female monkeys of all ages (which were restricted only for four months) also had similar [Ca2+]i responses to ad libitum fed controls. Our data suggest that age-related changes in [Ca2+]i responses are similar between humans and M. mulatta, and that over these intervals, no effects of caloric restrictions can be detected. © 1995 Wiley-Liss, Inc.

    Notes: Therapeutic drug monitoring is commonly used in both the ambulatory and hospital patient care settings. Routine measurement of concentrations of therapeutic agents in biological fluids is critical for certain drugs to maintain therapeutic benefit with minimizing drug-associated toxicities. Many analytical laboratory techniques are currently available to measure drug concentrations in biological samples. Recently there has been an increased interest in the use of capillary electrophoresis (CE) for measuring concentrations of therapeutic drugs in patient samples. However, while there are numerous reports of CE being used to measure drug concentrations in solution and pharmaceutical dosage forms, there are relatively few reports of the use of CE for measuring therapeutic agents in patient samples. The purpose of this paper is to provide an overview of methods currently used to measure therapeutic drugs in patient samples along with possible future trends for the use of CE in therapeutic drug monitoring.

    Notes: The retention behavior of polycyclic aromatic hydrocarbons (PAHs) in packed-column supercritical fluid chromatography (SFC) is studied for monomeric and polymeric C18 columns. Molecular shape discrimination (shape selectivity) is assessed through the use of Standard Reference Materials (SRMs), adn changes in selectivity are studied as a function of temperature, pressure, and mobile phase composition. Examples of separations of complex PAH isomer mixtures are presented, and guidelines are provided for modification and optimization of shape selectivity in SFC.

    Notes: Flims of metal oxides, such as Ta2O5, Nb2O5, Al2O3, HfO2, ZrO2 and TiO2 have been fabricated by use of different precursor materials, deposition techniques and annealing techniques. Several analytical methods were applied to study the layers. New data of fundamental properties of these metal oxides are reported and related to practical features that are of importance in device design and manufacturing of advanced, highly integrated devices. This overview may facilitate the choice of an optimal combination of precursor material, deposition technique and corresponding annealing procedure for a specific application of these metal oxide films in microelectronics.

    Notes: The crystal structure of the title compound, diphenyl 1-hydroxy-1-phenylethylphosphonate (1), was determined by the single-crystal X-ray diffraction method. The crystallographic data for 1 are as follow: C20H19O4P, Mr = 354.34, monoclinic, P21/n, a = 9.787(1) Å, b = 20.235(1) Å, c = 9.797(1) Å, β = 106.18(3)°, V = 1863.3(4) Å3, Z = 4, Dcalc = 1.26 g/cm3, λ(Mo-Ka) = 0.71073 Å, μ = 1.6 cm-1, F(000) = 744, R = 0.018, and Rw = 0.032 for 2258 observed reflections. The solid-state structure in a dimeric packing mode exhibits intermolecular hydrogen bonding of the type P = O···H-O. Infrared solution studies (CCl4) indicate that upon high dilution (10-4 M) the dimers completely dissociate to give conformers with and without intramolecular hydrogen bonds. Theoretical studies (PM3) were undertaken to determine the energy profile about the P-C torsional angle, which exhibited low energy barriers to rotation with no clear minimum energy conformation. © 1996 John Wiley & Sons, Inc.
