    Notizen: Abstract Agents (hardware or software) that act autonomously in an environment have to be able to integrate three basic behaviors: planning, execution, and learning. This integration is mandatory when the agent has no knowledge about how its actions can affect the environment, how the environment reacts to its actions, or, when the agent does not receive as an explicit input, the goals it must achieve. Without an “a priori” theory, autonomous agents should be able to self-propose goals, set-up plans for achieving the goals according to previously learned models of the agent and the environment, and learn those models from past experiences of successful and failed executions of plans. Planning involves selecting a goal to reach and computing a set of actions that will allow the autonomous agent to achieve the goal. Execution deals with the interaction with the environment by application of planned actions, observation of resulting perceptions, and control of successful achievement of the goals. Learning is needed to predict the reactions of the environment to the agent actions, thus guiding the agent to achieve its goals more efficiently. In this context, most of the learning systems applied to problem solving have been used to learn control knowledge for guiding the search for a plan, but few systems have focused on the acquisition of planning operator descriptions. As an example, currently, one of the most used techniques for the integration of (a way of) planning, execution, and learning is reinforcement learning. However, they usually do not consider the representation of action descriptions, so they cannot reason in terms of goals and ways of achieving those goals. In this paper, we present an integrated architecture, lope, that learns operator definitions, plans using those operators, and executes the plans for modifying the acquired operators. The resulting system is domain-independent, and we have performed experiments in a robotic framework. The results clearly show that the integrated planning, learning, and executing system outperforms the basic planner in that domain.

    Notizen: Abstract The effect of LAB 198 999 [3,5-dioxo-4-butyryl-cyclohexane carboxylic acid ethyl ester; a new plant growth retardant which competitively inhibits 2-oxoglutarate-dependent gibberellin (GA) dioxygenases] on elongation and in-vivo [3H]GA1 and [3H]GA20 metabolism in cowpea (Vigna sinensis L. cv Blackeye pea No. 5) epicotyls has been investigated. Gibberellins and LAB 198 999 were injected into the epicotyl at 25–30 mm from the apex. In intact seedlings, epicotyl elongation was inhibited by LAB 198 999 (25 μg · epicotyl-1), and the inhibition was counteracted by GA1 but not by GA20. In contrast to intact seedlings, the inhibitor enhanced epicotyl elongation in de-bladed seedlings and expiants, in the latter case proportionally to the amount of inhibitor applied (up to 50 μg · epicotyl-1), but not in explants made from paclobutrazol-treated seedlings. The inhibitor also enhanced dramatically the elongation induced in paclobutrazol-treated expiants by GA1, but not by GA20. The promotive effect of LAB 198 999 was associated with increased contents of GA1 and GA8 in the growing region of the epicotyl, indicating a dependence on endogenous GAs. The effect of LAB 198999 decreased progressively with the age of the seedlings, probably as a consequence of a decreased level of GAs in the epicotyl. Gibberellin substrates and metabolites in the growing region of the epicotyl (upper 20 mm) were fractionated and identified tentatively by high-performance liquid chromatography and radiocounting using a homogeneous on-line radioactivity detector. The metabolism of [3H]GA1(t) (tentative) to [3H]GA8(t), and that of [3H]GA20(t) to [3H]GA1(t) and [3H]GA29(t) in the epicotyl were blocked by LAB 198 999, that of the former more efficiently than the latter. The results presented support the hypothesis that GA1 is the active GA controlling elongation of cowpea epicotyls. They also show that both the promotion of epicotyl elongation in explants and the enhancement of the effect of exogenous GA1 by LAB 198 999 are the result of the inhibitor blocking the in-vivo 2β-hydroxylation of GA1.

    Notizen: Abstract Acute treatment of rats with recombinant tumour necrosis factor (TNF-α) caused an enhanced proteolytic rate —measured as tyrosine released in the presence of cycloheximide — insoleus muscle (34%). The cytokine treatment also decreased the rate of protein synthesis in this muscle (22%) while it had no effect upon the same parameter inextensor digitorum longus (EDL) (26%) muscle. In addition, treatment of rats with TNF-α increased amino acid uptake by transport system A in the incubated muscles both insoleus (45%) andEDL (99%) in the presence of insulin in the incubating medium. This effect was not associated with a direct action of TNF on muscle since the addition of different concentrations of the cytokine to the preparations did not alter the uptake of α-(methyl)-aminoisobutyric acid by the incubated muscles. It can be concluded that acute TNF-α treatment causes changes in protein metabolism in red-type muscles — suchsoleus — while little effects are seen in white-type muscles — such as EDL. The results presented may, to some extent, be related to the cachectic response associated with cancer and inflammation.

