    Notes: Abstract  Miniaturized interference filters were designed and fabricated using two different manufacturing technologies. Applying micromachined ceramic masks during the coating processes interference filters with 1 mm lateral feature size and an alignment accuracy of 50 μm were arranged in an array consisting of three different filters. The filter edge definition obtained by this method was smaller than 50 μm. By applying ion assisted deposition (IAD), a low-temperature coating process, the spectral sensitivity of receiver cells has been modified by coating the cells directly. A combination of coating processes, microlithographic masking procedures, and dry etching technologies made it possible to arrange three different stripe filters with minimum filter features of about 5 μm side by side. The accuracy during mask alignment and the filter edge definition was also within 1 and 2 μm, respectively.

    Notes: Attracting, motivating and retaining knowledge workers have become important in a knowledge-based and tight labour market, where changing knowledge management practices and global convergence of technology has redefined the nature of work. While individualisation of employment practices and team-based work may provide personal and organisational flexibilities, aligning HR and organisational strategies for competitive advantage has become more prominent. This exploratory study identifies the most and least effective HR strategies used by knowledge intensive firms (KIFs) in Singapore for attracting, motivating and retaining these workers. The most popular strategies were not always the most effective, and there appear to be distinctive ‘bundles’ of HR practices for managing knowledge workers. These vary according to whether ownership is foreign or local. A schema, based on statistically significant findings, for improving the effectiveness of these practices in managing knowledge workers is proposed. Cross-cultural research is necessary to establish the extent of diffusion of these practices.

    Notes: Abstract The three-dimensional structures of tryptophan synthase, carbamoyl phosphate synthetase, glutamine phosphoribosylpyrophosphate amidotransferase, and asparagine synthetase have revealed the relative locations of multiple active sites within these proteins. In all of these polyfunctional enzymes, a product formed from the catalytic reaction at one active site is a substrate for an enzymatic reaction at a distal active site. Reaction intermediates are translocated from one active site to the next through the participation of an intermolecular tunnel. The tunnel in tryptophan synthase is ~25 A in length, whereas the tunnel in carbamoyl phosphate synthetase is nearly 100 A long. Kinetic studies have demonstrated that the individual reactions are coordinated through allosteric coupling of one active site with another. The participation of these molecular tunnels is thought to protect reactive intermediates from coming in contact with the external medium.

    Notes: Magnetohydrodynamics (MHD) offers a unique opportunity to study the behavior of two-dimensional turbulent flows. A strong external magnetic field B perpendicular to the flow direction of an electrically conducting fluid will suppress velocity gradients in the direction of B. The resulting approximation is known as quasi-two-dimensional MHD. An experimental configuration is presented which meets this requirement, along with a spatially extended probe used to visualize the two-dimensional flow kinematics inside the opaque liquid metal flow. As a prototypical example, the wake behind a circular cylinder is investigated for Reynolds numbers up to R=10 000. New and unexpected vortex patterns are observed that deviate significantly from usual hydrodynamic flows. Also, stability limits for the transition from stationary to nonstationary flow patterns are experimentally determined for the cylinder wake and another type of shear flow profile. These results confirm existing theoretical predictions and thus validate the quasi-two-dimensional approach. © 2001 American Institute of Physics.

