    Notes: Abstract A soybean chitinase which has an apparent molecular mass of 28 kDa by SDS-PAGE, and has chitinase specific activity of 133 units per mg protein at pH 5.2 and an apparent pI of 5.7, was purified from mature dry seeds. Based upon the selected part (the residue positions 10–17) of the determined N-terminal 38 amino acid sequence, a 23-mer degenerate oligonucleotide was synthesized and used for the PCR cloning of the chitinase cDNA. The resulting 1340 bp cDNA was comprised of a 5′-untranslated region of 39 bases, a coding region corresponding to a 25 amino acid signal sequence, followed by a mature 308 amino acid sequence (calculated molecular mass 34269, calculated pI 4.7), and a 235 nucleotide 3′-terminal untranslated region including 24 bases of the poly(A) tail. By comparing the deduced primary sequence with those of plant chitinases known to date, this enzyme was more than 50% identical to every class III acidic chitinase, but has no significant similarity to other families of chitinases. The comparison also showed that the C-termininal region of this chitinase is markedly extended, by at least 31 residues. Northern blot analysis demonstrated that this mRNA species is remarkably transcribed from the early stage until the late middle stage of seed development, whilst it is hardly expressed in the leaves and the stems of soybean. Spatial and temporal expression of this single gene imply that this class III chitinase is mainly devoted to the seed defense, not only in development but also in dormancy of soybean seed. This is the first reported isolation and cDNA cloning of a class III acidic endochitinase from seeds. According to the chitinase nomenclature we propose that this enzyme would be classified into a new class of chitinase PR-8 family, together with a Sesbania homologue.

    Description: Hydrolysis and photolysis kinetics of Fubianezuofeng (FBEZF) in water were investigated in detail. The hydrolysis half-lives of FBEZF depending on pH, initial concentration, and temperature were (14.44 d at pH = 5; 1.60 d at pH = 7), (36.48 h at 1.0 mg L−1; 38.51 h at 5.0 mg L−1; and 31.51 h at 10.0 mg L−1), and (77.02 h at 15 °C; 38.51 h at 25 °C; 19.80 h at 35 °C; and 3.00 h at 45 °C), respectively. The photolysis half-life of FBEZF in different initial concentrations were 8.77 h at 1.0 mg L−1, 8.35 h at 5.0 mg L−1, and 8.66 h at 10.0 mg L−1, respectively. Results indicated that the degradation of FBEZF followed first-order kinetics, as the initial concentration of FBEZF only had a slight effect on the UV irradiation effects, and the increase in pH and temperature can substantially accelerate the degradation. The hydrolysis Ea of FBEZF was 49.90 kJ mol−1, which indicates that FBEZF belongs to medium hydrolysis. In addition, the degradation products were identified using ultra-high-performance liquid chromatography coupled with an Orbitrap high-resolution mass spectrometer. One degradation product was extracted and further analyzed by 1H-NMR, 13C-NMR, 19F-NMR, and MS. The degradation product was identified as 2-(4-fluorobenazyl)-5-methoxy-1,3,4-oxadiazole, therefore a degradation mechanism of FBEZF in water was proposed. The research on FBEZF can be helpful for its safety assessment and increase the understanding of FBEZF in water environments.

    Description: The activating immunoreceptor NKG2D (natural killer group 2, member D) and its ligands play important roles in the innate and adaptive immune responses. UL16-binding protein 3 (ULBP3), an NKG2D ligand, is overexpressed on certain epithelial tumor cells. In this study, we investigated the effect of ULBP3 expression on the cytotoxic activity of natural killer (NK) cells. ULBP3 were measured by flow cytometry analysis, immunohistochemistry, and time-resolved fluoroimmunoassay. The cytotoxicity of NK cells was determined with the lactate dehydrogenase release assay. We found that ULBP3 was overexpressed on tumor cell lines and tumor tissues. Serum from cancer patients, but not from healthy donors, contained elevated levels of soluble ULBP3 (sULBP3). Importantly, high expression of ULBP3 on the cell surface of tumor cells augmented NKG2D-mediated NK cell cytotoxicity. However, low levels of sULBP3 (〈15 ng/ml) weakened the cytotoxicity of NK cells by decreasing NKG2D expression on NK cells. Further analysis showed that serum samples from most cancer patients (〉70%) contained the low level of sULBP3. Our results demonstrate that tumor cells express surface and soluble ULBP3, which regulate NK cell activity. Thus, ULBP3 is a potential therapeutic target for improving the immune response against cancer. Scientific Reports 4 doi: 10.1038/srep06138

