    Notes: Abstract  The product of the Wilms' tumor gene WT1 is a transcription factor overexpressed not only in leukemic blast cells of almost all patients with acute myeloid leukemia, acute lymphoid leukemia, and chronic myeloid leukemia, but also in various types of solid tumor cells. Thus, it is suggested that the WT1 gene plays an important role in both leukemogenesis and tumorigenesis. Here we tested the potential of WT1 to serve as a target for immunotherapy against leukemia and solid tumors. Four 9-mer WT1 peptides that contain HLA-A2.1-binding anchor motifs were synthesized. Two of them, Db126 and WH187, were determined to bind to HLA-A2.1 molecules in a binding assay using transporter associated with antigen processing-deficient T2 cells. Peripheral blood mononuclear cells from an HLA-A2.1-positive healthy donor were repeatedly sensitized in vitro with T2 cells pulsed with each of these two WT1 peptides, and CD8+ cytotoxic T lymphocytes (CTLs) that specifically lyse WT1 peptide-pulsed T2 cells in an HLA-A2.1-restricted fashion were induced. The CTLs also exerted specific lysis against WT1-expressing, HLA-A2.1-positive leukemia cells, but not against WT1-expressing, HLA-A2.1-negative leukemia cells, or WT1-nonexpressing, HLA-A2.1-positive B-lymphoblastoid cells. These data provide the first evidence of human CTL responses specific for the WT1 peptides, and provide a rationale for developing WT1 peptide-based adoptive T-cell therapy and vaccination against leukemia and solid tumors.

    Notes: Abstract  The decline of Japanese red pine trees (Pinus densiflora Sieb. et Zucc.) at Mt. Gokurakuji (693 m a.s.l.), 30 km west of Hiroshima city, west Japan, was studied. The effects of air pollution and acid deposition on the physiological characteristics of the trees, especially those of the needles, were investigated. Ozone concentration was not correlated with the physiological status of the needles and SO2 concentration was not high in the declined area. NO2 concentration correlated negatively with needle longevity while it correlated positively with ethylene emission from 1-year-old needles. Average needle longevity was about 2.8 years in non-declined areas; however the longevity was 1.3 years in the most polluted area. The minimal fluorescence at night (F 0)of 1-year-old needles decreased with increasing NO2 concentration. The maximum stomatal conductance (gl), net photosynthesis (P n)and intercellular CO2 concentration (C i) in the declined areas were lower than in the non- declined areas (about 50%, 30% and 20% lower, respectively). The lower C isuggested that the major part of the decrease in P ncan be explained by stomatal restriction. The soil pH, N content and C/N ratio showed no significant difference between the declined and non-declined areas. The physiological disorders of needles were due to the damage by air pollutants, and important roles of NO2 are suggested. Lowering of P n and the shortening of needle longevity appear to be the main causes of the decline in pines in the forest decline area.

    Notes: The morphological change of p-type porous silicon during annealing has been investigated. The x-ray diffraction (XRD) pattern was composed of a sharp Bragg reflection peak and a diffuse scattering. The diffuse scattering is not related to the presence of the amorphous phase. The shape of the XRD pattern started to change at an annealing temperature as low as 400 °C, and the 2θ angle of the sharp peak varied at a temperature as low as 350 °C. These changes at low temperatures seem to be closely related to the desorption of hydrogen and the resultant change of the dangling bond density in porous silicon. The molecular orbital calculations also support the participation of dangling bonds in the structural reorganization in the surface region. © 2001 American Institute of Physics.

    Notes: Abstract  The manned submersible Shinkai 2000 investigated yellow patches on the near-summit slope of Shiribeshi Seamount in the Japan Sea. Yellowish patches are often associated with seepage, and the possibility of seepage at Shiribeshi Seamount was tested by the following four lines of observation: (i) high subsurface temperature was measured at a ring-like patch, although no increase in subsurface temperature was observed at other patches; (ii) high gamma ray (γ-ray) intensity from the thorium series was recorded in the patch zone; (iii) the yellowish deposit was composed of calcite, quartz and amorphous iron compound, as seen at the yellowish patches in other seeps and volcanoes; (iv) lipid phosphate, a measure of microbial abundance, in sediments of the ring-like patch was determined, and the recorded microbial abundance was higher inside the patch than outside it. The four lines of observation are explained consistently by postulating that the seepage of warm fluid contained Fe and γ-ray sources. A hydrothermal origin of the yellow patches is not ruled out for the extinct but young (0.9 Ma) arc volcano.