    Notizen: Abstract In addition to the previously-reported gibberellins: GA1; GA8, GA20 and GA29 (García-Martínez et al., 1987, Planta 170, 130–137), GA3 and GA19 were identified by combined gas chromatography-mass spectrometry in pods and ovules of 4-d-old pollinated pea (Pisum sativum cv. Alaska) ovaries. Pods contained additionally GA17, GA81 (2α-hydroxy GA20) and GA29-catabolite. The concentrations of GA1, GA3, GA8, GA19, GA20 and GA29 were higher in the ovules than in the pod, although, with the exception of GA3, the total content of these GAs in the pod exceeded that in the seeds. About 80% of the GA3 content of the ovary was present in the seeds. The concentrations of GA19 and GA20 in pollinated ovaries remained fairly constant for the first 12 ds after an thesis, after which they increased sharply. In contrast, GA1 and GA3 concentrations were maximal at 7 d and 4–6 d, respectively, after anthesis, at about the time of maximum pod growth rate, and declined thereafter. Emasculated ovaries at anthesis contained GA8, GA19 and GA20 at concentrations comparable with pollinated fruit, but they decreased rapidly. Gibberellins a1 and A3 were present in only trace amounts in emasculated ovaries at any stage. Parthenocarpic fruit, produced by decapitating plants immediately above an emasculated flower, or by treating such flowers with 2,4-dichlorophenoxyacetic acid or GA7, contained GA19 and GA20 at similar concentrations to seeded fruit, but very low amounts of GA1 and GA3 Thus, it appears that the presence of fertilised ovules is necessary for the synthesis of these last two GAs. Mature leaves and leaf diffusates contained GA1, GA8, GA19 and GA20 as determined by combined gas chromatography-mass spectrometry using selected ion monitoring. This provides further evidence that vegetative tissues are a possible alternative source of GAs for fruit-set, particularly in decapitated plants.

    Notizen: Summary The development of the semilunar valves takes place in association with septation of the outflow tract in the embryonic heart. Although numerous studies have focused on this process, the causal mechanisms of valvular development remain obscure. This paper reports an immunohistochemical analysis of tenascin and type-I collagen distribution in developing chick heart valves. Tenascin is a glycoprotein that is present on some embryonic extracellular matrices. It plays several significant roles in tissue differentiation, cell growth, and tissue interactions; it is also important for the formation of specific zones of connective tissue that fulfill mechanical functions. Our results show that tenascin is present during valvular morphogenesis and histogenesis, and that its distribution is associated with zones specialized in bearing mechanical loads.

    Notizen: Abstract The response of unpollinated ovary explants ofPisum sativum L. cv. Alaska No. 7 to several plant growth regulators and nutrients has been studied. Explants consisted of a segment of stem and an emasculated flower with or without the adjacent leaf. They were made on the day equivalent to anthesis and were cultured in a liquid medium. Growth regulators were applied either in the solution or directly to the ovaries. Giberellic acid (GA3) in the presence of sucrose, but not indole-3-acetic acid or N6-(Δ2-isopentenyl)-adenine (2iP), induced fruit set and development of parthenocarpic fruits, the final length of these being a function of the intensity of the GA3 treatment. The capacity of ovaries to respond fully to GA3 was not lost after incubation of explants in water or 50 mM sucrose for 1 day and was similar in explants made between the day of anthesis and 3 days later. Limited growth was obtained with 100 mM sucrose alone but this effect was counteracted by 2′-isopropyl-4′-(trimethyl ammonium chloride)-5′-methylphenyl piperidine-1-carboxylate (AMO-1618). This inhibitor was ineffective when GA3 was applied to the ovary. The development of the fruit was proportional to the length of the segment of stem up to 5 cm. The presence of the leaf in the explant enhanced the development of the fruit. These results indicate that a gibberellin is necessary for setting and development of fruits from cultured ovaries and that this effect depends on an appropriate source of nutrients. The course of development of parthenocarpic fruits on explants was similar to that of seeded fruits on the intact plant. The cultured pea ovary systemoffers convenient means to investigate the role of gibberellins and nutrients in fruit set and development.
