Thus, we developed a simple, sensitive and universal protein-sensing platform based on peptide and graphene oxide (GO). The design consists of a fluorophore (TAMRA, TAM), a peptide containing eight arginines and peptide ligand that could recognize the target protein, and GO used as a quencher. To demonstrate the feasible use of the sensor for target detection, Bcl-xL was evaluated as the model target. The sensor was proved to be sensitive and applied for the detection of the target proteins in buffer, 2% serum and living cells.

    Description: N-acylhomoserine lactone (AHL)-based quorum sensing (QS) is important for the regulation of proteobacterial virulence determinants. Thus, the inhibition of AHL synthases offers non-antibiotics-based therapeutic potentials against QS-mediated bacterial infections. In this work, functional AHL synthases of Pseudomonas aeruginosa LasI and RhlI were heterologously expressed in an AHL-negative Escherichia coli followed by assessments on their AHLs production using AHL biosensors and high resolution liquid chromatography–mass spectrometry (LCMS). These AHL-producing E. coli served as tools for screening AHL synthase inhibitors. Based on a campaign of screening synthetic molecules and natural products using our approach, three strongest inhibitors namely are salicylic acid, tannic acid and trans-cinnamaldehyde have been identified. LCMS analysis further confirmed tannic acid and trans-cinnemaldehyde efficiently inhibited AHL production by RhlI. We further demonstrated the application of trans-cinnemaldehyde inhibiting Rhl QS system regulated pyocyanin production in P. aeruginosa up to 42.06%. Molecular docking analysis suggested that trans-cinnemaldehyde binds to the LasI and EsaI with known structures mainly interacting with their substrate binding sites. Our data suggested a new class of QS-inhibiting agents from natural products targeting AHL synthase and provided a potential approach for facilitating the discovery of anti-QS signal synthesis as basis of novel anti-infective approach.     Description: This paper presents a variational model based segmentation approach for polarimetric synthetic aperture radar (PolSAR) images. The formulation for PolSAR image segmentation is based on a scaled Wishart distribution based continuous Potts model, which can partition the image domain into distinct regions with respect to the statistical property of PolSAR data. To make the segmentation efficient, a duality based optimization approach is utilized to minimize the energy functional. Moreover, an automatic initialization approach which takes the unsupervised H--a classification result of the polarimetric data as input is used to initialize the segmentation process. This approach can estimate the appropriate number of clusters and the corresponding classification map for the PolSAR data, which are used as the input of the following variational segmentation approach. In such a way, the proposed approach is carried out in a fully unsupervised way. Both of the polarimetric decomposition features and the statistical characteristics are used to get the final segmentation result, which helps to increase the accuracy. Experimental results demonstrate the effectiveness of the proposed approach. Without any artificial supervision, the proposed approach can produce superior segmentation results than results obtained with random initialized variational approach and Wishart--H--a classification approach.

    Description: Traditionally, P wave arrival times have been used to locate regional earthquakes. In contrast, the travel times of surface waves dependent on source excitation and the source parameters and depth must be determined independently. Thus surface wave path delays need to be known before such data can be used for location. These delays can be estimated from previous earthquakes using the cut-and-paste technique, Ambient Seismic Noise tomography, and from 3D models. Taking the Chino Hills event as an example, we show consistency of path corrections for (〉10 s) Love and Rayleigh waves to within about 1 s obtained from these methods. We then use these empirically derived delay maps to determine centroid locations of 138 Southern California moderate-sized (3.5 〉 Mw 〉 5.7) earthquakes using surface waves alone. It appears that these methods are capable of locating the main zone of rupture within a few (∼3) km accuracy relative to Southern California Seismic Network locations with 5 stations that are well distributed in azimuth. We also address the timing accuracy required to resolve non-double-couple source parameters which trades-off with location with less than a km error required for a 10% Compensated Linear Vector Dipole resolution.

    Description: Recent reports on modeling USArray data reveal mostly vertical microplates with little resemblance to preliminary reference Earth model (PREM). Such complexity at plate boundaries makes it difficult to form reliable images of ocean basins using global paths. Here, we report on modeling stacked seismograms obtained from the first broadband array (TriNet) situated on the edge of the Pacific Plate, southern California, with no major subduction zone blocking its view. Extended records, including multi-S and ScS waves up to four bounces from 18 Tonga-Fiji deep events (140 to 620 km) are analyzed to check the validity of existing models and derive the whole mantle shear velocity structure along this corridor. Synthetics generated from 3-D tomographic models do not fit the upper mantle triplication data or the mantle reverberations associated with the ScS multiples as well as the 1-D model PAC06. We construct a hybrid model (HPAC), which remains one dimensional down to 800 km (PAC06). The lower portion of HPAC is essentially the tomography model S20RTS with velocity variation inflated by a factor of 2 for the lowermost 600 km. Thus, the mid-Pacific large low shear velocity province (LLSVP) has a lower shear velocity of about 2% relative to PREM and extends into the midmantle, similar to that beneath South Africa. Moreover, rapid changes in the differential (ScS-S) and (ScS2-S) times as a function of distance suggest ultra low velocity zones near the eastern edge and under the LLSVP, again similar to that found beneath Africa.

    Description: Quorum sensing regulates bacterial virulence determinants, therefore making it an interesting target to attenuate pathogens. In this work, we screened edible, endemic plants in Malaysia for anti-quorum sensing properties. Extracts from Melicope lunu-ankenda (Gaertn.) T. G. Hartley, a Malay garden salad, inhibited response of Chromobacterium violaceum CV026 to N-hexanoylhomoserine lactone, thus interfering with violacein production; reduced bioluminescence expression of E. coli [pSB401], disrupted pyocyanin synthesis, swarming motility and expression of lecA::lux of Pseudomonas aeruginosa PAO1. Although the chemical nature of the anti-QS compounds from M. lunu-ankenda is currently unknown, this study proves that endemic Malaysian plants could serve as leads in the search for anti-quorum sensing compounds.

    Description: We combined zeolitic imidazolate framework nanoparticles (ZIF-8: ˜150 nm diameter) with Matrimid ® 5218 polymer to form permeable mixed matrix membranes, featuring different weight fractions of nanoparticles (up to 30 wt. % loading). We used ball-on-disc micro-tribological method to measure the frictional coefficient of the nanocomposite membranes, as a function of nanoparticle loading and annealing heat treatment. The tribological results reveal that the friction and wear of the unannealed samples rise steadily with greater nanoparticle loading because ZIF-8 is relatively harder than the matrix, thus promoting abrasive wear mechanism. After annealing, however, we discover that the nanocomposites display an appreciably lower friction and wear damage compared with the unannealed counterparts. Evidence shows that the major improvement in tribological performance is associated with the greater amounts of wear debris derived from the annealed nanocomposite membranes. We propose that detached Matrimid-encapsulated ZIF-8 nanoparticles could function as “spacers,” which are capable of not only reducing direct contact between two rubbing surfaces but also enhancing free-rolling under the action of lateral forces.
