    Notes: Abstract Metachromatic leukodystrophy (MLD) is a lysosomal storage disease with autosomal recessive inheritance caused by a deficiency of the enzyme arylsulfatase A (ASA). We have identified a new mutation in the ASA gene of a patient with adult-type MLD. In this mutation, the glycine at position 122, a highly conserved residue in the AS gene family, was replaced by serine. In a transient expression study, COS cells transfected with the mutant cDNA carrying 122Gly→Ser did not show an increase of ASA activity and produced little material immunoreactive to an anti-ASA antibody, despite normal mRNA levels.

    Notes: Abstract Purpose. Reduction of the frequency of injections and localization of the absorption of drug molecules to the injection site would be of great advantage in epidural pain treatment. The epidural use of a controlled release gel of lidocaine and ibuprofen was studied. Methods. The effect of a poloxamer gel (25%) containing 2% lidocaine ⋅HC1 and 2% ibuprofen ⋅ Na on the duration of analgesia after epidural administration to pigs was compared with drug in solution. Analgesia was assessed by observing the motor function and the nociceptive reflex-withdrawal response to painful pressure stimulation on the feet. Pharmacokinetic and histological examinations were performed. Results. Analgesia lasted significantly longer after epidural lidocaine gel injection in comparison with the solution. The gel prolonged the systemic absorption, thereby increasing the epidural availability of lidocaine for spinal analgesia. Although the absorption of ibuprofen was prolonged after epidural gel injection, the duration of analgesia as compared with the solution was not prolonged. After epidural injection, only slight inflammatory changes were observed in the tissue structures of the epidural space, but none in the spinal cord. Conclusions. These results demonstrate poloxamer gel to be a promising controlled-release, injectable epidural formulation for the management of pain.

    Notes: Scattering of SH wave by an elastic half space with a lining structure and a crack in anyposition and direction is studied with Green’s function, complex function and multi-polarcoordinate method. First, a suitable Green’s function is constructed, which is a fundamental solutionto the displacement field for the elastic space possessing circular lining structure while bearingout-of-plane harmonic line source load at arbitrary point. Then a crack in any position and directionis constructed by means of crack-division in half space. Finally the displacement field and stressfield are established in the case of coexistence of circular lining structure and crack, and theexpression of dynamic stress intensity factor (DSIF) at the tip of crack is given. According tonumerical examples, the influences of different parameters on DSIF are discussed

    Notes: Abstract The glycinin gene family encoding the glycinin subunits in soybean plants is composed of at least five gene members. A genomic clone λS312 containing the Gy4 gene from a genomic library of cv. Forrest was isolated and partially characterized. The organization of this gene was found to be similar to that of a null allele from cv. Raiden, but different from the Gy4 gene from cv. Dare. The complete nucleotide sequence of this gene has been determined. It is 2599 bp long consisting of four exons and three introns. Comparing the DNA sequences between this gene and the gene from Dare and a null allele from Raiden, the difference found in the coding region was 5′-GCAGTGCAAG-3′ (nt 824 to 833) in the former case versus 5′-TGGAGTTGCAATT-3′ (nt 1314 to 1326) in the latter case in the exon 2 domain, resulting in three amino acid differences and one amino acid absence. Some other differences were also found in the non-coding region. The coding sequence and 5′-flanking region of the Gy4 gene, when compared with that of other legumin genes as well as group 1 glycinin subunit genes, revealed some interesting features: (1) a transposable element-like sequence was found in the hypervariable region (HVR) of the exon 3 domain, which was lacking in the legumin and the glycinin group 1 genes; (2) in the 5′-flanking region from nt −145 to −1, two high-homology sequences were found: one from nt −141 to nt −132, the other from nt −118 to nt −92 which includes the ‘legumin box’ and the RY repeat element.

    Notes: The murine intracisternal A particle (IAP) proviral elements are expressed at low levels in undifferentiated F9 embryonal carcinoma cells but are highly expressed when F9 cells are induced to differentiate into parietal endoderm-like cells. IAP elements are also expressed in parietal endoderm-like PYS-2 cells. We previously identified an IAP proximal enhancer (IPE) element that mediates a F9 differentiation-specific enhancer activity. We also identified a 60 kDa IPE binding (IPEB) protein whose activity is high in PYS-2 cells, where IAP is expressed, but very low in F9 cells.Transcription of IAP elements has also been shown in the adult mouse thymus and in activated splenic B cells. We have now shown by DNA affinity chromatography, sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and band-shift analysis that the 60 kDa IPEB is expressed in adult T lymphocytes and in resting as well as lipopolysaccharide activated splenic B cells but not in adult liver cells, suggesting an important role for IPEB in IAP transcription in vivo. In addition, we find IPEB expressed in the fetal mouse at sites of lymphoid development, such as the liver, spleen, and thymus, suggesting it may play an important role in gene expression during lymphoid development. In support of this, we find IPEB in the human T cell tumor lines, Jurkat and Molt 13, as well as the Daudi B cell line and in the normal calf thymus and in the thymus and spleen of the chicken and rat. © 1995 Wiley-Liss, Inc.

    Notes: Acrylate copolymer composed of octadecyl acrylate (OA), 2-ethyl hexyl acrylate (2-EHA), hydroxythyl acrylate (HEA), and vinyl acetate (VA) was synthesized. The copolymer was used to adhere polyethylene (PE) with T-type peel, strength 4.4 N/cm. The interfacial characteristics of copolymer and PE were investigated via ATR-FTIR, transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), and dynamic contact angle (DCA). Results show that apolar moieties of the adhesive enrich on the interface. Moreover, the time dependence of peel strength coincides with that of enrichment in nonpolar moieties on the interface. © 1995 John Wiley & Sons, Inc.
