    Beschreibung: Seawater pH lowering, known as ocean acidification, is considered among the major threats to marine environment. In this study, post-spawning adults of the sea urchin Paracentrotus lividus were maintained at three pH values (8.0, 7.7, 7.4) for 60 days. Physiological, biochemical, cellular, behavioural and reproductive responses were evaluated in males and females. Significant differences between sexes were observed, with higher ammonia excretion and lower catalase activity in males. Respiration rate (after 21 days), catalase activity in gonads and total coelomocyte count showed the same increasing trend in males and females under low pH. Ammonia excretion, gonadosomatic index and lysozyme activity exhibited opposite responses to low pH, with an increasing trend in males and decreasing in females. Results demonstrated that exposure to low pH could result in different response strategies of male and female sea urchins at a physiological, biochemical and immunological level. Reduced female gonadosomatic index under low pH suggested decreased energy investment in reproduction.

    Beschreibung: CO2-driven ocean acidification affects many aspects of sea urchin biology. However, even in the same species, OA effects are often not univocal due to non-uniform exposure setups or different ecological history of the experimental specimens. In the present work, two groups of adult sea urchins Paracentrotus lividus from different environments (the Lagoon of Venice and a coastal area in the Northern Adriatic Sea) were exposed to OA in a long-term exposure. Animals were maintained for six months in both natural seawater (pHT 8.04) and end-of-the-century predicted condition (-0.4 units pH). Monthly, physiological (respiration rate, ammonia excretion, O:N ratio) and behavioural (righting, sheltering) endpoints were investigated. Both pH and time of exposure significantly influenced sea urchin responses, but differences between sites were highlighted, particularly in the first months. Under reduced pH, ammonia excretion increased and O:N decreased in coastal specimens. Righting and sheltering were impaired in coastal animals, whereas only righting decreased in lagoon ones. These findings suggested a higher adaptation ability in sea urchins from a more variable environment. Interestingly, as the exposure continued, animals from both sites were able to acclimate. Results revealed plasticity in the physiological and behavioural responses of sea urchins under future predicted OA conditions.

    Beschreibung: Whereas low levels of thermal stress, irradiance, and dietary restriction can have beneficial effects for many taxa, stress acclimation remains understudied in marine invertebrates, despite being threatened by climate change stressors such as ocean acidification. To test for life-stage and stress-intensity dependence in eliciting enhanced tolerance under subsequent stress encounters, we initially conditioned pediveliger Pacific geoduck (Panopea generosa) larvae to (i) ambient and moderately elevated pCO2 (920 µatm and 2800 µatm, respectively) for 110 days, (ii) secondarily applied a 7-day exposure to ambient, moderate, and severely elevated pCO2 (750 µatm, 2800 µatm, and 4900 µatm, respectively), followed by 7 days in ambient conditions, and (iii) implemented a 7-day third exposure to ambient (970 µatm) and moderate pCO2 (3000 µatm). Initial conditioning to moderate pCO2 stress followed by second and third exposure to severe and moderate pCO2 stress increased respiration rate, organic biomass, and shell size suggesting a stress-intensity-dependent effect on energetics. Additionally, stress-acclimated clams had lower antioxidant capacity compared to clams under ambient conditions, supporting the hypothesis that stress over postlarval-to-juvenile development affects oxidative status later in life. Time series and stress intensity-specific approaches can reveal life-stages and magnitudes of exposure, respectively, that may elicit beneficial phenotypic variation.

