    Notes: Abstract. Tapasin is a M r 48,000 glycoprotein and has a specialized role in MHC class I-restricted antigen presentation. It is encoded by a gene which maps centromeric to the MHC class II region of human Chromosome 6 within 200 kb of HLA-DP. There is variable dependence upon tapasin for MHC class I expression among different MHC class I alleles. HLA-B*4402 and to a lesser extent HLA-A1 and B8 are tapasin dependent, whereas HLA-B27, A2 and to a lesser extent B7 and A3 are tapasin independent. We investigated whether tapasin is polymorphic and whether these Tapasin alleles are in linkage with any MHC class I alleles. We identified three new mutations within intron 4, which are in a particular linkage with the previously described exon 4 (G16003C) dimorphism. The intronic mutations are G16146T, G16232A, and T16317A (numbering according to cosmid clone F0811; GenBank accession number Z97184). The allele frequency of Tapasin*01 (G16003) was 0.47 and Tapasin*02 (C16003) was 0.53 in this UK population. Four of the eight possible intronic haplotypes were identified and their cis linkage with the tapasin dimorphism ascertained. Tapasin*01 was associated with all the identified haplotypes, while Tapasin*02 was only associated with the wild-type intronic sequence (GGT). There was no significant linkage (P〉0.01) of the Tapasin dimorphism or new Tapasin alleles to any of the MHC class I A, B, or C alleles studied or to the extended A1 B8 DR3 haplotype.

    Notes: The ground state rotational spectra and internal dynamics in five isotopomers 14N2–IH, 14N2–ID, 15N2–IH, 14N15N–IH, and 15N14N–IH of the nitrogen–hydrogen iodine dimer have been investigated by pulsed-nozzle, Fourier-transform microwave spectroscopy. Analysis of the recorded spectra yields rotational, centrifugal distortion, nuclear quadrupole and spin-rotation coupling constants for the five isotopomers. The spectroscopic constants are interpreted in terms of a ground-state NN–IH structure using the parameters rcm, kσ, and the oscillation angles θN2 and θHI of the N2 and IH moieties. For 14N2–IH, the values rcm=4.197 872(67) Å, kσ=1.461 52(32) N/m, θN2=25.61(22)°, and θHI=23.454 6(91)° are obtained. The rotational transitions of 14N2–IH, 14N2–ID, and 15N2–IH show a doubling, which is a consequence of tunneling associated with the interchange of the two nitrogen nuclei. The experimental findings are compared with results from ab initio calculations. © 2000 American Institute of Physics.

    Notes: A fully three-dimensional morphed potential energy surface is reported for Ar:HBr. The morphed potential was obtained from parametrized scaling and shifting transformations of an ab initio potential. The optimum parameters of the morphed potential were determined by a regularized nonlinear least-squares fit to available experimental data. The rovibrational dynamics of the complex were computed using an adiabatic separation of the H–Br intramolecular stretching mode from the intermolecular modes of the system. The ground rovibrational state of the morphed potential was found to have the hydrogen-bound structure Ar–HBr. This state was 10.99 cm−1 more stable than the corresponding state having the van der Waals structure, Ar–BrH, in agreement with experimental data. However, the global minimum of the morphed potential was found to have the van der Waals structure, Ar–BrH. This structure was 20.9 cm−1 lower in energy than the local minimum having the hydrogen-bound structure, Ar–HBr. © 2001 American Institute of Physics.

    Notes: Total internal reflection microscopy is a technique for monitoring changes in the distance between a single microscopic sphere and a flat plate by measuring the intensity of light scattered by the sphere when illuminated by an evanescent wave. A histogram of scattering intensities can be used to construct the potential energy profile as a function of distance relative to the most probable distance. Thus potential energies can be measured to within a fraction of kT while changes in distance can be measured to within 1 nm. An autocorrelation of the scattering intensities can be used to deduce an average diffusion coefficient of the sphere, which is found to be only a few percent of the Stokes–Einstein value, owing to the close proximity of the plate. The analysis of the intensity-autocorrelation function presented here can be used to deduce an absolute value for the most probable separation distance, without a priori knowledge of the functional form of the PE profile and in the presence of a constant background scattering intensity. This "hydrodynamic" separation distance is found to be within a few percent of the "optical" separation distance found independently by comparing the intensity at the most probable distance with the intensity of the same particle in contact with the plate. Since the particle does not need to be brought into contact with the plate, the hydrodynamic method is well suited for determining the absolute separation distance with deformable particles like liquid droplets, vesicles or biological cells. Moreover, the hydrodynamic separation can be immediately calculated without any additional experiments. However, accurate determination of the hydrodynamic separation requires an accurate value for the particle size, which must be determined independently. © 2000 American Institute of Physics.

    Notes: The UK retail environment is extremely competitive, forcing retailers to constantly develop and enhance both their offerings and their formats. Probably the most significant changes taking place are coming from not the products being sold themselves, but rather when, how and where they are being sold. For example, new technologies such as the Internet, CD-Roms, electronic kiosks and digital television are opening up new opportunities for retailers through remote purchasing and delivery. While a number of large retailers have made heavy investments into electronic retailing, such as Arcadia, Marks & Spencer, Tesco and Sainsbury, it is uncertain how they are going to use this technology to gain competitive or market advantages. Given the media attention which technologies such as the Internet has attracted, there is the danger that businesses may adopt an unfocussed use. This report identifies and evaluates the ways in which major UK grocery retailers are using the Internet to create value for their users over and above that which is offered by bricks and mortar stores. Web sites of three grocery retailers were evaluated via preselected criteria in order to assess how they created value for their customers. Applying Holbrook’s typology of consumer value, the findings of the research suggest that each of the retailers surveyed are similar in terms of the nature of the value they have created for consumers online, these being value-as-convenience and value-as excellence.

    Notes: Comparative analysis of 70 16S rDNA sequences representing 20 Rhizobium species (including pathogenic Agrobacterium spp.) was conducted using Maximum Likelihood to establish relationships of species using multiple sequences. There is no significant internal division of the Rhizobium clade to suggest that it represents more than one genus. Plant pathogenic (Agrobacterium) species are distributed within the genus. The analysis supported the synonymy of some species (Rhizobium gallicum and Rhizobium mongolense) and the need for comparative investigations of the tumorigenic and nodulating properties of Rhizobium tropici and Rhizobium rhizogenes. Misidentification of some sequences may conceal one or more putative novel species. Some sequences appear to be misidentified because of faulty sequencing or incomplete or inadequate analysis.
