    Comparative study of the thiourea carrier in erythrocytes (1977)

    Notes: Summary A densimeter technique was used to measure the rate of exit of thiourea from erythrocytes of various species of mammals. The cells were first equilibrated with a 200mm thiourea solution in 1% NaCl. An aliquot of these cells was added to 1% NaCl containing 4.6–23.1mm thiourea. Facilitated diffusion was demonstrated in each case. Using exit times or initial rates, calculations of half-saturation constants (ϕ) inmm and maximum transport rates (K) in isotones per min were made by three different methods. The following values were obtained: human −ϕ=60, 42, 35;K=1.2, 2.9, 0.9; rabbit −ϕ=46, 33, 32;K=0.8, 2.1, 0.8; mouse −ϕ=46,40, 30;K=3.4, 8.5, 3.2; rat −ϕ=65, 42, 23;K=6.1, 15.3, 3.7; ox −ϕ=107, 63, 88;K=0.6, 1.4, 0.4; sheep −ϕ=56, 38, 56;K=0.9, 2.2, 0.6; and pig −ϕ=110, 64, 49;K=1.6, 3.6, 1.1.

Nicotinamide adenine dinucleotide phosphate (NADP) and nicotinamide adenine dinucleotide (NAD) linked isocitrate dehydrogenase and NADP linked alcohol dehydrogenase have been detected in Euglena gracilis var. bacillaris.The NADP isocitrate dehydrogenase showed half-maximal activity at a concentration of 3 × 10−5 M DL-isocitrate, but did not follow simple Michaelis-Menten kinetics with respect to substrate concentration. The optimal NADP concentration was about 0.06 mM, and activity fell off sharply on either side of this optimum. Fresh preparations of the enzyme migrated as single bands in disc electrophoresis, but two enzymatically active bands were present after frozen storage.The NAD isocitrate dehydrogenase followed Michaelis-Menten kinetics with respect to substrate. In crude extracts, no requirement for adenosine monophosphate, adenosine diphosphate, or sulfhydryl compounds could be found.NADP alcohol dehydrogenase activity could be found with either ethanol or propanol as substrate. Low concentrations of coenzyme A were moderately inhibitory.In tris(hydroxymethyl) aminomethane buffer (tris buffer), Euglena extracts reduced NAD slowly in the absence of exogenous substrate. In the absence of tris, no such reduction occurred. A similar phenomenon was observed with NADP.

    Notes: Using a densimeter technique, the effect of tannic acid and n-butanol on the permeability to non-electrolytes of erythrocytes of various species was studied. If both tannic acid and butanol decrease the permeability it is suggested that a carrier mechanism is involved. If tannic acid has little or no effect and butanol increases the permeability it is suggested that the penetration of such a molecule depends only on diffusion. According to this theory carriers are involved in the following systems: Urea  -  mouse, sheep, pig, ox and man; Ethylene glycol  -  man and possibly mouse; Glycerol  -  man, mouse, rabbit; Erythritol  -  mouse and possibly man; Ribose  -  mouse, man; Mannose, glucose, sorbose  -  man. No carriors are involved in the following systems: Urea  -  chicken; Ethylene glycol  -  pig, chicken, ox, sheep; Thiourea  -  rabbit, chicken, sheep, man, pig, ox; Glycerol  -  chicken, pig, sheep, ox; Erythritol  -  rabbit; Ribose  -  rabbit.

    Notes: Studies of the effect of triton X-100 on the fragility of rabbit erythrocytes and on shape and volume changes of rabbit and sheep erythrocytes suspended in an isotonic salt solution suggest that this subtance alters the erythrocytes of other species in a manner similar to that reported previously for human red cells. Using a densimeter technique it was found that the permeability of relatively slowly moving non-electrolyte-carrier-complexes is decreased by triton X-100 but more rapidly penetrating systems are not affected by this substance. The permeability of some molecules which cross the membrane by diffusion is increased by triton X-100. It is suggested that triton X-100 affects both the aqueous channels and the portion of the membrane across which carriers move. Its action is less than that of tannic acid or butanol.