    Notes: Silk fibroin is a simple protein expected to have functional applications in medicine and bioelectronics. The primary structure of this protein is quite simple, and the main secondary structures are β-sheet crystals and amorphous random coils. In the present study, we investigated pulsed laser deposition (PLD) of fibroin with the β-sheet structures as targets. The primary and secondary structures in films deposited were analyzed using infrared spectroscopy. Normal laser deposition at 351 nm using neat fibroin targets produced thin films of fibroin with a random coiled structure. Ablation was triggered by two-photonic excitation of the peptide chains, which resulted in the destruction of β-sheet structure in PLD. In order to avoid the two-photonic excitation, we adopted a PLD method utilizing anthracene (5–0.1 wt %) in a photosensitized reaction involving doped fibroin targets. Laser light (351 or 355 nm) was absorbed only by anthracene, which plays an important role converting photon energy to thermal energy with great ablation efficiency. Thin fibroin films deposited by this method had both random coil and β-sheet structures. As the dopant concentration and laser fluence decreased, the ratio of β-sheet domain to random coil increased in thin deposited films. © 2001 American Institute of Physics.

    Notes: An adaptive mesh generation method for two- and three-dimensional finite element methods using edge elements is proposed. Since the tangential component continuity is preserved when using edge elements, the strategy of creating new nodes is based on evaluation of the normal component of the magnetic vector potential across element interfaces. The evaluation is performed at the middle point of edge of a triangular element for two-dimensional problems or at the gravity center of triangular surface of a tetrahedral element for three-dimensional problems. At the boundary of two elements, the error estimator is the ratio of the normal component discontinuity to the maximum value of the potential in the same material. One or more nodes are set at the middle points of the edges according to the value of the estimator as well as the subdivision of elements where new nodes have been created. A final mesh will be obtained after several iterations. Some computation results of two- and three-dimensional problems using the proposed method are shown. © 2001 American Institute of Physics.

    Notes: The first gene characterizing the clag (cytoadherence linked asexual gene) family of Plasmodium falciparum was identified on chromosome 9. The protein product (Clag9) was implicated in cytoadhesion, the binding of infected erythrocytes to host endothelial cells, but little information on the biochemical characteristics of this protein is available. Other genes related to clag9 have been identified on different chromosomes. These genes encode similar amino acid sequences, but clag9 shows least conservation. Clag9 was detected in schizonts, merozoites and ring-stage parasites after protease digestion and peptide analysis by mass spectrometry. Using antisera raised against unique regions of Clag9 and against RhopH2, a component of the RhopH high-molecular-mass protein complex of merozoites, immunofluorescence co-localized the two proteins to the apical region of merozoites. Immunoelectron microscopy co-localized Clag9 and RhopH2 exclusively to the basal bulb region of rhoptries rather than to their apical ducts. The same Clag9-specific antibodies bound the RhopH complex, and the protein was detected in the complex purified by antibodies to RhopH2. Clag9 protein was also shown to be present in ring-stage parasites, carried through from the previous cycle with the RhopH complex, in a location identical to that of RhopH2. Transcription of the clag9 gene was shown to occur at the same time as the genes for other members of the RhopH complex, rhoph2 and 3. The results indicate that Clag9 is part of the RhopH complex and suggest that, within this complex, the protein previously designated RhopH1 is composed of more than one protein product of the clag gene family. The results cast doubt on a direct role for Clag9 in cytoadhesion; we suggest that the primary role of the RhopH complex is in remodelling the infected red blood cell after invasion by the merozoite. The complex may have multiple functions dependent on its exact composition, which may include, with respect to Clag9, a contribution to the mechanism of cytoadhesion.

    Notes: The Re atom in [Re(C9H6N2)Cl3(C18H15P)] is octahedrally coordinated by three Cl atoms in facial positions, two N atoms from 8-imidoquinoline (imq) and one P atom from triphenylphosphine. The Re—N(imido) distance [1.760 (9) and 1.772 (8) Å] for imq is very short and implies double-bond character. The trans influence of the P atom is indicated. Intra- and intermolecular π–π interactions between the π-rings in the complex are also observed.