To deal with medical diagnosis problems based on generalized fuzzy sets (FSs), the notion of single-valued neutrosophic multisets (SVNMs) is firstly used to express the diagnostic information in this article. Then the model of probabilistic rough sets (PRSs) over two universes is applied to analyze SVNMs, and the concepts of single-valued neutrosophic rough multisets (SVNRMs) over two universes and probabilistic rough single-valued neutrosophic multisets (PRSVNMs) over two universes are introduced. Based on SVNRMs over two universes and PRSVNMs over two universes, single-valued neutrosophic probabilistic rough multisets (SVNPRMs) over two universes are further established. Next, a three-way decisions model by virtue of SVNPRMs over two universes in the context of medical diagnosis is constructed. Finally, a practical case study along with a comparative study are carried out to reveal the accuracy and reliability of the constructed three-way decisions model.

    Description: Background: Dufulin is a novel, highly effective antiviral agent that activatives systemic acquired resistance of plants. This compound is widely used in China to prevent and control viral diseases in tobacco, vegetable and rice. Dufulin can treat plants infected by the tobacco mosaic virus and the cucumber mosaic virus. However, the achiral analysis and residue determination of dufulin remain underdeveloped because of its high enantioselectivity rates and high control costs. The enantioselectivity of an antiviral compound is an important factor that should be considered when studying the effect of chiral pesticides on the environment. The enantioselective degradation of dufulin in water remains an important objective in pesticide science. Results: The configuration of dufulin enantiomers was determined in this study based on its circular dichroism spectra. The S-(+)-dufulin and R-(-)-dufulin enantiomers were separated and identified using an amylose tris-(3,5-dimethylphenylcarbamate) chiral column by normal phase high-performance liquid chromatography. The degradation of the rac-dufulin racemate and its separate enantiomers complied with first-order reaction kinetics and demonstrated acceptable linearity. The enantioselective photolysis of rac-dufulin allowed for the faster degradation of R-(-)-dufulin, as compared with S-(+)-dufulin. However, S-(+)-dufulin was hydrolyzed faster than its antipode. Conclusion: The photolysation and hydrolyzation of dufulin in water samples normally complied with the first-order kinetics and demonstrated acceptable linearity (R2〉0.66). A preferential photolysation of the R-(-)-enantiomer was observed in water samples. Moreover, the S-(+)-enantiomer was hydrolyzed faster than its antipode.

Since SOC cannot be obtained directly by measurement, SOC estimation methods are required. In this paper, three model-based methods, including the extended particle filter (EPF), cubature particle filter (CPF), and unscented particle filter (UPF), are compared in terms of complexity, accuracy, and robustness. The second-order resistor-capacitor (RC) equivalent circuit model is selected as the circuit model of the lithium-ion battery, and the parameters of the model are obtained by off-line identification. Then, the City test is applied to compare the performance of the methods. The experimental results show that the EPF method exhibits low complexity and fast running speed, but poor accuracy and robustness. Compared with the EPF method, the complexity of the CPF and UPF methods is relatively high, but these models offer improved accuracy and robustness.

This paper provides a case study of Hulusu Coal Mine in Inner Mongolia, where roadway excavation speed was insufficient to meet production needs. Moreover, deformation in the roofs of the roadways was severe. To achieve rapid excavation and control the stability of deep roadways, a new support system was proposed and constructed in a roadway at a depth of 640 m. The system consisted of long flexible bolts pretensioned to high levels and spaced at large intervals. Roadway excavation and construction of a support system were conducted simultaneously. Field measurements indicated that the new support system effectively controlled deformation and fracture development during excavation and mining. Maximum displacements of the roof during excavation and mining were 10 and 30 mm, respectively. The axial load on bolts surged during excavation as a result of slight deformations caused by excavation operations. This active response of the bolts is beneficial to the prevention of roof deformation during excavation and mining. During mining, fissures propagated up to only a depth of 1.4 m into the surrounding rock mass. The new support system formed a thick reinforced anchorage zone, which greatly improved the bearing capacity of the roof. Compared with the previous support system, the new system allowed the maximum excavation speed (31.5 m/day) to increase by 85.3%. This successful case provides a practical reference for similar roadway projects.
