    Notes: Abstract A monocyte-derived steroid-induced factor has been shown previously to induce dispersive locomotion in human neutrophils and to lower adhesion to an albumin-coated glass surface. In this paper we show that this factor inhibits adhesion of neutrophils to bovine aorta and human endothelial cells by an undetermined mechanism. It induces unique changes in neutrophil shape with a characteristic monopolar pattern of F-actin distribution, which may correlate with the dispersive locomotion observed in the absence of a concentration gradient. This factor also inhibits N-formyl-methionyl-leucyl-phenylalanine-induced chemotaxis of neutrophils in a modified Boyden chamber assay. The reduction of adhesion and the inhibition of chemotaxis by the factor in vitro indicate a possible in vivo anti-inflammatory role.

    Notes: Abstract An inherited amino acid transport deficiency results in low concentrations of glutathione (GSH) in the erythrocytes of certain sheep. Earlier studies based on phenotyping according to GSH concentrations indicated that the gene Tr H, which controls normal levels of GSH, behaves as if dominant or incompletely dominant to the allele Tr h, which controls the GSH deficiency. The present paper shows that when sheep are classified according to amino acid transport activity, the Tr H gene behaves as if codominant to Tr h. Erythrocytes from sheep homozygous for the Tr H gene exhibit rapid saturable l-alanine influx (apparent K m ,21.6mm; V max, 22.4 mmol/liter cells/hr). Cells from sheep homozygous for the Tr h gene exhibit slow nonsaturable l-alanine uptake (0.55 mmol/liter cells/hr at 50mm extracellular l-alanine). Cells from heterozygous sheep show saturable l-alanine uptake with a diminished V max (apparent K m, 19.1mm; V max, 12.7 mmol/liter cells/hr). These erythrocytes have a significantly lower GSH concentration than cells from Tr H, TrH sheep but similar intracellular levels of dibasic amino acids.

    Notes: Abstract Nucleoside transport in sheep red cells is controlled by two allelomorphic genes, the gene for nucleoside transport deficiency (Nu I) being dominant to that for the functional presence of carrier-mediated nucleoside transport activity (Nu i). Sheep are also polymorphic with respect to their red-cell nucleoside phosphorylase (NP) activity, some having high activities and others low activities of this enzyme. The gene for high activity (NP H) is incompletely dominant to that for low activity (NP L). Inheritance data indicate that theNu locus is genetically linked to that for the B blood-group system and, in addition, exerts a pleiotropic effect on NP activity, Nu permeability stabilizing the heat-labileNP L gene product. Nu-permeable cells have a higher ATP content than Nu-impermeable red cells, and within the Nu-impermeable subgroup, NP deficiency causes a further reduction in red cell ATP concentration. It is concluded that the nucleoside inosine supplements glucose as a physiological energy substrate in sheep red cells.

    Notes: Abstract Nucleoside transport in sheep erythrocytes is under the genetic control of two allelomorphic genes (Nu I and Nu i ), where Nu I codes for the functional absence of a high-affinity nucleoside transport system and is dominant to the gene (Nu i ) coding for the presence of the transport system. Kinetic and inhibitor experiments show that the high-affinity transport system is not present in heterozygous erythrocytes, demonstrating that the Nu I gene is completely dominant over the Nu i gene. It is suggested that the Nu locus may not represent the structural gene locus of the nucleoside transport system. Instead, it may be a regulator gene locus.

    Notes: Abstract Red blood cell γ-glutamylcysteine synthetase (GC-S) activities and glutathione (GSH) concentrations were investigated in pure-bred Finnish Landrace, Tasmanian Merino, and Finnish Landrace × Tasmanian Merino sheep Previous studies were confirmed that a dominant gene (L) at the GSH locus leads to partial GC-S and GSH deficiency, while a recessive gene (h) at the Tr locus codes for defective red cell amino acid transport, concomitant GSH deficiency, and significantly elevated levels of GC-S. In addition, new results are presented which suggest that there is a dominant gene (A) at another locus distinct from GSH and Tr (provisionally designated GC-S) which has a marked epistatic enhancing effect on measured GC-S activities.