    Notes: The effect of supplemental UV-A (320–400 nm) radiation on tissue absorption at 355 nm, levels of various antioxidants (ascorbate, glutathione, carotenoids and flavonoids) and of antioxidant scavenging capacity were investigated with leaves and petals of Rosa hybrida, cv. Honesty and with leaves, petals and sepals of Fuchsia hybrida, cv. Dollarprinzessin. Supplemental UV-A did not result in visible changes in plant morphology of either species. In leaves it induced small increases in levels of chlorophylls a and b, the carotenoids antheraxanthin, lutein and β-carotene, and high increases in the flavonols quercetin and kaempferol. Petals hardly responded, while the coloured sepals of fuchsia showed an increase in quercetin derivatives. HPLC of unhydrolysed flavonoids showed that individual quercetin derivatives in leaves of both species and kaempferol derivatives in rose leaves increased 2-fold. Some kaempferol derivatives in fuchsia leaves were more than 2-fold enhanced or were newly induced by supplemental UV-A. Increases in l-ascorbic acid levels in fuchsia leaves, and decreases in rose leaves as result of supplemental UV-A were observed, but differences appeared statistically not significant, while l-ascorbate levels remained unchanged in the other tissues investigated. Anthocyanins and reduced glutathione levels were unaffected in all tissues. The combined UV-A induced increases in concentrations of these antioxidant species, did not lead to significant increases in antioxidant capacity of tissues, measured as Trolox equivalents in 50%-ethanol extracts. Light absorption at 355 nm of leaf extracts was significantly increased upon UV-A exposure. Our results indicate that the major protection towards UV-A exposure, in particular in the leaves, will originate from absorption of irradiation, and not from scavenging reactive oxygen species.

    Notes: Glycolysis is one of the main pathways of carbon catabolism in Bacillus subtilis. Although the biochemical activity of glycolytic enzymes has been studied in detail, no information about the expression of glycolytic genes has so far been available in this organism. Therefore, transcriptional analysis of all glycolytic genes was performed. The genes cggR, gapA, pgk, tpi, pgm and eno, encoding the enzymes required for the interconversion of triose phosphates, are transcribed as a hexacistronic operon as demonstrated by Northern analysis. This gapA operon is repressed by the regulator CggR. The presence of sugars and amino acids synergistically results in the induction of the gapA operon. The transcriptional start site upstream of cggR was mapped by primer extension. Transcripts originating upstream of cggR are processed near the 3′ end of cggR. This endonucleolytic cleavage leads to differential stability of the resulting processing products: the monocistronic cggR message is very rapidly degraded, whereas the mRNA species encoding glycolytic enzymes exhibit much higher stability. An additional internal constitutive promoter was identified upstream of pgk. Thus, gapA is the most strongly regulated gene of this operon. The pfk pyk operon encoding phosphofructokinase and pyruvate kinase is weakly induced by glucose. In contrast, the genes pgi and fbaA, coding for phosphoglucoisomerase and fructose-1,6-bisphosphate aldolase, are constitutively expressed.

    Notes: Enterococcus faecium strain 9631355 was isolated from animal sources on the basis of its resistance to the growth promotant avilamycin. The strain also exhibited high-level resistance to evernimicin, a drug undergoing evaluation as a therapeutic agent in humans. Ribosomes from strain 9631355 exhibited a dramatic reduction in evernimicin binding, shown by both cell-free translation assays and direct-binding assays. The resistance determinant was cloned from strain 9631355; sequence alignments suggested it was a methyltransferase and therefore it was designated emtA for evernimicin methyltransferase. Evernimicin resistance was transmissible and emtA was localized to a plasmid-borne insertion element. Purified EmtA methylated 50S subunits from an evernimicin-sensitive strain 30-fold more efficiently than those from a resistant strain. Reverse transcription identified a pause site that was unique to the 23S rRNA extracted from resistant ribosomes. The pause corresponded to methylation of residue G2470 (Escherichia coli numbering). RNA footprinting revealed that G2470 is located within the evernimicin-binding site on the ribosome, thus providing an explanation for the reduced binding of the drug to methylated ribosomes.

    Notes: Abstract A needle-like apatite-leucite glass-ceramic was prepared in the SiO2-Al2O3-Na2O-K2O-P2O5-F system. Nucleation and crystallization processes were studied in bulk and powdered samples. The crystallization of leucite follows the mechanism of surface crystallization. After the precipitation of NaCaPO4 crystals and another unknown crystal phase, the formation of needle-like apatite is based on a volume nucleation and crystallization process. The mechanism of the formation of needle-like apatite differs to those of apatite precipitation in glass-ceramics. The morphology of needle-like apatite is comparable to that of apatite in natural teeth. The properties of the glass-ceramic, especially the good chemical durability, the optical properties, as well as mechanical and thermal properties allow glass-ceramic to be used as a main component in a bio-material for the veneering of metal restorations in dentistry.