    Beschreibung: This volume considers the Prabodhacandrodaya Nāṭaka (c. 1760 CE), an allegorical drama composed by Brajvāsīdās in Brajbhāṣā. It contributes to the study of vernacular nāṭakas with its first complete English translation. Moreover, the critical analysis shows that the foundational Sanskrit texts for Vedānta and those for Bhakti play a part in the Prabodhacandrodaya Nāṭaka’s philosophical and religious edifice. At the same time, the investigation demonstrates that Brajvāsīdās expresses several philosophical ideas by adaptively reusing the Rāmcaritmānas by Tulsīdās (c. 1574 CE). Brajvāsīdās composes a dohā by combining one line of his invention with a line from the Mānas. This method is employed throughout all the personified metaphysical concepts. That Brajvāsī not only read Bhakti but also Vedānta through the Rāmcaritmānas highlights the philosophical and literary creativity in 18th c. North India. It points to the necessity to rethink the sources of Vedānta philosophies, by including works non-conventional for language and genre, because not in Sanskrit and not śāstras.     Beschreibung: Long-term observations and experiments in diverse drylands reveal how ecosystems and services are responding to climate change. To develop generalities about climate change impacts at dryland sites, we compared broadscale patterns in climate and synthesized primary production responses among the eight terrestrial, nonforested sites of the United States Long-Term Ecological Research (US LTER) Network located in temperate (Southwest and Midwest) and polar (Arctic and Antarctic) regions. All sites experienced warming in recent decades, whereas drought varied regionally with multidecadal phases. Multiple years of wet or dry conditions had larger effects than single years on primary production. Droughts, floods, and wildfires altered resource availability and restructured plant communities, with greater impacts on primary production than warming alone. During severe regional droughts, air pollution from wildfire and dust events peaked. Studies at US LTER drylands over more than 40 years demonstrate reciprocal links and feedbacks among dryland ecosystems, climate-driven disturbance events, and climate change.

    Notizen: The plant uncoupling mitochondrial protein (PUMP) and the plant mitochondrial potassium channel (PmitoKATP) are two recently discovered energy-dissipating systems present in plant mitochondria, which may play a role as defence systems under environmental stress. To verify whether hyperosmotic stress affects the two dissipating systems in durum wheat (Triticum durum Desf.), their functioning was studied in early etiolated seedlings maintained under moderate and severe salt (NaCl) and osmotic (mannitol) stress. As measures of mitochondrial stress mitochondrial integrity, membrane potential maintenance and oxygen uptake coupled with ATP synthesis during succinate and proline oxidation were investigated. Both PUMP and PmitoKATP were activated under stress conditions. Activation was clearly evident even under moderate stress when proline oxidation was inhibited, although mitochondrial integrity and succinate oxidation were still unaffected. Under severe stress, which significantly affected all the tested indicators of mitochondrial integrity and functionality, PUMP and PmitoKATP activation was further enhanced. Interestingly, both systems were activated by reactive oxygen species and were able to control mitochondrial superoxide anion production. These results suggest that PUMP and PmitoKATP serve as early antioxidant defence systems in response to hyperosmotic stress and that they are involved in a prolonged response to stress.

    Notizen: : Geotrichum fragrans and another Geotrichum sp., 2 newly isolated microorganisms, produce -γ-decalactone, which exhibits odors characteristic of fruit and chocolate derivatives. The production of-γ-decalactone by Geotrichum species in broth, to which enzymatically hydrolyzed castor oil had been added, was investigated. The study of different microbial lipases, aimed at producing the best hydrolyzed oil for use as a precursor for the production of -γ-decalactone, was also studied. The addition of ricinoleic acid or hydrolyzed castor bean oil to the culture medium enhanced -γ-decalactone production. The greatest enhancement of -γ-decalactone production was noted, with a yield of 600 mg/L, when 5% of hydrolyzed castor bean oil was added to the medium and fermented by Geotrichum fragrans for 96 h.

    Notizen: This paper describes the first identification of chemotaxis genes in Bacillus cereus. We sequenced and studied the genomic organization and the expression of the cheA and fliY genes in two different B. cereus strains, ATCC 14579 and ATCC 10987. While cheA encodes a highly conserved protein acting as the main regulator of the chemotactic response in flagellated eubacteria, fliY, which has been previously described only in B. subtilis, is one of the three genes encoding proteins of the flagellar switch complex. Although the sequences and relative position of cheA and fliY were found to be identical in the two B. cereus strains analyzed, the restriction fragment containing both genes was located differently on the physical maps of B. cereus ATCC 14579 and ATCC 10987. Evidence is shown that the genomic organization and the expression of fliY and cheA in B. cereus differ significantly from that described for B. subtilis, which is considered a model microorganism for chemotaxis in Gram-positive bacteria.
